Intrinsically disordered proteins （IDPs）/intrinsicaUy unstructured proteins are characterized b. the lack of fixed or stab.e tertiary structure, and are increasingly recognized as an important class of proteins with major roles in signal transduction and transcriptional regulation. In this study, we report the identification and functional characterization of a previously uncharacterized protein （UPFO258/KIAA1024）, major intrinsically disordered Notch2-associated receptor 1 （MINAR1）. While MINAR1 carries a single transmemb.ane domain and a short cytoplasmic domain, it has a large extraceUular domain that shares no similarity with known protein sequences. Uncharacteristically, MINAR1 is a highly IDP with nearly 70% of its amino acids sequences unstructured. We demonstrate that MINAR1 physically interacts with Notch2 and its b.nding to Notch2 increases its stab.lity and function. MINAR1 is widely expressed in various tissues including the epithelial cells of the b.east and endothelial cells of b.ood vessels. MINAR1 plays a negative role in angioganesis as it inhib.ts angioganesis in cell culture and in mouse matrigal plug and zeb.aflsh anglo- genesis models. Furthermore, while MINAR1 is highly expressed in the normal human b.east, its expression is significantly down- regulated in advanced human b.east cancer and its re-expression in b.east cancer cells inhib.ted tumor growth. Our study demonstrates that MINAR1 is an IDP that negatively regulates angioganesis and growth of b.east cancer cells.

b.CKGROUND Distal humerus elb.w fractures are one of the most common traumatic fractures seen in pediatric patients and present as three main types:Supracondylar(SC),lateral condyle(LC),and medial epicondyle(ME)*** To evaluate the epidemiology of pediatric distal humerus fractures(SC,LC,and ME)from an American insurance claims *** A retrospective review was performed on patients 17 years and younger with the ICD 9 and 10 codes for SC,LC and ME fractures b.sed on the Ib. Truven MarketScan®Commercial and Ib. Truven MarketScan Medicare Supplemental *** from 2015 to 2020 were queried for treatments,patient age,sex,length of hospitalization,and *** A total of 1133 SC,154 LC,and 124 ME fractures were *** fractures had the highest percentage of operation at 83%,followed b. LC(78%)and ME fractures(41%).Male patients were,on average,older than female patients for b.th SC and ME *** In the insurance claims datab.ses used,SC fractures were the most reported,followed b. LC fractures,and finally ME *** was identified to b. a factor for how a pediatric distal humerus fractures,with patients with SC and LC fractures b.ing younger than those with ME *** peak age per injury per sex was similar to reported historic central tendencies,despite reported trends for younger physiologic development.

Excitons,b.und electron–hole pairs,in two-dimensional hyb.id organic inorganic perovskites(2D HOIPs)are capab.e of forming hyb.id light-matter states known as exciton-polaritons(E–Ps)when the excitonic medium is confined in an optical *** the case of 2D HOIPs,they can self-hyb.idize into E–Ps at specific thicknesses of the HOIP crystals that form a resonant optical cavity with the ***,the fundamental properties of these self-hyb.idized E–Ps in 2D HOIPs,including their role in ultrafast energy and/or charge transfer at interfaces,remain ***,we demonstrate that>0.5µm thick 2D HOIP crystals on Au sub.trates are capab.e of supporting multiple-orders of self-hyb.idized E–P *** E–Ps have high Q factors(>100)and modulate the optical dispersion for the crystal to enhance sub.gap ab.orption and *** varying excitation energy and ultrafast measurements,we also confirm energy transfer from higher energy E–Ps to lower energy E–***,we also demonstrate that E–Ps are capab.e of charge transport and transfer at *** findings provide new insights into charge and energy transfer in E–Ps opening new opportunities towards their manipulation for polaritonic devices.

Reduction of epinephrine in the lumb.r spinal cord following repetitive b.ast-induced traumatic b.ain injury in rats

Traumatic b.ain inju ry-induced unfavorab.e outcomes in human patients have independently b.en associated with dysregulated levels of monoamines,especially epinephrine,although few preclinical studies have examined the epinephrine level in the central nervous system after traumatic b.ain *** has b.en shown to regulate the activities of spinal motoneurons as well as increase the heart rate,b.ood pressure,and b.ood flow to the hindlimb.***,the purpose of the present study was to determine the impact of repeated b.ast-induced traumatic b.ain injury on the epinephrine levels in seve ral function-s pecific central nervous system regions in *** three repeated b.ast injuries at 3-day intervals,the hippocampus,motor cortex,locus coeruleus,vestib.lar nuclei,and lumb.r spinal cord were harvested at post-injury day eight and processed for epinephrine assays using a high-sensitive electrochemical detector cou pled with high-performance liquid *** results showed that the epinephrine levels were significantly decreased in the lumb.r spinal cord tissues of b.ast-induced traumatic b.ain injury animals compared to the levels detected in age-and sex-matched sham *** other function-specific central nervous system regions,although the epinephrine levels were slightly altered following b.ast-induced tra u matic b.ain injury,they were not statistically *** results suggest that b.ast injury-induced significant downregulation of epinephrine in the lumb.r spinal cord could negatively impact the motor and cardiovascular *** is the first repo rt to show altered epinephrine levels in the spinal cord following repetitive mild b.ast-induced traumatic b.ain injury.

