Unicellular micro-alga Chlamydomonas reinhardtii has been recognized as a promising host for expressing recombinant proteins albeit its limited utility due to low levels of heterologous protein expression. Here, transcription of the 3.4-kb mosquito-larvicidal cry4Ba gene from Bacillus thuringiensis in transgenic C. reinhardtii chloroplasts under control of the promoter and 5’-untranslated region of photosynthetic psbA gene was accomplished. Inverted repeats in chloroplast genomes of the host strain with deleted endogenous psbA genes were selected as recombination targets. Two transformant lines were obtained by dual-phenotypic screening via exhibition of resistance to spectinomycin and restoration of photosynthetic activity. Stable and site-specific integration of intact cry4Ba and psbA genes into chloroplast genomes found in both transgenic lines implied homoplasmy of organelle populations. Achievement in cotranscription of cry4Ba and psbA transgenes revealed by RT-PCR and Northern blot analyses demonstrates the sufficiency of this system’s transcription machinery, offering the further innovation for insecticidal protein production.

The real‐time reverse transcription‐polymerase chain reaction(RT‐PCR)test is the gold standard for severe acute respiratory syndrome coronavirus 2(SARS‐CoV‐2)*** specimen collection and obtaining a sufficient specimen are the most essential steps for laboratory *** nasopharyngeal(NP)swab is recommended as the reference collection ***,NP swab collection is invasive and uncomfortable for patients and poses some risk to healthcare *** study aimed to compare the efficacy of SARS‐CoV‐2 RNA detection from surgical masks with the NP swab method using RT‐PCR *** 269 patients,RT‐PCR RNA from NP swabs was detected among 82 patients(30.5%)and was undetected among 187 patients(69.5%).All patients were tested for SARS‐CoV‐2 RNA from surgical ***‐CoV‐2 RNA was detected in 25/82(30.5%)surgical mask filters,while undetected among 57(69.5%).For the surgical mask with an average use time of 7.05 h,the sensitivity was 30.5%,the specificity was 100.0%,with positive predictive value of 100.0%and negative predictive value of 76.2%.Therefore,surgical masks could be an alternative non‐invasive specimen source for SARS‐CoV‐2 RT‐PCR *** results of our study suggest that the test could be employed after wearing surgical masks for at least 8‐12 h,with increased sensitivity when used for more than 12 h.

0引言。当电机暴露在超过其极限的热应力时,其定子绕组绝缘特性会恶化。通过分析定子线圈的极化和去极化电流(PDC)的特性,了解定子线圈在热应力作用下的绝缘特性变化规律。本文使用了定子线圈的槽部作为研究对象,该线圈来自额定6.6kV、F级(155℃)绝缘的同步电机。根据测试温度100℃、140℃、180℃和220℃,将12根定子线圈分为四组,每个试样在规定的温度下加热4个周期(每个周期6h),在热老化4个周期后进行了PDC测量以分析测试样品的绝缘特性;同时也检测了频域参数如电容比、介损因数DDF,通过合理分析为今后将时域PDC参数转换为频域参数奠定了基础。

The purpose of this study was to develop a calibration model to evaluate the moisture content of tapioca starch using the near infrared(NIR)spectral data in conjunction with partial least square(PIS)*** prediction ability was asessed using a separate prediction data *** groups of tapioca starch samples were used in this study:tapioca starch cake,dried tapioca starch and combined tapioca *** opt imum model obtained from the baseline ofset spectra of dried tapioca starch samples at the outlet of the factory drying process provided a cofficient of determination(R^(2)),standard error of prediction(SEP),bias and residual prediction deviation(RPD)of 0.974,0.16%,-0.092%and 7.4,*** NIR spectroscopy protocol developed in this study could be a rapid method for evaluation of the moisture content of the tapioca starch in factory *** indicated the possibility of real-tine online monitoring and control of the tapioca starch cake feeder in the drying *** addition,it was determined that there was.a stronger influence of the NIR absorption of both water and starch on the prediction of moisture content of the model.

