Bone marrow-derived mesenchymal stem cell(MSC)is one of the most actively studied cell types due to its regenerative potential and immunomodulatory *** cell expansion methods using 2D tissue culture plates and 2.5D microcarriers in bioreactors can generate large cell numbers,but they compromise stem cell potency and lack mechanical preconditioning to prepare MSC for physiological loading expected in *** overcome these challenges,in this work,we describe a 3D dynamic hydrogel using magneto-stimulation for direct MSC manufacturing to *** our technology,we found that dynamic mechanical stimulation(DMS)enhanced matrix-integrinβ1 interactions which induced MSCs spreading and *** addition,DMS could modulate MSC biofunctions including directing MSC differentiation into specific lineages and boosting paracrine activities(e.g.,growth factor secretion)through YAP nuclear localization and FAK-ERK *** our magnetic hydrogel,complex procedures from MSC manufacturing to final clinical use,can be integrated into one single platform,and we believe this‘all-in-one’technology could offer a paradigm shift to existing standards in MSC therapy.

Despite the global administration of approved COVID-19 vaccines(e.g.,ChAdOx1 nCoV-19?,mRNA-1273?,BNT162b2?),the number of infections and fatalities continue to rise at an alarming rate because of the new variants such as Omicron and its *** COVID-19 vaccines that are licensed for human use,most of the vaccines that are currently in clinical trials are administered via parenteral ***,it has been proven that the parenteral vaccines do not induce localized immunity in the upper respiratory mucosal surface,and administration of the currently approved vaccines does not necessarily lead to sterilizing *** further supports the necessity of a mucosal vaccine that blocks the main entrance route of COVID-19:nasal and oral mucosal *** the mechanism of immune regulation of M cells and dendritic cells and targeting them can be another promising approach for the successful stimulation of the mucosal immune *** paper reviews the basic mechanisms of the mucosal immunity elicited by mucosal vaccines and summarizes the practical aspects and challenges of nanotechnology-based vaccine platform development,as well as ligand hybrid nanoparticles as potentially effective target delivery agents for mucosal vaccines.

Probing temperature-dependent interlayer coupling in a MoS2/h-BN heterostructure

Stacking of atomically thin layers of two-dimensional materials has revealed extraordinary physical phenomena owing to van der Waals(vdW)interaction at the ***,most of the studies focused on the transition metal dichalcogenide(TMD)/TMD heterostructure,while the interlayer coupling of the TMD/hexagonal boron nitride(h-BN)heterostructure has not been extensively explored despite its *** this study,the temperature-dependent interlayer coupling is demonstrated in a heterostructure of molybdenum disulfide(MoS2)and *** interface between MoS2 and the insulating substrate exerts a significant spectroscopic impact on MoS2 through substrate-induced local strain,charged impurity,and vdW *** non-resonant conditions,temperature-dependent peak shifts in Raman and photoluminescence(PL)spectra of MoS2 reveal the evolution of interlayer *** frequencies and PL peak energies at different temperatures demonstrate how substrate-induced strain,impurity,and vdW interactions at the interface influence phonon vibration and excitonic transition of *** resonant conditions at low temperature,anomalous Raman modes appear in the MoS2/h-BN heterostructure because of the enhanced electron-phonon coupling and vdW *** anomalous Raman modes are quantitatively investigated by the deconvolution of the resonance Raman spectra and described by interlayer coupling at low temperature,in agreement with complementary indications from the temperature-dependent evolution of non-resonant Raman and PL spectra.

Two-dimensional(2D)materials have recently provided a new perspective on optoelectronics because of their unique layered structure and excellent physical ***,their potential use as optoelectric devices has been limited by the trade-off between photoresponsivity and response ***,based on a vertically stacked atomically thin p-n junction,we propose a gap-mode plasmon structure that simultaneously enables enhanced responsivity and rapid *** atomically thin 2D materials act as a spacer for enhancing the gap-mode plasmons,and their short transit length in the vertical direction allows fast photocarrier *** demonstrate a high responsivity of up to 8.67 A/W with a high operation speed that exceeds 35 MHz under a 30 nW laser *** photocurrent,absorption,and a numerical simulation are used to verify the effectiveness of the gap-mode plasmons in the *** believe that the design strategy proposed in this study can pave the way for a platform to overcome the trade-off between responsivity and response time.

