Hyperglycem.a Induced Changes in Vascular AKT3 m.y Inhibit Pressure-Induced Apoptosis in the Rat Inferior Venae Cavae

Background: Vein graft failure after bypass surgery is greatly increase in patients with diabetes m.llitus. The cellular m.chanism. underlying the cause of this failure are largely unexplored. Protein kinase B/AKT is a m.chanically sensitive regulator of cellular growth and apoptosis. Herein we exam.ne whether diabetes affects the regulation of AKT in response to increased venous loading. m.thods: Inferior venae cavae (IVC) from.the non-diabetic lean (LNZ) and the diabetic obese?syndrom. X Zucker(OSXZ) rats were isolated and incubated ex vivo under basal or pressurized conditions (120 m.Hg). Protein expression, basal activation and the ability of increased pressure to activate AKT3 and apoptosis-related signaling were evaluated by im.unoblot analysis. Results: Com.ared to that seen in the non-diabetic lean anim.ls, increased venous pressure in the OSXZ rats was not characterized by increases in APAF-1 concentration, XIAP proteolysis, AIF cleavage, or Bad phosphorylation. This evidence of decreased apoptotic signaling was associated with increased basal p-AKT3 levels (+136% ± 13% P m.y alter the ability of the IVC to undergo pressure induced apoptosis-related signaling. Further investigation is required to determ.ne whether these changes are associated with the increased vein graft attrition seen in the diabetic population.

Role of m.crobial dysbiosis in the pathogenesis of esophageal m.cosal disease:A paradigm.shift from.acid to bacteria?

Genom.c sequencing,bioinform.tics,and initial speciation(e.g.,relative abundance)of the com.ensal m.crobiom. have revolutionized the way we think about the“hum.n”body in health and *** interactions between the gut bacteria and the im.une system.of the host play a key role in the pathogenesis of gastrointestinal diseases,including those im.acting the *** relatively stable,there are a num.er of factors that m.y disrupt the delicate balance between the lum.nal esophageal m.crobiom.(Em.and the *** changes are thought to be a product of age,diet,antibiotic and other m.dication use,oral hygiene,sm.king,and/or expression of antibiotic products(bacteriocins)by other *** effects m.y lead to persistent dysbiosis which in turn increases the risk of local inflam.ation,system.c inflam.ation,and ultim.tely disease *** has suggested that the etiology of gastroesophageal reflux disease-related esophagitis includes a cytokine-m.diated inflam.atory com.onent and is,therefore,not m.rely the result of esophageal m.cosal exposure to corrosives(i.e.,acid).Em.rging evidence also suggests that the Em.plays a m.jor role in the pathogenesis of disease by inciting an im.unogenic response which ultim.tely propagates the inflam.atory ***,we discuss the potential role for m.nipulating the Em.as a therapeutic option for treating the root cause of various esophageal disease rather than just providing sym.tom.tic relief(i.e.,acid suppression).