Iron （Fe） has a low solid solubility in aluminum （Al）, and it usually forms Fe-rich intermetallic compounds. Scandium （Sc） is an element that can act as a grain refiner, modify the eutectic silicon and change the morphology of Fe-rich intermetallic compounds at the same time. The present work was conducted to study the effect of Sc on the mechanical properties of Al-7Si-0.3Mg. The alloy was prepared by squeeze casting at two levels of Fe （0.2 and 0.4 wt%） and three levels of Sc （0 wt%, 0.2 wt% and 0.4 wt%）. Sc is found to increase the mechanical properties of the alloy, including its hardness, yield strength and ultimate tensile strength. At 0.2 wt% Fe, adding Sc increases the strength while maintaining good elongation. At 0.4 wt% Fe, adding Sc increases the strength but decreases the elongation slightly. The distributions and morphologies of intermetallic compounds and eutectic silicon affect the elongation. Both Fe-rich intermetallic compounds and Sc-rich intermetallic compounds act as crack initiation sites. The 0.2 wt% Fe ＋ 0.2 wt% Sc alloy has the lowest amount of these intermetallic compounds, and eutectic silicon is small and fibrous. So, it has the highest elongation.

Objective: To find additional factors suggestive of hepatitis C virus(HCV) infection in the general population by using data from a hepatitis virus survey. Methods: This study collected data of HCV infection from a hepatitis virus survey. The survey was conducted in 13 provinces in the northeast Thailand in 2014 and 2015. During the survey, a blood test was performed to screen for HCV. A questionnaire was also distributed to all participants asking about baseline characteristics, risk factors for HCV infection, and daily life activities. Risk factors for HCV infection were executed. Results: There were 2 112 participants for the survey. Of those, 110 participants(5.21%) tested positive for HCV infection. After adjustment by multivariate logistic regression, three factors were significantly associated with HCV infection, namely male gender, age, and family history of liver cancer. The adjusted ORs and 95% CI of these factors were 3.14(1.50, 6.56), 3.78(1.12, 12.76), and 2.28(1.08, 4.80), respectively. Conclusions: Male gender, increasing age, and family history of liver cancer are predictors of HCV infection in endemic areas. Males with a family history of liver cancer in their firstdegree relatives should be tested for HCV infection regardless of symptoms.

BACKGROUND: In emergency conditions, the actual weight of infants and young children are essential for treatments. The RAMATHIBODI Pediatric Emergency Drug Card or RAMA Ped Card has also been developed to estimate actual weight of the subjects. This study aimed to validate the RAMA Ped Card in correctly identifying the actual weight of infants and young ***: This study was a prospective study. We enrolled all consecutive patients under 15 years of age who visited the emergency department(ED). All eligible patients' actual weight and height were measured at the screening point of the ED. The weight of each patient was also measured using the unlabeled RAMA Ped Card. The Cohen's kappa values and agreement percentages were ***: During the study period, there were 345 eligible patients. The RAMA Ped Card had a 61.16% agreement with the actual weight with a kappa of 0.54(P<0.01), while the agreement with the actual height had a kappa of 0.90 and 91.59% agreement. Sub-group analysis found kappa scores with good range in two categories: in cases of accidents and in the infant group(kappa of 0.68 and 0.65, respectively).CONCLUSION: The RAMA Ped Card had a fair correlation with the actual weight in child patients presenting at the ED. Weight estimation in infant patients and children who presented with accidents were more accurate.

Xanthomonas oryzae ***(Xoo)is a widespread pathogen causing bacterial leaf blight(BLB)disease,devastating rice productivity in many cultivated areas of Thailand.A specific and simple method for Xoo detection is required to improve surveillance of disease transmission and *** study developed a recombinase polymerase amplification(RPA)assay assisted with CRISPR-cas12a assay(RAC)for Xoo detection from bacterial cell suspension of infected rice samples without DNA *** efficiency of the RAC system for Xoo detection using either Xoo80 or Xoo4009 locus was optimized to amplify and determine the sensitivity and specificity using a Xoo DNA template from bacterial cell suspension of infected rice samples without DNA *** RAC system using the Xoo4009 locus gave a higher specificity than Xoo80 locus,because only Xoo species was amplified positive RPA product with fluorescence signal by cas12a digestion,which indicated no cross *** RAC using the Xoo4009 locus enabled diagnosis of Xoo presence from both plant extracted samples of Xoo artificially inoculated rice leaves within 3 d post-inoculation without symptomatic BLB appearance,and Xoo naturally infected *** exhibited that RAC using the Xoo4009 locus offered sensitivity,specificity and simplicity for Xoo detection,with low intensities of Xoo-DNA(1×10^(3) copies/μL)and Xoo-cell(2.5×10^(3) cfu/mL).This developed RAC system showed significantly potential for Xoo detection at point-of-care application for early signs of BLB disease outbreak in rice fields.
