Fluorescence Intensity Decay Shape Analysis Microscopy （FIDSAM） for Quantitative and Sensitive Live-Cell Imaging： A Novel Technique for Fluorescence Microscopy of Endogenously Expressed Fusion-Proteins

Fluorescent studies of living plant cells such as confocal microscopy and fluorescence lifetime imaging often suffer from a strong autofluorescent background contribution that significantly reduces the dynamic image contrast and the quantitative access to sub-cellular processes at high spatial resolution. here, we present a novel technique--fluorescence intensity decay shape analysis microscopy （FIDSAM） to enhance the dynamic contrast of a fluorescence image of at least one order of magnitude. The method is based on the analysis of the shape of the fluorescence intensity decay （fluorescence lifetime curve） and benefits from the fact that the decay patterns of typical fluorescence label dyes strongly differ from emission decay curves of autofluorescent sample areas. Using FIDSAM, we investigated Arabidopsis thaliana hypocotyl cells in their tissue environment, which accumulate an eGFP fusion of the plasma membrane marker protein LTI6b （LTI6b-eGFP） to low level. Whereas in conventional confocal fluorescence images, the membranes of neighboring cells can hardly be optically resolved due to the strong autofluorescence of the cell wall, FIDSAM allows for imaging of single, isolated membranes at high spatial resolution. Thus, FIDSAM will enable the sub-cellular analysis of even low-expressed fluorophoretagged proteins in living plant cells. Furthermore, the combination of FIDSAM with fluorescence lifetime imaging provides the basis to study the local physico-chemical environment of fluorophore-tagged biomolecules in living plant cells.

As the first and rate-limiting enzyme of proline degradation, PROLINE DEhYDROGENASE1 （PDh1） is tightly regulated during plant stress responses, including induction under hypoosmolarity and repression under water deficit. The plant receptor histidine kinases AhKs, elements of the two-component system （TCS） in Arabidopsis thaliana, are pro- posed to function in water stress responses by regulating different stress-responsive genes. however, little information is available concerning AhK phosphorelay-mediated downstream signaling. here we show that the Arabidopsis type-B response regulator 18 （ARR18） functions as a positive osmotic stress response regulator in Arabidopsis seeds and affects the activity of the PDh1 promoter, known to be controlled by C-group bZlP transcription factors. Moreover, direct physical interaction of ARR18 with bZIP63 was identified and shown to be dependent on phosphorylation of the conserved aspar- tate residue in the ARR18 receiver domain. We further show that bZlP63 itself functions as a negative regulator of seed germination upon osmotic stress. Using reporter gene assays in protoplasts, we demonstrated that ARR18 interaction negatively interferes with the transcriptional activity of bZIP63 on the PDh1 promoter. Our findings provide new insight into the function of ARR18 and bZIP63 as antagonistic regulators of gene expression in Arabidopsis.

*** In July 2018,the Antarctic community came together to meet at the 13th Workshop on Antarctic Meteorology and Climate(WAMC)in Madison,Wisconsin,USA(Fig.1);and in the following year in June 2019,the 14th WAMC was held in Charleston,South Carolina,USA(Fig.2).With a growing history,the WAMC addresses the topics of Antarctic meteorology and climate(Kameda et al.,2008;Colwell et al.,2016;Lazzara et al.,2018)as well as weather-related issues of scientific and operational *** workshops bring together researchers,operational forecasters,numerical modelers,observational specialists,and *** themes of both workshops included Antarctic meteorological observations,Antarctic atmospheric numerical modeling,Antarctic meteorological and climate research,and Antarctic weather forecasting and operational *** 2018 and 2019 WAMC were both followed by a one-day focus on the Year of Polar Prediction-Southern hemisphere(YOPP-Sh),when preparations and follow-up discussions were made with regard to the YOPP Special Observing Period from 16 November 2018 to 15 February 2019.

Over the past 70 years,the world has witnessed extraordinary growth in crop productivity,enabled by a suite of technological advances,including higher yielding crop varieties,improved farm management,synthetic agrochemicals,and agricultural *** this"Green Revolution"intensified crop production,and is credited with reducing famine and malnutrition,its benefits were accompanied by several undesirable collateral effects(Pingali,2012).These include a narrowing of agricultural biodiversity,stemming from increased monoculture and greater reliance on a smaller number of crops and crop varieties for the majority of our *** reduction in diversity has created vulnerabilities to pest and disease epidemics,climate variation,and ultimately to human health(harlan,1972).
