Irisin is a myokine th.t is generated by cleavage of th. membrane protein fibronectin type Ⅲ domain-containing protein 5(FNDC5) in response to ph.sical exercise. Studies reveal th.t irisin/FNDC5 h.s neuroprotective functions against Alzh.imer's disease, th. most common form of dementia in th. elderly, by improving cognitive function and reducing amyloid-β and tau path.logies as well as neuroinflammation in cell culture or animal models of Alzh.imer's disease. Alth.ugh.current and ongoing studies on irisin/FNDC5 sh.w promising results, furth.r mech.nistic studies are required to clarify its potential as a meaningful th.rapeutic target for alleviating Alzh.imer's disease. We recently found th.t irisin treatment reduces amyloid-β path.logy by increasing th. activity/levels of amyloid-β-degrading enzyme neprilysin secreted from astrocytes. h.rein, we present an overview of irisin/FNDC5's protective roles and mech.nisms against Alzh.imer's disease.

Background:Immune ch.ckpoint inh.bitors (ICIs) are effective in a subset of patients with.metastatic solid tumors. h.wever, th. patients wh. would benefit most from ICIs in biliary tract cancer (BTC) are still controversial.Materials and meth.ds:We molecularly ch.racterized tissues and blood from 32 patients with.metastatic BTC treated with.th. ICI pembrolizumab as second-line th.rapy.Results:All patients h.d microsatellite stable (MSS) type tumors. Th.ee of th. 32 patients ach.eved partial response (PR), with.an objective response rate (ORR) of 9.4% (95% confidence interval [CI], 2.0-25.2) and nine sh.wed stable disease (SD), exh.biting a disease control rate (DCR) of 37.5% (95% CI, 21.1-56.3). For th. 31 patients wh. h.d access to PD-1 ligand 1 (PD-L1) combined positive score (CPS) testing (cut-off value ≥1%), th. ORR was not different between th.se wh. h.d PD-L1-positive (PD-L1+; 1/11, 9.1%) and PDL1-(2/20, 10.0%) tumors ( = 1.000). Th. tumor mutational burden (TMB) of PD-L1+ BTC was comparable to th.t of PD-L1-BTC ( = 0.630). TMB and any exonic somatic mutations were also not predictive of pembrolizumab response. Molecular analysis of blood and tumor samples demonstrated a relatively h.gh.natural killer (NK) cell proportion in th. periph.ral blood before pembrolizumab treatment in patients wh. ach.eved tumor response. Moreover, th. tumors of th.se patients presented h.gh.enrich.ent scores for NK cells, antitumor cytokines, and Th. signatures, and a low enrich.ent score for cancer-associated fibroblasts.Conclusions:Th.s study sh.ws th. molecular ch.racteristics associated with.th. efficacy of pembrolizumab in BTC of th. MSS type.

With.th. rise of remote collaboration,th. demand for advanced storage and collaboration tools h.s rapidly ***,traditional collaboration tools primarily rely on access control,leaving data stored on cloud servers vulnerable due to insufficient *** paper introduces a novel mech.nism th.t encrypts data in‘bundle’units,designed to meet th. dual requirements of efficiency and security for frequently updated collaborative *** bundle includes updated information,allowing only th. updated portions to be reencrypted wh.n ch.nges *** encryption meth.d proposed in th.s paper addresses th. inefficiencies of traditional encryption modes,such.as Ciph.r Block Ch.ining(CBC)and Counter(CTR),wh.ch.require decrypting and re-encrypting th. entire dataset wh.never updates *** proposed meth.d leverages update-specific information embedded with.n data bundles and metadata th.t maps th. relationsh.p between th.se bundles and th. plaintext *** utilizing th.s information,th. meth.d accurately identifies th. modified portions and applies algorith.s to selectively re-encrypt only th.se *** approach.significantly enh.nces th. efficiency of data updates wh.le maintaining h.gh.performance,particularly in large-scale data *** validate th.s approach.we conducted experiments measuring execution time as both.th. size of th. modified data and th. total dataset size *** sh.w th.t th. proposed meth.d significantly outperforms CBC and CTR modes in execution speed,with.greater performance gains as data size ***,our security evaluation confirms th.t th.s meth.d provides robust protection against both.passive and active attacks.
