Thioredoxin is a small ubiquitous protein that is involved in the dithiol-disulfide exchange reaction, byway of two cysteine residues located on the molecule surface. In order to elucidate the role of thioredoxin in Chlorobaculum tepidurn, an anaerobic green sulfur bacterium that uses various inorganic sulfur compounds and H2S as electron donors under strict anaerobic conditions for growth, we applied the thioredoxin affinity chromatography method （Motohashi et al., 2001）. In this study, 37 cytoplasmic proteins were captured as thioredoxin target candidates, including proteins involved in sulfur assimilation. Furthermore, six of the candidate proteins were members of the reductive tricarboxylic acid cycle （pyruvate orthophosphate dikinase, pyruvate flavodoxin/ferredoxin oxidoreductase, ^-oxoglutarate synthase, citrate lyase, citrate synthase, malate dehydrogenase）. The redox sensitivity of three enzymes was then examined： citrate lyase, citrate synthase, and malate dehydrogenase, using their recombinant proteins. Based on the information relating to the target proteins, the significance of thioredoxin as a reductant for the metabolic pathway in the anaerobic photosynthetic bacteria is discussed.

High-resolution modeling and evaluation of ozone air quality of Osaka using MM5-CMAQ system

High-resolution modeling approach is increasingly being considered as a necessary step for improving the monitoring and predictions of regional air quality. This is especially true for highly urbanized region with complex terrain and land-use. This study uses Community Multiscale Air Quality （CMAQ） model coupled with MM5 mesoscale model for a comprehensive analysis to assess the suitability of such high-resolution modeling system in predicting ozone air quality in the complex terrains of Osaka, Japan. The 1-km and 3-kin grid domains were nested inside a 9-km domain and the domain with 1-km grid covered the Osaka region. High-resolution Grid Point Value-Mesoscale Model （GPV-MSM） data were used after suitable validation. The simulated ozone concentrations were validated and evaluated using statistical metrics using performance criteria set for ozone. Daily maxima of ozone were found better simulated by the 1-krn grid domain than the coarser 9-km and 3-km domains, with the maximum improvement in the mean absolute gross error about 3 ppbv. In addition, 1-km grid results fared better than other grids at most of the observation stations that showed noticeable differences in gross error as well as correlation. These results amply justify the use of the integrated high-resolution MM5-CMAQ modeling system in the highly urbanized region, such as the Osaka region, which has complex terrain and land-use.

The glass forming ability of Gd-Co-Al ternary alloy systems with a composition ranging from 50% to 70% (molar fraction) for Gd and from 5% to 40% (molar fraction) for Al were investigated by copper mold casting and Gd60Co25Al15 bulk glass alloy cylinders with the maximum diameter of 5 mm were obtained. The reduced glass transformation temperature (Tg/Tm) and the distance of supercooling region △Tx are 0.616 and 45 K, respectively for this Gd-Co-Al alloy. The compressive fracture strength (σf) and elastic modulus (E) of Gd-Co-Al glassy alloys are 1 170-1 380 MPa and 59-70 GPa, respectively. The Gd-Al-Co bulk glassy alloys with high glass forming ability and good mechanical properties are promising for the future development as a new type function materials.

Barley(Hordeum vulgare ssp.vulgare)is one of the first crops to be domesticated and is adapted to a wide range of environments.Worldwide barley germplasm collections possess valuable allelic variations that could further improve barley productivity.Although barley genomics has offered a global picture of allelic variation among varieties and its association with various agronomic traits,polymorphisms from East Asian varieties remain scarce.In this study,we analyze exome polymorphisms in a panel of 274 barley varieties collected worldwide,including 137 varieties from East Asian countries and Ethiopia.We reveal the underlying population structure and conduct genome-wide association studies for 10 agronomic traits.Moreover,we examin genome-wide associations for traits related to grain size such as awn length and glume length.Our results demonstrate the value of diverse barley germplasm panels containing Eastern varieties,highlighting their distinct genomic signatures relative to Western subpopulations.

Survival of pathogenic bacteria in compost with special reference to Escherichia coli

Application of compost in agricultural practice could potentially cause contamination of foodstuffs with pathogenic bacteria such as Escherichia coli O157：H7 （E. Coli O157）. We investigated pathogenic bacteria in compost collected from the compost facilities, and evaluated the survival of E. coli K12 and O157 in laboratory experiments. Out of 19 compost product samples, coliform bacteria and salmonella were detected in 7 and 3 samples respectively. The number of coliform bacteria was 1.8 × 10^2 to 2.5 × 10^6 CFU/g dw and that of salmonella was 4.2 × 10^1 to 6.0 × 10^3 CFU/g dw. Moreover. coliform bacteria, fecal coliform, E. coli and salmonella were detected during composting at 54℃ to 67℃. The results indicated that moisture content was a very important factor to the heat sensitivity of pathogenic bacteria in compost, E. coli in compost of high moisture content was more sensitive than that in compost of low moisture content, cells harvested in logarithmic phase was more sensitive than these in stationary phase, and E. coli K12 was more sensitive than E. coli O157. Based on the D values, the lethal time of E. coli K12 and 0157 from 10^8 to 10^0 CFU/g dw were 16.3 and 28.8 min, respectively, at 60℃ in compost with 40% moisture content. However, some E. coli cells survived in composting process at 54℃ to 67℃. Water potential（low moisture content） and physiological aspects of bacteria（stationary phase） could explain only in part of the prolonged survival of E. coli in compost, and there should be some other factors that are conducive to bacterial survial in compost.

This paper proposes an operator based fault detection method for an actuator fault of an aluminum plate thermal process with input constraints. Operator-based robust right coprime factorization(RCF)approach is utilized in this method. After developing a mathematical model of the thermal process,a robust tracking operator system is designed for the process with input constraints. Following this, design of the fault detection system is given by using operator-based robust RCF approach. Finally, experiment is conducted to support the proposed design method

Multicomponent alloys of Zr_(50)M_(50),Zr_(50)(M,Ag)_(50)and Zr_(50)(M,Pd)_(50)(M=Fe,Co,Ni,Cu)can be melt-spun to obtain amorphous ribbons.The maximum thickness for fully amorphous ribbons varies with composition in the range 34-53μm.In contrast,fully amorphous ribbons are not obtainable for binary Zr_(50)Ni_(50)or ternary Zr_(50)(Ni,Cu)_(50)alloys.Heating-induced crystallization occurs through:two stages of amorphous[am]→[~(am')+B2]→[B2+B33]for Zr_(50)M_(50);and[am]→[am'+B2]→[B2+AgZr]for Zr_(50)(M,Ag)_(50);and a single stage of[am]→[B2]for Zr_(50)(M,Pd)_(50),while no B2 phase is formed for the binary and ternary Zr_(50)Q_(50)(Q=Ni or/and Cu)alloys.As-spun amorphous ribbons have good bending plasticity.Remarkably,Zr_(50)M_(50)ribbons in tension show 0.22-0.28%plastic elongation and work-hardening(the yield stress is~820 MPa,the fracture stress is~1200 MPa).When cold-rolled at room temperature to 30%reduction in thickness,Zr_(50)M_(50)ribbons show 10%increase in hardness,while retaining good bending plasticity.Cold-rolling induces precipitation of spheroidal B2 and irregular B33 particles,while deformation in tension induces B2,B33 and also plate-like monoclinic precipitates.The B2 and B33 particles form by polymorphic transformation,and include a high density of internal defects.This novel deformationinduced precipitation has not been recognized for any Zr_(50)Q_(50)binary or ternary alloys.The new multicomponent systems are encouraging for future progress as structural amorphous alloys.
