Dendritic cells(DCs)play a critical role in controlling T helper 2(Th2)cell-dependent diseases,but the signaling mechanism that triggers this function is not fully understood.We showed that p38αactivity in DCs was decreased upon HDM stimulation and dynamically regulated by both extrinsic signals and Th2-instructive cytokines.p38α-specific deletion in cDC1s but not in cDC2s or macrophages promoted Th2 responses under HDM stimulation.Further study showed that p38αin cDC1s regulated Th2-cell differentiation by modulating the MK2−c-FOS−IL-12 axis.Importantly,crosstalk between p38α-dependent DCs and Th2 cells occurred during the sensitization phase,not the effector phase,and was conserved between mice and humans.Our results identify p38αsignaling as a central pathway in DCs that integrates allergic and parasitic instructive signals with Th2-instructive cytokines from the microenvironment to regulate Th2-cell differentiation and function,and this finding may offer a novel strategy for the treatment of allergic diseases and parasitic infection.

Rebuilding insight into the pathophysiology of Alzheimer’s disease through new blood-brain barrier models

The blood-brain barrier is a unique function of the microvasculature in the brain parenchyma that maintains homeostasis in the central nervous system.Blood-brain barrier breakdown is a common pathology in various neurological diseases,such as Alzheimer’s disease,stroke,multiple sclerosis,and Parkinson’s disease.Traditionally,it has been considered a consequence of neuroinflammation or neurodegeneration,but recent advanced imaging techniques and detailed studies in animal models show that blood-brain barrier breakdown occurs early in the disease process and may precede neuronal loss.Thus,the blood-brain barrier is attractive as a potential therapeutic target for neurological diseases that lack effective therapeutics.To elucidate the molecular mechanism underlying blood-brain barrier breakdown and translate them into therapeutic strategies for neurological diseases,there is a growing demand for experimental models of human origin that allow for functional assessments.Recently,several human induced pluripotent stem cell-derived blood-brain barrier models have been established and various in vitro blood-brain barrier models using microdevices have been proposed.Especially in the Alzheimer’s disease field,the human evidence for blood-brain barrier dysfunction has been demonstrated and human induced pluripotent stem cell-derived blood-brain barrier models have suggested the putative molecular mechanisms of pathological blood-brain barrier.In this review,we summarize recent evidence of blood-brain barrier dysfunction in Alzheimer’s disease from pathological analyses,imaging studies,animal models,and stem cell sources.Additionally,we discuss the potential future directions for blood-brain barrier research.

Regarding the social problem of food loss in Japan,about 10%-20%of agricultural products(e.***.vegetables and fruits)are seen as“non-standardized foods”or“surplus foods”by the Agricultural Cooperative Association.Those foods are then discarded.The Tamaki Laboratory of the School of Business,Aoyama Gakuin University is promoting the“SDGs Food Loss Regeneration Project”through industry and academic collaboration—as a research study—to solve this social problem.Therefore,the purpose of this research is the following three subjects:(1)Indicate the design of VCM(value chain management).VCM corresponds to the planning,manufacturing,and sales meetings of“SDGs Moist Vegetable Cake”,and the roles of stakeholders engaged in each area of VCM.(2)Design various BOMs(bill of materials)corresponding to the VCM.Describe how to use BOM and the significance of using each BOM in cooperation.(3)In order to verify the effectiveness of the“SDGs Food Loss Regeneration Project”with“SDGs Moist Vegetable Cake”,conduct and analyze questionnaire surveys and consider the results.

Dynamic External Fixator Combined with Volar Locking Plate Fixation for the Treatment of AO Type C3 Distal Radius Fractures

Background: AO type C3 distal radius fracture (DRF) is the most difficult-to-treat fracture type because of a tendency to shorten and collapse. The purpose of this study is to investigate the clinical and radiographic outcomes of comminuted intra-articular DRFs treated with a dynamic external fixator combined with a volar locking plate (VLP). Methods: Eleven patients (mean age, 61 years) with comminuted intra-articular DRFs were treated with a dynamic type of external fixator combined with a VLP. Following reduction and fixation with a VLP, the dynamic external fixator was applied and the distal ball joint of the fixator was aligned with the lunate-capitate line. The ball joint was unlocked approximately 2 weeks after surgery to allow wrist mobilization. The fixator was removed 3 - 6 weeks (mean, 5 weeks) after surgery. Consequently, clinical and radiographic assessments were carried out at the final follow-up. Results: At the final follow-up, the mean range values of wrist extension and flexion were 76°and 64°, respectively. Compared with the contralateral side, the mean grip strength was 84%. The mean Modified Mayo Wrist Score and the Disabilities of the Arm, Shoulder and Hand score were 88 and 9, respectively. No significant differences in the radiographic parameters exist between after surgery and final follow-up. Conclusions: This study indicated that the dynamic wrist fixator combined with a VLP is effective for the treatment of AO type C3 DRFs.

Comparison between surgical outcomes of colorectal cancer in younger and elderly patients

AIM:To compare the outcome of surgical treatment of colorectal adenocarcinoma in elderly and younger patients.METHODS:The outcomes of 122 patients with colorectal adenocarcinoma who underwent surgical treatment between January 2004 and June 2009 were analyzed.The clinicopathological and blood biochemistry data of the younger group(<75 years) and the elderly group (≥75 years) were compared.RESULTS:There were no significant differences between the two groups in operation time,intraoperative blood loss,hospital stay,time to resumption of oral intake,or morbidity.The elderly group had a significantly higher rate of hypertension and cardiovascular disease.The perioperative serum total protein and albumin levels were significantly lower in the elderly than in the younger group.The serum carcinoembryonic antigen level was lower in the elderly than in the younger group,and there was a significant decreasing trend after the operation in the elderly group.CONCLUSION:The short-term outcomes of surgical treatment in elderly patients with colorectal adenocarcinoma were acceptable.Surgical treatment in elderly patients was considered a selectively effective approach.

