＜正＞ BACKGROUND Voglibose is an alpha- glucosidase inhibitor. Due to negligible oral absorption, measuring drug concentration in the blood is impractical. So we proposed a pharmacodynamic assessment method to reflect drug effect, and this study aimed to validate this *** A placebo - controlled, selective two - period clinical study was conducted in 20 healthy male subjects. Period Ⅰ: On day 1. subjects received a placebo and a sucrose - rich fluid meal 20 min after dosing. Blood samples were taken during 3 hours. On day 2. subjects received 0.3 mg voglibose instead of *** Ⅱ: 9 subjects in whom effects of drug were observed in Period Ⅰ participated in a multiple dose study （placebo: 8. 11 pm on day - 1. and 9 am on day 1 / voglibose: 2. 8. 11 pm on day 1. and 9 am on day 2）. RESULTS The average percent decreases of AUEClh （area under the serum glucose level- time curve to 1h） and Gmax （maximum serum glucose level） were 19.6% （P＜0.001） and 22.2% （P＜0.001）. respectively. CONCLUSIONS Significant drug effects of voglibose were revealed after multiple doses. Changes of AUEClh and Gmax compared to placebo may be alternative parameters to AUC and Cmax for an equivalence study.

＜正＞ CKD-501 is a novel peroxisome proliferator-activated receptor α/γ dual agonist for the treatment of diabetes mellitus. This study aimed to investigate safety, tolerability, and pharmacokinetics of CKD-501 in healthy volunteers. A randomized, placebo-controlled, double-blind, parallel-group, dose-rising study was performed. Thirty-six healthy male subjects received single oral doses ranging from 0.5-8 mg CKD- 501 or placebo. In the multipledose study, 24 subjects received 1-4 mg once daily for 7 days. Serial blood and urine samples were collected. No serious adverse events （AEs） were observed and AEs reported were all of mild severity. In the single-dose study, mean Cmax and AUCinf increased linearly up to the 8 mg dose *** and t1/2 ranged from 0.5-4 h and 7.8-9.8 h. respectively. Less than 1 % CKD-501 was excreted in urine. After multiple dosing, accumulation index was around 1.2. Mean apparent clearance, Tmax and t1/2 in steady state were independent of dose and time. Single oral doses up to 8 mg CKD- 501 and multiple doses up to 4 mg were safe and well tolerated. Mean Cmax and AUCinf were dose proportional and there was no remarkable accumulation after multiple dosing.

The purpose of this study is to establish a gene delivery system for interstitial tissue-specific protein expression in mice testes using modified recombinant baculovirus. Green fluorescent protein （GFP）-expressing recombinant baculovirus （GFP-baculovirus）, in which the insect cell-specific polyhedron promoter was replaced by the cytomegalovirus （CMV）-IE promoter, was used to transfect testicular cells in vitro, and for intra-tunica albuguineal injection of the interstitial tissue of the testis. GFP expression was monitored in frozen testes sections by fluorescence microscopy. Expression of GFP in testicular tissues was also assessed by reverse transcription polymerase chain reaction （RT-PCR）, and protein expression was assessed by Western blot. Testicular cells in vitro were infected efficiently by modified recombinant GFP-baculovirus. lntra-tunica albuguineal injection of GFP- baculovirus into the mouse testis resulted in a high level of GFP expression in the interstitial tissues. RT-PCR analysis clearly showed GFP gene expression in the testis, particularly interstitial tissues. Intra-tunica albuguineal injection of a modified baculovirus that encoded recombinant rat insulin-like growth factor binding protein （IGFBP）-5 resulted in an increase in IGFBP-5 in testis and semen. In conclusion, we have developed an efficient delivery system for gene expression in vivo in testicular cells, particularly cells of the interstitial tissue using intratunica albuguineal injection of a modified recombinant baculovirus. This method will be particularly relevant for application that requires gene delivery and protein expression in the testicular cells of the outer seminiferous tubule of the testis.

Blockade of programmed death-1(PD-1)reinvigorates exhausted CD8^(+)T cells,resulting in tumor regression in cancer ***,reinvigoration of exhausted CD8^(+)T cells following PD-1 blockade was shown to be CD28-dependent in mouse ***,we examined the role of CD28 in anti-PD-1 antibody-induced human T cell reinvigoration using tumor-infiltrating CD8^(+)T cells(CD8^(+)TILs)obtained from non-small-cell lung cancer ***-cell analysis demonstrated a distinct expression pattern of CD28 between mouse and human CD8^(+)***,we found that human CD28^(+)CD8^(+)but not CD28^(–)CD8^(+)TILs responded to PD-1 blockade irrespective of B7/CD28 blockade,indicating that CD28 costimulation in human CD8^(+)TILs is dispensable for PD-1 blockade-induced reinvigoration and that loss of CD28 expression serves as a marker of anti-PD-1 antibody-unresponsive CD8^(+)*** and phenotypically,PD-1 blockade-unresponsive human CD28^(–)PD-1^(+)CD8^(+)TILs exhibited characteristics of terminally exhausted CD8^(+)T cells with low TCF1 ***,CD28^(–)PD-1^(+)CD8^(+)TILs had preserved machinery to respond to IL-15,and IL-15 treatment enhanced the proliferation of CD28^(–)PD-1^(+)CD8^(+)TILs as well as CD28^(+)PD-1^(+)CD8^(+)*** together,these results show that loss of CD28 expression is a marker of PD-1 blockade-unresponsive human CD8^(+)TILs with a TCF1–signature and provide mechanistic insights into combining IL-15 with anti-PD-1 antibodies.

