Animal models are necessary to investigate the pathogenic features underlying motor neuron degeneration and for therapeutic development in amyotrophic lateral sclerosis（ALS）. Measures of model validity allow for a critical interpretation of results from each model and caution from over-interpretation of experimental models. Face and construct validity refer to the similarity in phenotype and the proposed causal factor to the human disease, respectively. More recently developed models are restricted by limited phenotype characterization, yet new models hold promise for novel disease insights, thus highlighting their importance. In this article, we evaluate the features of face and construct validity of our new zebrafish model of environmentally-induced motor neuron degeneration and discuss this in the context of current environmental and genetic ALS models, including C9 orf72, mutant Cu/Zn superoxide dismutase 1 and TAR DNA-binding protein 43 mouse and zebrafish models. In this mini-review, we discuss the pros and cons to validity criteria in each model. Our zebrafish model of environmentally-induced motor neuron degeneration displays convincing features of face validity with many hallmarks of ALS-like features, and weakness in construct validity. However, the value of this model may lie in its potential to be more representative of the pathogenic features underlying sporadic ALS cases, where environmental factors may be more likely to be involved in disease etiology than single dominant gene mutations. It may be necessary to compare findings between different strains and species modeling specific genes or environmental factors to confirm findings from ALS animal models and tease out arbitrary strain-and overexpression-specific effects.

Objective:Treatment delays in prostate cancer have been characterised,although not explicitly in men undergoing transperineal prostate *** aimed to determine if delays to radical prostatectomy correlate with adverse outcomes using a contemporary population-based cohort of men diagnosed by transperineal ***:This study analysed men with prostate cancer of the International Society for Urological Pathology grade group≥2,diagnosed by transperineal prostate biopsies who underwent prostatectomy,using the prospectively data from 1 January 2014 to 30 June 2018 Prostate Cancer Outcomes *** were analysed according to stratified demographic and disease *** intervals from biopsy(28,60,90,120,and 270 days)were compared using odds ratios and regression analyses for proportion of upgrading,early biochemical recurrence,pT3 disease at prostatectomy,and positive surgical ***:In total,2008 men were *** were 306(16.7%)men with upgrading,151(8.4%)with biochemical recurrence,1068(54.1%)with pT3 disease,and 464(23.1%)with positive surgical margins(percentages excluded patients with missing data).All adverse outcomes studied were significantly associated with higher prostate-specific antigen and grade at *** of 120-270 days did not adversely alter the incidence of Gleason upgrading,pT3,or ***(most frequent 60-89 days,28%)were associated with positive surgical margins but not *** modelling demonstrated no increased likelihood of most adverse outcomes for up to 270 ***:Men with prostate cancer of grade group≥2 diagnosed through transperineal biopsy may wait up to 270 days for a prostatectomy without a greater likelihood of upgrading,pT3 disease,positive surgical margins,or biochemical recurrence.

The availability of high‐strength fabrics and progress in the development of large‐scale inflatable technology made possible the creation of temporary and quickly deployable structures for protection of underground *** structures are relatively lightweight and portable,and can maintain the required rigidity while in *** benefits have prompted the development of inflatable structures for use in confined spaces,such as tunnels and large‐diameter pipes to act as barriers for containing flooding with minimal infrastructure *** work presents experimental results obtained from the evaluation of frictional characteristics of the fabric material that constitute the structural membrane of confined inflatable structures developed for protection of underground transportation tunnels and other large *** tests at coupon level and slippage tests in a reduced‐scale inflatable structure were performed in order to evaluate the frictional characteristics of Vectran *** at coupon level were performed to determine the friction coefficient for different surface types and *** with the reduced‐scale inflatable structure contributed to the understanding of the friction characteristics at system level when subjected to different pressurization or depressurization sequences designed to induce *** results indicate that friction coefficient values at coupon level are about 29 percent higher than values derived from reduced‐scale tests.

For the two-dimensional Navier-Stokes equations of isentropic magnetohydrodynamics （MHD） with γ-law gas equation of state, γ≥ 1, and infinite electrical resistivity, we carry out a global analysis categorizing all possible viscous shock profiles. Precisely, we show that the phase portrait of the traveling-wave ODE generically consists of either two rest points connected by a viscous Lax profile, or else four rest points, two saddles and two nodes. In the latter configuration, which rest points are connected by profiles depends on the ratio of viscosities, and can involve Lax, overcompressive, or undercompressive shock profiles. Considered as three-dimensional solutions, undercompressive shocks axe Lax-type （Alfven） waves. For the monatomic and diatomic cases γ= 5/3 and γ=7/5, with standard viscosity ratio for a nonmagnetic gas, we find numerically that the the nodes are connected by a family of overcompressive profiles bounded by Lax profiles connecting saddles to nodes, with no undercompressive shocks occurring. We carry out a systematic numerical evans function analysis indicating that all of these two-dimensional shock pro- files are linearly and nonlinearly stable, both with respect to two- and three-dimensional perturbations. For the same gas constants, but different viscosity ratios, we investigate also cases for which undercompressive shocks appear; these are seen numerically to be stable as well, both with respect to two-dimensional and （in the neutral sense of convergence to nearby Riemann solutions） three-dimensional perturbations.
