Tissue-specific Tregs in cancer metastasis:opportunities for precision immunotherapy

Decades of advancements in immuno-oncology have enabled the development of current immunotherapies,which provide longterm treatment responses in certain metastatic cancer ***,cures remain infrequent,and most patients ultimately succumb to treatment-refractory metastatic *** insights suggest that tumors at certain organ sites exhibit distinctive response patterns to immunotherapy and can even reduce antitumor immunity within anatomically distant tumors,suggesting the activation of tissue-specific immune tolerogenic mechanisms in some cases of therapy *** immune cells known as regulatory T cells(Tregs)are present within all tissues in the body and coordinate the suppression of excessive immune activation to curb autoimmunity and maintain immune *** the high volume of research on Tregs,the findings have failed to reconcile tissue-specific Treg functions in organs,such as tolerance,tissue repair,and regeneration,with their suppression of local and systemic tumor immunity in the context of immunotherapy *** improve the understanding of how the tissue-specific functions of Tregs impact cancer immunotherapy,we review the specialized role of Tregs in clinically common and challenging organ sites of cancer metastasis,highlight research that describes Treg impacts on tissue-specific and systemic immune regulation in the context of immunotherapy,and summarize ongoing work reporting clinically feasible strategies that combine the specific targeting of Tregs with systemic cancer *** knowledge of Tregs in the framework of their tissue-specific biology and clinical sites of organ metastasis will enable more precise targeting of immunotherapy and have profound implications for treating patients with metastatic cancer.

Magnetohydrodynamic flows tuning in a conduit with multiple channels under a magnetic field applied perpendicular to the plane of flow

In this study, three-dimensional liquid-metal magnetohydrodynamic flows in a conduit with multiple channels under a uniform magnetic field are numerically investigated. The geometry of the conduit is of a four-parallel-channels system including one inflow channel and three outflow channels. The liquid-metal flows into the inflow channel, then turns through 180° in the transi- tion segment, finally flows into three different outflow channels. This kind of channel system can induce counter flow and co-flow, which is rarely investigated before though the conceptual designs of duct flow in the blanket have suggested this type of flow. A structured grid system is chosen after a series of mesh independence tests in the present study. The axial velocity in the side layer near the first partitioning wall, located between the inflow channel and the first outflow channel, is the highest with the lowest electric potential formed therein. The pressure almost linearly decreases in the main flow direction, except in the transition segment. Moreover, the pressure gradient in the first outflow channel is the largest among the three outflow channels. The interdependency of the current, fluid velocity, pressure, electric potential is examined in order to describe the electromagnetic characteristics of the liquid-metal flows.

Optimal deployment schedule of an active twist rotor for performance enhancement and vibration reduction in high-speed flights

The best active twist schedules exploiting various waveform types are sought taking advantage of the global search algorithm for the reduction of hub vibration and/or power required of a rotor in high-speed conditions. The active twist schedules include two non-harmonic inputs formed based on segmented step functions as well as the simple harmonic waveform input. An advanced Particle Swarm assisted Genetic Algorithm（PSGA） is employed for the optimizer. A rotorcraft Computational Structural Dynamics（CSD） code CAMRAD II is used to perform the rotor aeromechanics analysis. A Computation Fluid Dynamics（CFD） code is coupled with CSD for verification and some physical insights. The PSGA optimization results are verified against the parameter sweep study performed using the harmonic actuation. The optimum twist schedules according to the performance and/or vibration reduction strategy are obtained and their optimization gains are compared between the actuation cases. A two-phase non-harmonic actuation schedule demonstrates the best outcome in decreasing the power required while a four-phase non-harmonic schedule results in the best vibration reduction as well as the simultaneous reductions in the power required and vibration. The mechanism of reduction to the performance gains is identified illustrating the section airloads, angle-of-attack distribution, and elastic twist deformation predicted by the present approaches.