Increase in medium-size rainfall events will enhance the C-sequestration capacity of biological soil crusts

Biological soil crusts(BSCs) play important roles in the carbon(C) balance in arid regions. Net C balance of BSCs is strongly dependent on rainfall and consequent activation of microbes in the BSCs. The compensation-rainfall size for BSCs(the minimum rainfall amount for a positive net C balance) is assumed to be different with BSCs of different developmental stages. A field experiment with simulated rainfall amount(SRA) of 0, 1, 5, 10, 20, and 40 mm was conducted to examine the C fluxes and compensation-rainfall size of BSCs in different parts of fixed dunes in the ecotone between the Badain Jaran Desert and the Minqin Oasis. We found algae-lichen crust on the interdunes and crest, algae crust on the leeward side, and lichen-moss crust on the windward. Even a small rainfall(1 mm) can activate both photosynthesis and respiration of all types of BSCs. The gross ecosystem production, ecosystem respiration, and net ecosystem exchange were significantly affected by SRA, hours after the simulated rainfall, position on a dune, and their interactions. The rapid activation of photosynthesis provides a C source and therefore could be responsible for the increase of C efflux after each rewetting. C-uptake and-emission capacity of all the BSCs positively correlated with rainfall size, with the lowest C fluxes on the leeward side. The compensation rainfall for a net C uptake was 3.80, 15.54, 8.62, and 1.88 mm for BSCs on the interdunes, the leeward side, the crest, and the windward side, respectively. The whole dune started to show a net C uptake with an SRA of 5 mm and maximized with an SRA of about 30 mm. The compensation-rainfall size is negatively correlated with chlorophyll content. Our results suggest that BSCs will be favored in terms of C balance, and sand dune stabilization could be sustained with an increasing frequency of 5-10 mm rainfall events in the desert-oasis transitional zone.-

Granulysin and its clinical significance as a biomarker of immune response and NK cell related neoplasms

Granulysin is a cytotoxic granular protein that was identified from human T cells by using the gene subtraction method in 1987. Based on its amino acid homology, granulysin belongs to the saposin-like protein family. The bioactive 9-k Da form of granulysin is processed from the 15-k Da pro-product in the cytoplasmic granules. It is expressed in CD8-positive αβT cells 5 d after mitogenic stimulation and constitutively in natural killer(NK) cells and γδT cells, although regulation of its expression has not yet been precisely determined. The 9-k Da granulysin form has anti-microbial activity against microorganisms such as bacteria, fungi, mycobacteria and parasites, as well as tumoricidal activity against some tumors at 1-10 μmol/L concentrations. Granulysin is secreted in both Ca-dependent and-inde-pendent manners. In sera, only the 15-k Da form is detectable and is expected to be a biomarker for immune potency, acute viral infection, anti-tumor immune reaction, acute graft vs host disease, and NK cell associated neoplasm.

Differential expression of hepatic apurinic/apyrimidinic endonuclease 1,a DNA repair enzyme,in chronic hepatitis

AIM:To determine hepatic expression of apurinic/apyrimidinic endonuclease 1(APE-1)and 8-hydroxydeoxyguanosine(8-OHdG)in patients with chronic hepatitis B and C.METHODS:Liver biopsies were obtained from 27 patients with chronic hepatitis B virus(HBV),30 with chronic hepatitis C virus(HCV),6 with autoimmune hepatitis(AIH),and 6 with primary biliary cirrhosis(PBC).Normal liver tissue was obtained from surgical resection specimens of four patients.Hepatic APE-1 protein and mRNA expression were assayed by Western blot and by real-time polymerase chain reaction,respectively.Hepatocellular APE-1 and 8-OHdG expression were determined by immunohistochemistry.RESULTS:The staining intensity of hepatocellular nuclear APE-1 was lower in the HBV group than in the other groups(P < 0.05).Hepatic APE-1 protein levels were reduced in the HBV group relative to the other groups.Hepatic APE-1 mRNA levels were also lower in the HBV group.The proportion of hepatocytes with 8-OHdG-positive nuclei was increased in the HCV,AIH and PBC groups(P < 0.05),but not in the HBV group.Hepatocellular nuclear APE-1 levels were positively correlated with hepatocellular 8-OHdG levels in both the HBV and HCV groups(HBV,r = 0.34,P < 0.05;HCV,r = 0.54,P < 0.01).CONCLUSION:An imbalance between oxidative DNA damage and APE-1 expression may contribute to hepatocarcinogenesis in chronic viral hepatitis.

Our Japanese industry-university project group of Aoyama Gakuin University investigated and studied new educational programs to cultivate“future strategy design”human resources over several years.The purpose of this research paper is to describe two types of educational programs.The first program proposes the system creator,which is capable of new product design and innovative management that use advanced technologies such as internet of things(IoT),artificial intelligence(AI),and augmented and virtual reality.The second program proposes the business producer that is capable of platform service business modeling,product development and strategy,and customer service.The characteristics of these programs involve applying new educational methods to combine project-based learning(PBL)and active learning.