In 2008, the very extensive tropical peats were estimated to be about 182 million ha spanning South America, Asia and Africa. About 20.3%(36.9 million ha) of this area exist in Asia. Peats are classified based on their degree of decomposition, namely Fibrists, Hemists, Saprists and Folists. This makes them different in characteristics. The activities of microorganisms vary in different types of peat due to, for example, the sapric layer of well humified peat can provide water and food to microorganisms during heat stress. In another scenario, deeper peat is older and typically has lower levels of labile carbon to provide substrate for microbes compared to surface peat. A complete understanding of the microbial communities in different layers of peat is essential as microorganisms play major roles in peat decomposition and are important to ecosystem processes. These peats are a very important global carbon(C)store or reserve and could severely impact climate change if not managed well. Peatlands can store as much as 40 to 90 Gt C. Mis-management of peats could severely impact the environment particularly the emission of carbon into the atmosphere. For instance, clearing of peatlands using fire has been reported to release an estimated 88 t C ha^(-1) to the atmosphere. There are several factors which influence the environmental consequences of tropical peat especially in relation to climate change. The main influences are:(i) changes in temperature,(ii) changes in precipitation or rainfall,(iii) changes in atmospheric composition, and(iv) fire and haze. This paper is a brief review on these four influences in relation to climate change. It is apparent from the brief review that there is a need for continued short and long-term research to better understand tropical peats and how they affect our climate. This will hopefully provide the basis for predicting better what could happen under various scenarios.

Selective inhibition of cell growth by activin in SNU-16 cells

AIM： To investigate whether activin regulates the cell proliferation of human gastric cancer cell line SNU-16 through the mRNA changes in activin receptors, Smads and p21^CIP1/WAF1. METHODS： The human gastric cancer cell lines were cultured, RNAs were purified, and RT-PCRs were carried out with specifically designed primer for each gene. Among them, the two cell lines SNU-5 and SNU-16 were cultured with activin A for 24, 48 and 72 h. The cell proliferation was measured by MTT assay. For SNU-16, changes in ActRIA, ActRIB, ActRIIA, ActRIIB, Smad2, Smad4, Smad7, and p21^CIP1/WAF1 mRNAs were detected with RT-PCR after the cells were cultured with activin A for 24, 48 and 72 h. RESULTS： The proliferation of SNU-16 cells was down regulated by activin A whereas other cells showed no change. Basal level of inhibin/activin subunits, activin receptors, Smads, and p21^CIP1/WAF1 except for activin βB mRNAs was observed to have differential expression patterns in the human gastric cancer cell lines, AGS, KATO III, SNU-1, SNU-5, SNU-16, SNU-484, SNU-601, SNU-638, SNU-668, and SNU-719. Interestingly, significantly higher expressions of ActR IIA and IIB mRNAs were observed in SNU-16 cells when compared to other cells. After activin treatment, ActR IA, IB, and IIA mRNA levels were decreased whereas ActR IIB mRNA level increased in SNU-16 cells. Smad4 mRNA increased for up to 48 h whereas Smad7 mRNA increased sharply at 24 h and returned to the initial level at 48 h in SNU-16 cells. In addition, expression of the p21^CIP1/WAF1 the mitotic inhibitor, peaked at 72 h after activin treatment in SNU-16 cells. CONCLUSION： Our results suggest that inhibition of cell growth by activin is regulated by the negative feedback effect of Smad7 on the activin signaling pathway, and is mediated through p21^CIP1/WAF1 activation in SNU-16 cells.

Gene expression profiles of the one-carbon metabolism pathway

One-carbon metabolism plays a critical role in both DNA methylation and DNA synthesis. Accumulating evidence has shown that interruptions of this pathway are associated with many disease outcomes including cardiovascular diseases and cancers. Mechanistic studies have been performed on genetic polymorphisms involved in one-carbon metabolism. However, expression profiles of these inter-related genes are not well-known. In this study, we examined the gene expression profiles of 11 one-carbon metabolizing genes by quantifying the mRNA level of the lymphocyte among 54 healthy individuals and explored the correlations of these genes. We found these genes were expressed in lymphocytes at moderate levels and showed significant inter-person variations, We also applied principle component analysis to explore potential patterns of expression. The components identified by the program agreed with existing knowledge about one-carbon metabolism. This study helps us better understand the biological functions of one-carbon metabolism.

