Copy number variation profile-based genomic typing of premenstrual dysphoric disorder in Chinese

Premenstrual dysphoric disorder(PMDD) affects nearly 5% of women of reproductive age. Symptomatic heterogeneity, together with largely unknown genetics, has greatly hindered its effective treatment. In the present study, analysis of genomic sequencing-based copy number variations(CNVs) called from 100 kb white blood cell DNA sequence windows by means of semisupervized clustering led to the segregation of patient genomes into the D and V groups, which correlated with the depression and invasion clinical types,respectively, with 89.0% consistency. Application of diagnostic CNV features selected using the correlation-based machine learning method enabled the classification of the CNVs obtained into the D group, V group, total patient group, and control group with an average accuracy of 83.0%. The power of the diagnostic CNV features was 0.98 on average, suggesting that these CNV features could be used for the molecular diagnosis of the major clinical types of PMDD. This demonstrated concordance between the CNV profiles and clinical types of PMDD supported the validity of symptom-based diagnosis of PMDD for differentiating between its two major clinical types, as well as the predominantly genetic nature of PMDD with a host of overlaps between multiple susceptibility genes/pathways and the diagnostic CNV features as indicators of involvement in PMDD etiology.

With the increasing incidence of heavy rainfall events,particularly over the monsoon regions,the highly dense populations are more vulnerable[1].Research initiatives on observation,modeling,and prediction of monsoon heavy rainfall have been promoted actively by World Weather Research Programme's(WWRP)Working Group on Tropical Meteorology Research(WGTMR)of the World Meteorological Organization(WMO)since *** of monsoon-heavy-rainfall workshops were held in Beijing(2011),Petaling Jaya(2012),and New Delhi(2015)to benefit scientists worldwide and forecasters from the National Meteorological and Hydrological *** international Research and Development Project,namely,the Southern China Monsoon Rainfall Experiment(SCMREX)[2]was established in 2013 to coordinate field campaign experiments and to conduct scientific research on presummer(April-June)heavy rainfall processes in southern China.

Background Many studies have shown that positive surgical margin and biochemical recurrence could impact the life of patients with prostate cancer treated with radical prostatectomy. With more and more patients with prostate cancer appeared in recent 20 years in China, it is necessary to investigate the risk of positive surgical margin and biochemical recurrence, and their possible impact on the prognosis of patients treated with radical prostatectomy. In this study, we analyzed the characteristics of patients with prostate cancer who had undergone radical prostatectomy in Macao area and tried to find any risk factor of positive surgical margin and biochemical recurrence and their relationship with the proanosis of these oatients.

Background Asthma is a significant chronic health problem worldwide. Management aims at disease control by reducing functional impairment and exacerbations and improving quality of life (QoL). We report a multi-center study to survey asthma control and QoL in four cities in the Pearl River Delta. Methods The conjoint survey involved ten Hong Kong pediatric hospitals/units, two Shenzhen hospitals, two Macao hos-pitals, and two Guangzhou hospitals on asthma control (using Asthma Control Test) and QoL (Pediatric Allergic Disease Quality of Life Questionnaire, PADQLQ). Acceptability of a treatment is graded as very good/good/fair/poor. Results Good asthma control was only reported in 80% subjects in Hong Kong, but higher in sister cities (85–94%, P<0.001). Allergic rhinitis, 'incense burning', and 'smoker in family' were prevalent among the four cities. Logistic regres-sion showed better control of asthma was associated with better PADQLQ (B=?0.029, P < 0.001), better acceptability of bronchodilator (B=?1.488, P = 0.025), negatively with 'smoker in family' (B=?0.83, P = 0.015) and various PADQLQ domains. Conversely, worse PADQLQ was associated with allergic rhinitis severity (B=4.77, P <0.001), poor control of asthma (B=7.56, P <0.001), increased frequency of traditional Chinese medicine use (B=1.7, P < 0.05), increased fre-quency of bronchodilator usage (B=1.05, P < 0.05), 'smoker in family' (B=4.05, P < 0.05), and incense burning at home (B=3.9, P < 0.05). Conclusions There are some clinical and cultural differences among the four southern Chinese cities within the Guangdong province. This study identifies potentially modifiable environmental and treatment factors associated with poor asthma control and QoL for health-care interventions. Having a smoker in the family is independently associated with poor asthma control and QoL.

