Mapping the metabolic responses to oxaliplatin-based chemotherapy with in vivo spatiotemporal metabolomics

Adjuvant chemotherapy improves the survival outlook for patients undergoing operations for lung metastases caused by colorectal cancer (CRC). However, a multidisciplinary approach that evaluates several factors related to patient and tumor characteristics is necessary for managing chemotherapy treatment in metastatic CRC patients with lung disease, as such factors dictate the timing and drug regimen, which may affect treatment response and prognosis. In this study, we explore the potential of spatial metabolomics for evaluating metabolic phenotypes and therapy outcomes during the local delivery of the anticancer drug, oxaliplatin, to the lung. 12 male Yorkshire pigs underwent a 3 h left lung in vivo lung perfusion (IVLP) with various doses of oxaliplatin (7.5, 10, 20, 40, and 80 mg/L), which were administered to the perfusion circuit reservoir as a bolus. Biocompatible solid-phase microextraction (SPME) microprobes were combined with global metabolite profiling to obtain spatiotemporal information about the activity of the drug, determine toxic doses that exceed therapeutic efficacy, and conduct a mechanistic exploration of associated lung injury. Mild and subclinical lung injury was observed at 40 mg/L of oxaliplatin, and significant compromise of the hemodynamic lung function was found at 80 mg/L. This result was associated with massive alterations in metabolic patterns of lung tissue and perfusate, resulting in a total of 139 discriminant compounds. Uncontrolled inflammatory response, abnormalities in energy metabolism, and mitochondrial dysfunction next to accelerated kynurenine and aldosterone production were recognized as distinct features of dysregulated metabolipidome. Spatial pharmacometabolomics may be a promising tool for identifying pathological responses to chemotherapy.

Boundary Layer Development on a Concave Surface： Flow Visualization and Hot Wire Velocity Measurements

Gortler vortices are key issues in the design of gas turbine blades. The present study deals with flow visualization over concave surface for gas turbine applications. The aim is to comprehend qualitatively the flow trends, particularly the Gortler vortices formation and development. Gortler vortices have the shape of mushroom-like vortices regularly spaced at 25 mm. These vortices grow and increase in strength more rapidly along the surface in the case of the same grid of turbulence applied to the measuring section. The curvature radius of the studied blade is 0.5 m and the stream turbulence intensity level is 2.6%. The velocity field is measured by hot wire anemometer in the streamwise direction. The velocity profile is found to be highly distorted by the momentum transfer associated with Gortler vortices. The results are compared to Blasius flow and to literature data for a blade with curvature radius equal to 2 m.

Generation and Propagation of Tsunami by a Moving Realistic Curvilinear Slide Shape with Variable Velocities in Linearized Shallow-Water Wave Theory

The process of tsunami evolution during its generation under the effect of the variable velocities of realistic submarine landslides based on a two-dimensional curvilinear slide model is investigated. Tsunami generation from submarine gravity mass flows is described in three stages. The first stage represented by a rapid curvilinear down and uplift faulting with rise time. The second stage represented by a unilaterally propagation in the positive x direction to a significant length to produce curvilinear two-dimensional models represented by a depression slump, and a displaced accumulation slide model. The last stage represented by the time variation in the velocity of the accumulation slide (block slide). By using transforms method, Laplace in time and Fourier in space, tsunami waveforms within the frame of the linearized shallow water theory for constant water depth are analyzed analytically for the movable source model. Effect of the water depths on the amplification factor of the tsunami generation by the submarine slump and slide for different propagation lengths and widths has been studied and the results are plotted. Comparison of tsunami peak amplitudes is discussed for different propagation lengths, widths and water depths. In addition, we demonstrated the tsunami propagation waveforms after the slide stops moving at different propagation times.