Effect of Freeze-Thaw and Urea in Solubility of GPC3-Csub Protein Expressed in Escherichia coli

Glypican-3 is a protein encoded by the Glypican-3 gene located on human X chromosome (Xq26), composed of two subunits, a 40 kDa n-terminal subunit, and a 30 kDa C-terminal subunit. Glypican-3 is a currently potential target molecule for liver cancer treatments because of its over-expression and growth effects on hepatocellular carcinoma (HCC). This study examined the expression and purification of a C-terminal subunit of Glypican-3 protein (GPC3-Csub) due to its application in both diagnosis and therapy for hepatocellular carcinoma. The gene encoding for GPC3-Csub was successfully cloned into plasmid pET28a fused with an affinity tag composed of six consecutive histidine residues (His-tag). Recombinant protein GPC3-Csub was expressed in Escherichia coli BL21 (DE3) in the condition of adding 3% ethanol with IPTG induction. GPC3-Csub was extracted using repeated freeze-thaw cycles with lysozyme, and inclusion bodies were solubilized by 8M Urea, SDS 10% in pH 12. His-tag fused GPC3-Csub proteins allowed it to be purified by affinity chromatography method using the nickel-nitrilotriacetic acid (ni-nTA) column. High expression of GPC3-Csub was confirmed by Coomassie staining and western-blot. GPC3-Csub could be isolated with a ni-nTA column and have a purity of about 90%.

For Al 6k21-T4 overlap weld joint,the shear-tensile strength by using the weaving laser was improved as compared to the case of linear *** the specimen of low strength,the porosity was distributed continuously along the intersection between the plates and fusion ***,for the optimized welding condition,large oval-shaped porosities were located only in the advancing track of the concave ***,the continuity of cracks and porosities played a key role to determine the ***,the weaving width was also the important parameter to control the ***,the concave part had more significant hot and cold cracking in the weld and heat-affected zone（HAZ）,respectively,than the convex part.

盐胁迫下耐盐性木霉菌的筛选与鉴定

对分离于甘肃省民勤县的6株木霉菌进行耐盐性筛选和鉴定。结果表明,随着naCl浓度(0 to 150 mmol/L)增加显著降低6株木霉菌(TG1 to TG6)的菌落直径。与其他5株木霉菌相比,不同浓度naCl胁迫下菌株TG1具有较强耐盐性。当naCl浓度为50 mmol/L时,培养2 d后菌株TG1产孢量和菌落直径与对照相比无显著差异,但是其孢子萌发率增加10.59%。菌株TG1在PDA培养基上菌落正面呈暗绿色,菌落背面呈暗黄色,分生孢子球形至椭圆形。分生孢子梗直角分枝,结合其形态特征将菌株TG1鉴定为***。构建菌株TG1的TEF和ITS系统发育树,结果表明菌株TG1的TEF和ITS序列分别与*** IMI 297702(EU401578.1)和***(MT889709.1)序列相似性为99.22%和100.00%。因此,结合形态特征和分子生物学分析将菌株TG1鉴定为*** TG1。与对照相比,菌株TG1在50、100和150 mmol/L naCl胁迫下产生IAA浓度分别为1.25、1.35和1.56μg/mL,而对照为0.92μg/mL。同时,在50、100和150 mmol/L naCl胁迫下,TG1菌株ACC脱氨酶活性分别为0.1、0.16和0.17μmolα-ketobutyrate mg/h,而对照为0.09μmolα-ketobutyrate mg/h。

Quantitative analysis of Arctic ice flow acceleration with increasing temperature

This study explores the ice flow acceleration(21.1%)of Pedersenbreen during 2016–2017 after the extremely warm winter throughout the whole Arctic in 2015/2016 using in situ data and quantitatively analyses the factors contributing to this *** data sets,including 2008–2018 air temperature data from ny-?lesund,ten-year in situ GPS measurements and Elmer/Ice ice flow modelling under different ice temperature scenarios,suggest that the following factors contributed to the ice flow acceleration:the softened glacier ice caused by an increase in the air temperature(1.5℃)contributed 2.7%–30.5%,while basal lubrication contributed 69.5%–97.3%.The enhanced basal sliding was mostly due to the increased surface meltwater penetrating to the bedrock under the rising air temperature conditions;consequently,the glacier ice flow acceleration was caused mainly by an increase in subglacial *** Pedersenbreen,there was an approximately one-year time lag between the change in air temperature and the change in glacier ice flow velocity.