In a prior practice and policy article published in Healthcare Science,we introduced the deployed application of an artificial intelligence(AI)model to predict longer‐term inpatient readmissions to guide community care interventions for patients with complex conditions in the context of Singapore's Hospital to Home(H2H)program that has been operating since *** this follow on practice and policy article,we further elaborate on Singapore's H2H program and care model,and its supporting AI model for multiple readmission prediction,in the following ways:(1)by providing updates on the AI and supporting information systems,(2)by reporting on customer engagement and related service delivery outcomes including staff‐related time savings and patient benefits in terms of bed days saved,(3)by sharing lessons learned with respect to(i)analytics challenges encountered due to the high degree of heterogeneity and resulting variability of the data set associated with the population of program participants,(ii)balancing competing needs for simpler and stable predictive models versus continuing to further enhance models and add yet more predictive variables,and(iii)the complications of continuing to make model changes when the AI part of the system is highly interlinked with supporting clinical information systems,(4)by highlighting how this H2H effort supported broader Covid‐19 response efforts across Singapore's public healthcare system,and finally(5)by commenting on how the experiences and related capabilities acquired from running this H2H program and related community care model and supporting AI prediction model are expected to contribute to the next wave of Singapore's public healthcare efforts from 2023 *** the convenience of the reader,some content that introduces the H2H program and the multiple readmissions AI prediction model that previously appeared in the prior Healthcare Science publication is repeated at the beginning of this article.

东南亚五加科包含14个属约500种,本文应用ITS片段对该区五加科植物的进化关系作了初步研究。研究显示该地区五加科植物具有复杂的起源,很多属属于亚洲掌状复叶类群或Hedereae族的一支中。该区特有类群Harmsiopanax形态上非常特殊,但其系统位置尚未不明朗。在Brassaiopsis属中,有几种形态差异较大的种,但它们属同一单系,加之各种间ITS序列差异较小,故应是新近起源于马来亚半岛和苏门达腊岛的种类。Wardenia simplex聚类在Brassaiopsis一支中,故不支持将Wardenia作为独立的属。东南亚地区对于Schefflera属的发育非常重要,已有的证据显示该区的Schefflera属植物属于该属的Heptapleurum类群。马来亚与泰国南部的Dendropanax lancifolius并没有与Dendropanax属的核心类群聚在一起,其系统地位需进一步研究。Macropanax maingayi是非常特殊的一个种,曾被独立分出,成立了单种属Hederopsis。本文的分析清楚表明它属于Macropanax属。Aralia merrillii因为其不同寻常的攀缘特性而被独立出来,建立了单种属Acanthophora,但ITS序列分析支持将它置于Aralia属中。新增的取样继续支持Arthrophyllum的单系性。Os-moxylon的原初分布范围在东南亚,它是五加科系统进化树上孤立的类群。

Development and Validation of a High-Density SNP Genotyping Array for African Oil Palm

High-density single nucleotide polymorphism （SNP） genotyping arrays are powerful tools that can measure the level of genetic polymorphism within a population. To develop a whole-genome SNP array for oil palms, SNP discovery was performed using deep resequencing of eight libraries derived from 132 Elaeis guineen- sis and Elaeis oleifera palms belonging to 59 origins, resulting in the discovery of 〉3 million putative SNPs. After SNP filtering, the Illumina OP200K custom array was built with 170 860 successful probes. Phenetic clustering analysis revealed that the array could distinguish between palms of different origins in a way consistent with pedigree records. Genome-wide linkage disequilibrium declined more slowly for the commercial populations （ranging from 120 kb at r2 = 0.43 to 146 kb at r2 = 0.50） when compared with the semi-wild populations （19.5 kb at r2 = 0.22）. Genetic fixation mapping comparing the semi-wild and commercial population identified 321 selective sweeps. A genome-wide association study （GWAS） detected a significant peak on chromosome 2 associated with the polygenic component of the shell thick- ness trait （based on the trait shell-to-fruit; S/F %） in tenera palms. Testing of a genomic selection model on the same trait resulted in good prediction accuracy （r = 0.65） with 42% of the S/F % variation explained. The first high-density SNP genotyping array for oil palm has been developed and shown to be robust for use in genetic studies and with potential for developing early trait prediction to shorten the oil palm breeding cycle.
