Metaxin deficiency alters mitochondrial membrane permeability and leads to resistance to TNF-induced cell killing

Metaxin, a mitochondrial outer membrane protein, is critical for TNF-induced cell death in L929 cells. Its deficiency, caused by retroviral insertion-mediated mutagenesis, renders L929 cells resistance to TNF killing. In this study, we further characterized metaxin deficiency-caused TNF resistance in parallel with Bcl-XLoverexpressionmediated death resistance. We did not find obvious change in mitochondria membrane potential in metaxindeficient(Metmut) and Bcl-XL-overexpressing cells, but we did find an increase in the release rate of the mitochondrial membrane potential probe rhodamine 123(Rh123) that was preloaded into mitochondria. In addition, overexpression of a function-interfering mutant of metaxin(MetaΔTM/C) or Bcl-XLin MCF-7.3.28 cells also resulted in an acquired resistance to TNF killing and a faster rate of Rh123 release, indicating a close correlation between TNF resistance and higher rates of the dye release from the mitochondria. The release of Rh123 can be controlled by the mitochondrial membrane permeability transition(PT) pore, as targeting an inner membrane component of the PT pore by cyclosporin A(Cs A)inhibited Rh123 release. However, metaxin deficiency and Bcl-XLoverexpression apparently affect Rh123 release from a site(s) different from that of Cs A, as Cs A can overcome their effect. Though both metaxin and Bcl-XLappear to function on the outer mitochondrial membrane, they do not interact with each other. They may use different mechanisms to increase the permeability of Rh123, since previous studies have suggested that metaxin may influence certain outer membrane porins while Bcl-XLmay form pores on the outer membrane. The alteration of the mitochondrial outer membrane properties by metaxin deficiency and Bcl-XL overespression, as indicated by a quicker Rh123 release,may be helpful in maintaining mitochondrial integrity.

In recent times,an image enhancement approach,which learns the global transformation function using deep neural networks,has gained ***,many existing methods based on this approach have a limitation:their transformation functions are too simple to imitate complex colour transformations between low-quality images and manually retouched high-quality *** order to address this limitation,a simple yet effective approach for image enhancement is *** proposed algorithm based on the channel-wise intensity transformation is ***,this transformation is applied to the learnt embedding space instead of specific colour spaces and then return enhanced features to *** this end,the authors define the continuous intensity transformation(CIT)to describe the mapping between input and output intensities on the embedding ***,the enhancement network is developed,which produces multi-scale feature maps from input images,derives the set of transformation functions,and performs the CIT to obtain enhanced *** experiments on the MIT-Adobe 5K dataset demonstrate that the authors’approach improves the performance of conventional intensity transforms on colour space ***,the authors achieved a 3.8%improvement in peak signal-to-noise ratio,a 1.8%improvement in structual similarity index measure,and a 27.5%improvement in learned perceptual image patch ***,the authors’algorithm outperforms state-of-the-art alternatives on three image enhancement datasets:MIT-Adobe 5K,Low-Light,and Google HDRþ.

The growing demand for electric vehicles highlights the need for energy storage solutions with higher densities,spotlighting Li metal anodes as potential successors to traditional Li-ion batteries(LIBs).Achieving longer calendar aging life for Li metal anodes is crucial for their practical use,given their propensity for corrosion due to a low redox potential,which leads to compromised cycling stability and significant capacity loss during *** research investigated that this susceptibility is mainly dependent on the surface area of Li metal anode and the properties of the solid electrolyte interphase(SEI),particularly its stability and growth *** research adds to this understanding by demonstrating that the amount of Li plating is a key factor in its corrosion during open-circuit storage,as assessed across various *** discovered that increasing the Li plating amount effectively reduces Coulombic efficiency(C.E.)loss during aging,due to a lower surface area-to-Li *** implies that the choice of electrolyte for optimal storage life should consider the amount of Li plating,with higher capacities promoting better storage characteristics.

BACKGROUND Porocarcinoma is a rare type of skin cancer that originates from sweat gland *** is an aggressive malignant skin cancer that is difficult to diagnose clinically owing to its rarity and similarity to squamous cell carcinoma(SCC).CASE SUMMARY This case involved a 92-year-old woman,a farmer by profession,presented with an exophytic and verrucous mass on her left palm that had formed 2 years prior and caused chronic pain and frequent ***,the patient was diagnosed with SCC using a punch biopsy;however,a repeat biopsy with addi-tional immunohistochemical tests was performed for ***,the patient was diagnosed with porocarcinoma and reconstruction was planned using a full-thickness skin *** treatment,the range of motion of the palm was preserved,and the aesthetic outcome was *** 6 mo of follow-up,the patient was satisfied with the *** Porocarcinoma is commonly misdiagnosed as SCC;therefore,clinicians should consider porocarcinomas when evaluating mass-like lesions on the hands.

This article presents an in-depth exploration of the acoustofluidic capabilities of guided flexural waves(GFWs)generated by a membrane acoustic waveguide actuator(MAWA).By harnessing the potential of GFWs,cavity-agnostic advanced particle manipulation functions are achieved,unlocking new avenues for microfluidic systems and lab-on-a-chip *** localized acoustofluidic effects of GFWs arising from the evanescent nature of the acoustic fields they induce inside a liquid medium are numerically investigated to highlight their unique and promising *** traditional acoustofluidic technologies,the GFWs propagating on the MAWA’s membrane waveguide allow for cavity-agnostic particle manipulation,irrespective of the resonant properties of the fluidic ***,the acoustofluidic functions enabled by the device depend on the flexural mode populating the active region of the membrane *** demonstrations using two types of particles include in-sessile-droplet particle transport,mixing,and spatial separation based on particle diameter,along with streaming-induced counter-flow virtual channel generation in microfluidic PDMS *** experiments emphasize the versatility and potential applications of the MAWA as a microfluidic platform targeted at lab-on-a-chip development and showcase the MAWA’s compatibility with existing microfluidic systems.