Effect of Initiation of Basal Insulin Glargine on Glycemic Control in Patients with Diabetes: Real Life Experience from Hong Kong

Introduction: To assess the changes in glycemic control after initiating or switching to a basal insulin analogue in patients with diabetes mellitus. Methods: A retrospective, observational analysis was conducted using electronic data from a Hong Kong regional hospital. Data from adult patients with type 1 and 2 diabetes mellitus (T1DM and T2DM, respectively) who had been prescribed with basal insulin glargine in 2008-2010, with recorded HbA1c levels at the time of initiation, at 6 and 12 months thereafter, were analysed. Results: Data from 106 eligible patients were analysed. Substantial reduction in HbA1c and fasting sugar levels were reported in both T1DM (Δ HbA1c = 1.5%, Δ FBG = 1.3 mmol/L p < 0.05) and T2DM (Δ HbA1c = 1.2%, Δ FBG = 2.9 mmol/L p < 0.05) patients after 12 months of therapy. A total of 42% of T1DM and 26% of T2DM patients achieved HbA1c levels < 7.0%. After adjustment, T2DM patients who were insulin naive achieved a statistically greater HbA1c reduction (Δ = 1.7%) than those who previous treated with premixed or basal bolus insulin (Δ = 0.3%) (p < 0.05). Percentage of patients experiencing hypoglycaemia reduced from 69% to 62% in T1DM but increased from 26% to 36% in T2DM patients. All hypoglycaemic episodes recorded were either asymptomatic or mild and self-limiting. Only 4% of the patients discontinued treatment at the end of 12 months. Conclusions: In real life clinical practice, a single daily basal insulin analogue therapy provided effective glycemic control with an acceptable risk of mild hypoglycaemia.

Long-Term Trends in Pre-Summer Daytime and Nocturnal Extreme Hourly Rainfall in a Coastal City of South China

The long-term trends in the occurrence frequency of pre-summer daytime and nocturnal extreme hourly rainfall(EXHR) during 1988-2018 in Hong Kong and their spatial distributions are examined and analyzed. Despite a significant increasing trend observed in the occurrence frequency of pre-summer EXHRs during the investigated period,the increase in daytime and nocturnal EXHRs show distinct spatial patterns. Nocturnal EXHRs show uniform increasing trends over the entire Hong Kong. However, the increase in daytime EXHRs is concentrated over the northern or eastern areas of Hong Kong, indicating a downstream shift of pre-summer EXHRs in Hong Kong with regard to the prevailing southwesterly monsoonal flows in south China. The clustering of weather types associated with daytime and nocturnal EXHRs further reveals that the increase in EXHRs over Hong Kong are mainly contributed by the increase of the events associated with southwesterly monsoonal flows with relatively high speeds. During the past few decades, the southwesterly monsoonal flows at coastal south China have undergone a substantial weakening due to the increased surface roughness induced by the urbanization over the Guangdong-Hong Kong-Macao Greater Bay Area since 1990s,leading to enhanced low-level convergence and thus significant increase in EXHRs at coastal south China. Meanwhile,daytime sea-wind circulation at coastal south China is markedly enhanced during the investigated period, which is the main reason for the northward shift of daytime EXHRs in Hong Kong. In addition, the blocked southwesterly monsoonal flows at coastal south China are detoured eastward, leading to stronger convergence and increase in EXHRs at eastern coast of Hong Kong, especially during daytime, when the easterly sea winds prevail at the region.

Alzheimer’s disease(AD),the most common dementia,is characterized by potentially neurotoxic aggregation of Aβ peptide and tau protein,and their deposition as amyloid plaques and neurofibrillary tangles(NFTs).Tau aggregation also occurs in other common neurodegenerative *** dementia(FTD)can be caused by tau mutations that increase the susceptibility of tau to hyperphosphorylation and aggregation,which may cause neuronal dysfunction and deposition of NFTs.17-allylamino-17-demethoxygeldanamycin(17-AAG)is a potent inhibitor of heat shock protein 90(Hsp90),a cytosolic chaperone implicated in the proper folding and functions of a repertoire of client proteins.17-AAG binds to Hsp90 and enhances degradation of Hsp90 client *** sought to determine whether 17-AAG can reduce Aβ and tau pathology in the brains of AD and FTD model mice expressing Aβ or P301L mutant tau,*** were randomized to receive 25,5,or 0 mg/kg 17-AAG thrice weekly from age eight to 11 *** was performed by rotarod test on motor function,on the area occupied by plaques in hippocampus or NFTs in medulla tissue sections,and on mortality.A high dose of 17-AAG tended to decrease NFTs in male mice(p=0.08).Further studies are required to confirm the effect of 17-AAG in diseases of tau aggregation.

Kalmaegi(1415)required the issuance of Gale or Storm Wind Signal No.8 in Hong Kong in 2014,even though it passed by at a distance of 370 km from Hong Kong during its closest *** surges triggered by Kalmaegi caused backflow of sea water in some low lying areas in both Hong Kong and *** paper reviews the use of observational data in monitoring the cyclone characteristics and studies the synoptic factors leading to its fast movement and extensive *** combined analysis of multi-platform satellite wind retrieval,in-situ surface observations and aircraft reconnaissance data over the northern part of the South China Sea is found to be useful in depicting the cyclone *** analysis suggests that the relatively large size of Kalmaegi may be attributed to monsoon shear pattern during its formation stage and the subsequent strengthening of southwesterlies over the northern part of the South China Sea.A strong subtropical ridge north of Kalmaegi not only provides strong steering and thus its high translational speed,but also leads to extensive gale force wind distribution over its northern *** performance of various numerical prediction models in forecasting the movement,intensity change and wind structure of Kalmaegi,as well as the storm surge triggered,is assessed and presented.
