The study was sought to enhance the synthesis of thermal stableβ-cyclodextrin glycosyltransferase(β-CGTase)using potato wastewater as a low-cost medium and assess the degree to which it is efficient for industrial production ofβ-cyclodextrin(β-CD)from raw potato *** bacteria producingβ-CGTase was isolated from Saudi Arabia and the promising strain was identified as Bacillus licheniformis using phylogenetic analysis of the 16S rRNA ***-encapsulated cultures exhibited twice-fold ofβ-CGTase production more than free *** electron microscopy(SEM)of polymeric capsules indicated the potential for a longer shelf-life,which promotes the restoration of activity in bacterial cells across semi-continuous fermentation ofβ-CGTase production for 252 *** optimal conditions forβ-CGTase synthesis using potato wastewater medium were at 36 h,pH of 8.0,and 50°C with 0.4%potato starch and 0.6%yeast extract as carbon and nitrogen sources,*** purified enzyme showed a specific activity of 63.90 U/mg with a molecular weight of∼84.6 kDa as determined by SDS-PAGE *** high enzyme activity was observed up to 60°C,and complete stability was achieved at 75°*** levels of activity and stability were shown at pH 8.0,and the pH range from 7.0–10.0,*** enzyme has an appreciable affinity for raw potato starch with a Km of 5.7×10−6 M and a Vmax of 87.71μmoL/mL/min.β-CD production was effective against 25 U/g of raw potato *** outcomes demonstrated its feasibility to develop a fermentation process by integrating the cost-effective production ofβ-CGTase having distinctive properties forβ-CD production with ecofriendly utilization of potato wastewater.

Complications and survival in patients undergoing colonic stenting for malignant obstruction

AIM:To investigate whether predicting patients that might be at a higher risk for complications might serve to improve the selection of patients undergoing colonic ***:A retrospective review of consecutive patients who underwent an attempted self-expandable metal stent(SEMS)insertion for malignant colonic obstruction between November 2006 and March *** patients were either referred for preoperative colonic decompression with the intent of a single surgical procedure,or for palliation of the malignant colorectal obstruction for unresectable ***’s test orχ2test was performed on categorical variables,and the t test for continuous *** and multivariable logistic regression were used to examine the association between independent variables and the presence of complications from SEMS ***:SEMS insertion was attempted in 73 *** comprised 55.71%and the mean age was 67.41±12.41 *** these,65.15%underwent subsequent surgery,while 34.85%received SEMS as palliation for advanced *** tumors were only4.76%.The majority of patients had stageⅣdisease(63.83%),while the remainder had stageⅢ(36.17%).SEMS were successfully inserted in 93.85%(95%CI:87.85%-99.85%).Perforations occurred in 4.10%,SEMS migration in 8.21%,and stent re-occlusion from ingrowth occurred in 2.74%of *** mean duration of follow up for the patients was 13.52±17.48 mo(range 0-73 mo).None of the variables:age,sex,time between the onset of symptoms to SEMS insertion,time between SEMS insertion and surgery,length of the stenosis,location of the stenosis,albumin level,or receiving neoadjuvant chemotherapy,could predict the development of complications from either SEMS insertion nor prolonged ***:None of the variables could predict the development of complications or *** studies are required to identify patients who would benefit the most from SEMS.

Sunitinib is an orally administered, multi-target tyrosine kinase inhibitor that has been approved by the FDA for the treatment of renal cell carcinoma and imatinib resistant gastro-intestinal tumors. Anti-leukemic activity of sunitinib has been examined in early clinical trials with limited success. However, recent trials on acute myeloid leukemia (AML) patients carrying FLT3 mutations have shown promising results. Effects of sunitinib on leukemic clonogenic cells and potential leukemic stem cells have not been examined so far. We analyzed the anti-proliferative and apoptotic properties of sunitinib on AML-derived cell lines. We also tested the effect of sunitinib on AML patient derived clonogenic cells (AML-CFC), as well as flow-sorted potential leukemic progenitors. Peripheral blood or bone marrow samples were obtained from newly diagnosed AML patients and flow sorted for CD34+ CD133+ or ALDH+ cells. Umbilical cord blood derived CD34+ cells were used as normal controls. Sunitinib induced growth arrest and apoptosis in AML derived cell lines. In addition, 7 μM sunitinib induced 75% reduction of AML-CFC as compared to DMSO treated control (±6.79%;n = 4). In contrast, 7 μM sunitinib treatment of umbilical cord blood derived normal CD34+ cells showed 29% reduction in AML-CFC (±6.77%;n = 5). Treatment of ALDH+ cells sorted from 2 AML cases and CD34+ CD133+ cells from one patient showed reduction of AML-CFC on treatment with sunitinib. Our study highlighted a potent anti-proliferative and proapoptotic effect of sunitinib on AML cell lines, AML patient derived clonogenic cells and potential leukemic stem cells.

Epidemiology and Patterns of Malignant Lymphoma in Northern Saudi Arabia

Objective: The objective of this study was to appraise the epidemiology and patterns of malignant lymphoma in Northern Saudi Arabia. Methodology: A full reportage of data regarding malignant lymphoma was retrieved from the King khalid Hospital, Hail, Northern Saudi Arabia, comprising records of malignant lymphoma from 2009 to 2016. Results: A review of 120 blood cancers records revealed 47/120 (39.2%) cases of lymphomas, 39 (83%) were Non-Hodgkin lymphoma (NHL), 6 (12.8%) were HL and 2 (4.2%) B-cell lymphomas. The overall crude incidence rate CIR) of lymphomas (from 2009 to 2016) was 6.7 per 100,000 persons of the Hail region population. The mean CIR per year was 0.84 per 100,000 persons. Conclusion: The prevalence of malignant lymphoma is relatively higher in northern Saudi Arabia with predominance of NHL amongst elder population.

The novel coronavirus,SARS-CoV-2,which has caused millions of death globally is recognized to be unstable and recalcitrant in the environment,especially in the way it has been evolving to form new and highly transmissible *** particular concerns are human-environment interactions and the handling and reusing the environmental materials,such as effluents,sludge,or biosolids laden with the SARS-CoV-2 without adequate treatments,thereby suggesting potential transmission and health *** study assesses the prevalence of SARS-CoV-2 RNA in effluents,sludge,and ***,we evaluate the environmental,ecological,and health risks of reusing these environmental materials by wastewater/sludge workers and farmers.A systematic review of literature from the Scopus database resulted in a total of 21 articles(11 for effluents,8 for sludge,and 2 for biosolids)that met the criteria for meta-analysis,which are then subdivided into 30 meta-analyzed *** prevalence of SAR-CoV-2 RNA in effluent and sludge based on random-effect models are 27.51 and 1012.25,respectively,with a 95%CI between 6.14 and 48.89 for the effluent,and 104.78 and 1019.71 for the ***,the prevalence of SARS-CoV-2 RNA in the biosolids based on the fixed-effect model is 30.59,with a 95%CI between 10.10 and *** prevalence of SARS-CoV-2 RNA in environmental materials indicates the inefficiency in some of the treatment systems currently deployed to inactivate and remove the novel virus,which could be a potential health risk concern to vulnerable wastewater workers in particular,and the environmental and ecological issues for the population at *** timely review portends the associated risks in handling and reusing environmental materials without proper and adequate treatments.
