Microbial responses to combined oxidation and catalysis treatment of 1,4-dioxane and co-contaminants in groundwater and soil

Hybrid coronary revascularization vs. percutaneous coronary interventions for multivessel coronary artery disease

Effect of estrogen on manganese-induced toxicity on embryonic astrocytes

Manganese (Mn) is a natural trace metal that is essential for many physiological functions in the human body. Astrocytes in the central nervous system are susceptible reservoirs for Mn accumulation. Estrogen, a steroidal hormone, has been shown to mitigate Mn-induced toxicity in cultures of postnatal astrocytes. However, differences in expression/inducibility of glutamate transporters and glutamine synthetase, transmitters, and the natural gonadal steroids and their receptors are known to occur in astrocyte cultures derived from various stages of fetal and postnatal development. Cultures of embryonic (E18) hippocampal astrocytes were examined in this study for the ability of 17 β-estradiol (E2) to protect them from Mn toxicity by up regulating gene expression of a glutamate transporter. Primary rat hippocampal astrocytes were pretreated with β-Estradiol (E2) in vitro and subsequently, Mn sulfate (MnSO4). The amount of toxic damage to the astrocytes was measured by quantifying glial fibrillary acidic protein (GFAP) with a sandwiched Enzyme-Linked Immunosorbent Assay (ELISA). ELISA analysis indicated Mn exposure at 100 μM, 300 μM, or 600 μM significantly increased GFAP levels. However, E2 concentrations at 10 nM or 30 nM significantly reduced Mn-induced GFAP concentrations at 100 μM. Cells pretreated with 10 nM or 30 nM of E2 significantly lowered GFAP levels. The Water-Soluble Tetrazolium-8 (WST-8) method was utilized to determine cell viability. The WST-8 assay showed that Mn concentrations of 100 μM, 300 μM, or 600 μM significantly reduced the dehydrogenase activity, thereby decreasing the number of viable astrocytes. Enzyme activity with 600 μM of Mn was significantly decreased when compared with 100 μM of Mn, revealing a dose-dependent effect. However, the dehydrogenase activity in cells treated with 600 μM Mn was significantly increased when pretreated with 10 nM of E2. Finite Element Analysis of Osteotomy Treatment for Kienbock’s Disease

Lateral closing wedge osteotomy is a treatment for Kienbock’s disease. It is one of the most frequently used treatment options, which has been reported with relatively good long-term results. However, the results about the treatment are still controversial in some literatures and some key mechanisms are still not clear. The objective of the cur-rent study was to study the biomedical mechanism of the treatment. A finite element model was developed based on the geometry of carpal bones. Various situations in-cluding inclination angle changes by cutting the radial with 0?, 5?, 10? and 15? osteot-omy angles were studied. The effectiveness of the treatment was also studied for the carpal structure with abnormal positions of the lunate bone. The results show that the effectiveness of the stress reduction with the angle depends on many situations such as the initial morphology of the carpal structure.

Oncomorphic <i>TP</i>53 Mutations in Gynecologic Cancers Lose the Normal Protein:Protein Interactions with the microRNA Microprocessing Complex

Mutations in the tumor suppressor TP53 occur in almost all advanced ovarian cancers and in many advanced serous endometrial cancers. Mutations in TP53 can alter the function of the p53 protein, and some mutations result in a mutated protein with oncogenic activity. Previously referred to as gain of function (GOF) p53 proteins, we now term these “oncomorphic” mutations to better describe their function as oncogenes. We reviewed the data from The Cancer Genome Atlas (TCGA) and demonstrate that of the patients diagnosed with endometrial cancer that harbor TP53 mutations, approximately 30% of these mutations are oncomorphic. In ovarian cancer, approximately 20% are oncomorphic. The wild type (WT) p53 protein transactivates genes and micro-RNAs (miRNAs) necessary in the response to cellular stress, which turn off growth and induce apoptosis. In addition to direct transcriptional activation, WT p53 also acts through protein:protein interactions with Drosha and the miRNA processing complex to mediate rapid, enhanced processing of a subset of anti-growth miRNAs. We validated the interaction of WT p53 with the Drosha complex in the cell line UCI-107. We observed that miRNAs that inhibit the expression of oncogenes were induced. Specifically, some miRNAs were induced very rapidly over minutes, consistent with enhanced processing, while others required hours, consistent with transcriptional activation. In contrast, the most common oncomorphic TP53 mutations failed to interact with the Drosha complex and lost the ability to rapidly induce the miRNAs which inhibit oncogene expression. These studies highlight one mechanism underlying the oncomorphic properties of specific TP53 mutations: loss of the enhanced processing of anti-proliferative miRNAs.

This review,which was adapted from a Tenth International Workshop on Tropical Cyclones(IWTC-10)report,discusses research findings and operational practices relevant to cyclone types and phase transitions(extratropical,subtropical,and tropical).The cyclone phase space(CPS)method is widely used in both historical investigations and real-time evaluation of cyclone type and transition;however,CPS parameter values depend on input data resolution,and universal thresholds do not currently exist to delineate when a cyclone transitions from one type to *** of phase transitions in a changing climate highlight potential latitude shifts in extratropical transition and increased potential for tropical transition,but realistic projections of future trends likely require high-resolution simulations that can capture the cyclone warm *** meteorological centers apply varied approaches to cyclone classification via CPS parameters and other criteria,some of which depend on the tropical basin,yet these approaches cannot fully address challenges in operational classification and subsequently in communicating risks associated with these phase *** recommend a multivariate historical assessment of tropical and subtropical cyclones across all basins in which they occur,including the South Atlantic Ocean and the Mediterranean Sea,to identify the potential for a more universal cyclone classification approach that meets operational needs.