Development of a novel in vivo lung perfusion(IVLP)procedure allows localized delivery of high-dose doxorubicin(DOX)for targeting residual micrometastatic disease in the ***,DOX delivery via IVLP requires careful monitoring of drug level to ensure tissue concentrations of this agent remain in the therapeutic window.A small dimension nitinol wire coated with a sorbent of biocompatible morphology(Bio-SPME)has been clinically evaluated for in vivo lung tissue extraction and determination of DOX and its key *** in vivo Bio-SPME-IVLP experiments were performed on pig model over various(150 and 225 mg/m^(2))drug doses,and during human clinical *** patients with metastatic osteosarcoma were treated with a single 5 and 7 μg/mL(respectively)dose of DOX during a 3-h *** both pig and human cases,DOX tissue levels presented similar trends during *** lung tissue concentrations of drug ranged between 15 and 293 μg/g over the course of the IVLP *** addition to DOX levels,Bio-SPME followed by liquid chromatography-mass spectrometry analysis generated 64 metabolic features during endogenous metabolite screening,providing information about lung status during drug ***-time monitoring of DOX levels in the lungs can be performed effectively throughout the IVLP procedure by in vivo Bio-SPME chemical biopsy ***-SPME also extracted various endogenous molecules,thus providing a real-time snapshot of the physiology of the cells,which might assist in the tailoring of personalized treatment strategy.

Numerical Solution of the Rotating Shallow Water Flows with Topography Using the Fractional Steps Method

The two-dimensional nonlinear shallow water equations in the presence of Coriolis force and bottom topography are solved numerically using the fractional steps method. The fractional steps method consists of splitting the multi-dimensional matrix inversion problem into an equivalent one dimensional problem which is successively integrated in every direction along the characteristics using the Riemann invariant associated with the cubic spline interpolation. The height and the velocity field of the shallow water equations over irregular bottom are discretized on a fixed Eulerian grid and time-stepped using the fractional steps method. Effects of the Coriolis force and the bottom topography for particular initial flows on the velocity components and the free surface elevation have been studied and the results are plotted.

Modeling of Tsunami Generation and Propagation by a Spreading Curvilinear Seismic Faulting in Linearized Shallow-Water Wave Theory

The processes of tsunami evolution during its generation in search for possible amplification mechanisms resulting from unilateral spreading of the sea floor uplift is investigated. We study the nature of the tsunami build up and propagation during and after realistic curvilinear source models represented by a slowly uplift faulting and a spreading slip-fault model. The models are used to study the tsunami amplitude amplification as a function of the spreading velocity and rise time. Tsunami waveforms within the frame of the linearized shallow water theory for constant water depth are analyzed analytically by transform methods (Laplace in time and Fourier in space) for the movable source models. We analyzed the normalized peak amplitude as a function of the propagated uplift length, width and the average depth of the ocean along the propagation path.

Effect of cold deformation and heat treatment on microstructure and mechanical properties of TC21 Ti alloy

In the present research work on TC21 titanium alloy(6.5 Al-3 Mo-1.9 Nb-2.2 Sn-2.2 Zr-1.5 Cr), the effects of cold deformation, solution treatment with different cooling rates and then aging on microstructure, hardness and wear property were investigated. A cold deformation at room temperature with 15% reduction in height was applied on annealed samples. The samples were solution-treated at 920 ℃ for 15 min followed by different cooling rates of water quenching(WQ), air cooling(AC) and furnace cooling(FC) to room temperature. Finally, the samples were aged at 590 ℃ for 4 h. Secondary α-platelets precipitated in residual β-phase in the case of solution-treated samples with AC condition and aged ones. The maximum hardness of HV 470 was obtained for WQ + aging condition due to the presence of high amount of residual β-matrix(69%), while the minimum hardness of HV 328 was reported for FC condition. Aging process after solution treatment can considerably enhance the wear property and this enhancement can reach up to about 122% by applying aging after WQ compared with the annealed samples.

URBAN PLANNING:A TOOL FOR URBAN POVERTY ALLEVIATION IN SUDAN

It is expected that over 50% of the world population will live in cities in 2005. The growing trend indicates that as many as one fifth of all households still remain with housing tenure problems which they try to solve through very expensive rentals insecurity socially and environmentally-hazardous squatting or unhealthy overcrowding sometimes ending up with none at all. And hence propcr planning is necessary in urban development policies to improve human settlement managements in a sustainable way. This article has tried to handle urban poverty in the Sudan to a reasonable level of detail. It dealt with selected areas with regard to promotion of sustainable human settlement. The study managed to identity some recent progress in Sudanese human settlements that is however still accompanied by numerous serious instances of decline in the urban environment especially that of the greater Khartoum. Such instances of environmental deterioration are profiled and analyzed by the study with respect to all human settlement aspects of management land-use infrastructure energy and transport. The paper's conclusion emphasized that improving all these aspects is of high priority.