Laboratory investigation of the anisotropic confinement-dependent brittle-ductile transition of a Utah coal

Development of a fault-rupture environment in 3D: A numerical tool for examining the mechanical impact of a fault on underground excavations

While faults are commonly simulated as a single planar or non-planar interface for a safety or stability analysis in underground mining excavation, the real 3D structure of a fault is often very complex, with different branches that reactivate at different times. Furthermore, these branches are zones of nonzero thickness where material continuously undergoes damage even during interseismic periods. In this study, the initiation and the initial evolution of a strike-slip fault was modeled using the FLAC3D software program. The initial and boundary conditions are simplified, and mimic the Riedel shear experiment and the constitutive model in the literature. The FLAC3D model successfully replicates and creates the 3D fault zone as a strike-slip type structure in the entire thickness of the model. The strike-slip fault structure and normal displacement result in the formation of valleys in the model. Three panels of a longwall excavation are virtually placed and excavated beneath a main valley. The characteristics of stored and dissipated energy associated with the panel excavations are examined and observed at different stages of shear strain in the fault to evaluate bump potential. Depending on the shear strain in the fault, the energy characteristics adjacent to the longwall panels present different degrees of bump potential, which is not possible to capture by conventional fault simulation using an interface.

Is the filamentary dark cloud GF 6 a star forming region?--Stability analysis and infrared properties

We present the results of mapping observations and stability analyses toward the filamentary dark cloud GF 6. We investigate the internal structures of a typical filamentary dark cloud GF 6 to know whether the filamentary dark cloud will form stars. We perform radio observations with both 12CO（J =1–0） and 13CO（J =1–0） emission lines to examine the mass distribution and its evolutionary *** 13CO gas column density map shows eight subclumps in the GF 6 region with sizes on a sub-pc scale. The resulting local thermodynamic equilibrium masses of all the subclumps are too low to form stars against the turbulent dissipation. We also investigate the properties of embedded infrared point sources to know whether they are newly formed stars. The infrared properties also indicate that these point sources are not related to star forming activities associated with GF 6. both radio and infrared properties indicate that the filamentary dark cloud GF 6 is too light to contract gravitationally and will eventually be dissipated away.

Radiologic factors related to double-bar insertion in minimal invasive repair of pectus excavatum

Background:Pectus excavatum is the most common congenital chest wall deformity,with a high incidence in live *** study aimed to evaluate the measured factors on CT images related to the number of pectus bars for surgical ***:A total of 497 patients who had undergone minimally invasive repair between April 2007 and July 2011 were classified into single-bar(n=358)and doublebar(n=139)insertion *** measured eight distinct distances and one angle on CT scans to reflect quantitative *** analysis and multivariate logistic regression analysis were performed to detect statistically significant association between radiologic measurements and the pectus bars ***:After adjusting for age and gender,the transverse distance(T),the transverse distance of the depression area(A),the inclined distance of the depression area(B),the AP distance of the depression area(C),the depression angle(G),and the eccentric distance of deformity(E)were significantly correlated with double-bar *** regression model showed that age(,P<0.0001),gender(P<0.0001),depression angle(G)(P<0.0001),direction of the depression(DD)(P<***1)and depression depth(D)(P<0.0001)were significantly associated with doublebar ***:CT scan provides useful factors which can be of assistance in predicting the number of pectus bars for the surgical correction of pectus excavatum.

Mg has received much attention as a next-generation implantable material owing to its biocompatibility,bone-like mechanical properties,and biodegradability in physiological *** application of various polymer coatings has been conducted in the past to reduce the rapid formation of hydrogen gas and the local change in pH during the initial phase of the chemical reaction with the body ***,we propose femtosecond(fs)laser-mediated Mg surface patterning for significant enhancement of the binding strength of the coating material,which eventually reduces the corrosion *** of the structural,physical,crystallographic,and chemical properties of the Mg surface have been conducted in order to understand the mechanism by which the surface adhesion increases between Mg and the polymer coating *** on the fs laser conditions,the surface structure becomes rough owing to the presence of several microscaled pits and grooves of nanoporous MgO,resulting in a tightly bonded poly(lactic-co-glycolic acid)(PLGA)*** corrosion rate of the PLGA-coated,fs laser-treated Mg is considerably slow compared with the non-treated Mg;the treated Mg is also more biocompatible compared with the non-treated *** fs laser-based surface modification technique offers a simple and quick method for introducing a rough coating on Mg;further,it does not require any chemical treatment,thereby overcoming a potential obstacle for its clinical use.