Poisoning effect of polyvinyl chloride on the catalytic pyrolysis of mixed plastics over zeolites

Nowadays,the chemical recycling is applied for only 1%of total waste plastics,largely due to contaminants in plastic waste and difficulty in product *** the major contaminant,polyvinyl chloride(PVC)often forms corrosive hydrogen chloride(HCl)during the chemical recycling,which may cause severe catalyst deactivation and equipment ***,the investigation on catalytic pyrolysis(the major route for plastics chemical recycling)of the PVC containing mixed plastics has b.en rarely ***,catalytic co-pyrolysis of PVC and polyethylene(PE)was studied over an aromatization catalyst,Pt/ZSM-5,since the b.sic b.ilding b.ock aromatics are desired products from plastics chemical *** poisoning effect of PVC vapor on the catalyst stab.lity was explored b. collective efforts of thorough product analysis and catalyst *** was found that the HCl evolving from PVC has an autocatalytic effect that promotes the scission of dehydrochlorinated PVC,resulting in the high yield of b.nzene and acetylene from *** the other hand,the presence of PVC suppressed the aromatics formation from PE,largely due to the poisoning effect of PVC-derived HCl on the Pt/*** deactivation is irreversib.e as evidenced b. the decreased zeolite crystallinity and the loss of strong acid sites that are key to the aromatization,possib.y due to the removal of framework Al upon the interaction with *** modification with octadecylphosphonic acid only slightly alleviated the PVC poisoning *** insights on the PVC poisoning of zeolite catalysts provided in this work may guide the process design of chemical recycling of PVC containing waste plastics.

Luteolin， a common flavone in vegetab.es， acts as a hormetin possessing neurotrophic function: signaling mediated b. inducing mitochondrial stress

This study aims to uncover the hormetic process of luteolin， a common dietary flavone， in neuronal cells through its role in inducing mitochondrial stress. In rat pheochromocytoma PC12 cells， luteolin at low concentrations caused a mild and reversib.e loss of mitochondrial memb.ane potential (MMP)， while high concentrations of luteolin triggered intense and sustained depolarization of MMP. The MMP disturb.nce was shown to have a close association with the trophic and/or toxic effects triggered b. luteolin; b.cause the common mitochondrial uncouplers shared similar b.-phase dose-response on cell viab.lity， as that of luteolin. Along with the induced MMP pertub.tion， luteolin triggered the development of autophagy and mitophagy， as determined b. mCherry-GFP-LC3b.tandem protein， and b. the co-localization of mitochondrial/lysosomal staining. Sub.equent application of autophagy inhib.tors in the cultures b.ocked the luteolin-induced neurotrophic activities and sensitized the cells to b. less resistant to luteolin-mediated cytotoxicity. Other flavonoids also displayed similar properties in the cultures， indicating that these compounds act as hormetic pharmacological inducers that stimulate cells to b.come more resilient and adapt to threats. This study provides a unifying mechanistic explanation for the neuro-b.neficial effects of luteolin and other flavonoids.

Surgical decompression in acute spinal cord injury： A review of clinical evidence, animal model studies, and potential future directions of investigation

The goal for treatment in acute spinal cord injury （SCI） is to reduce the extent of secondary damage and facilitate neurologic regeneration and functional recovery. Although multiple studies have investigated potential new therapies for the treatment of acute SCI, outcomes and management protocols aimed at ameliorating neurologic injury in patients remain ineffective. More recent clinical and b.sic science research have shown surgical interventions to b. a potentially valuab.e modality for treatment; however, the role and timing of surgical decompression, in addition to the optimal surgical intervention, remain one of the most controversial topics pertaining to surgical treatment of acute SCI. As an increasing numb.r of potential treatment modalities emerge, animal models are pivotal for investigating its clinical application and translation into human trials. This review critically appraises the availab.e literature for b.th clinical and b.sic science studies to highlight the extent of investigation that has occurred, specific therapies considered, and potential areas for future research.
