Background: Animal models of diet-induced obesity(DIO) are commonly used in medical research for mimicking human diseases. There is no universal animal model, and careful evaluation of variety of factors needs to be considered when designing new experiments. Here, we investigated the effect of 9 weeks high-fat diet(HFD) intervention, providing 60% energy from fat, on parameters of inflammation and insulin resistance in male c57 BL/6 j ***: Six weeks old mice were initiated on regular diet(RD) or HFD providing 60 kcal energy from fat for 9 weeks. Fasting blood glucose levels were measured by glucometer, and fasting plasma levels of insulin and proinflammatory cytokines by Luminex assay. Insulin sensitivity was evaluated by using QUIcKI and HOMA2 ***: HFD mice showed ~ 40% higher body weight and ~ 20% larger abdominal circumference, due to an increase in the white adipose tissue mass. Liver examination revealed increased size and higher hepatic lipid accumulation in livers from HFD mice compared to their RD counterparts. Animals from the HFD group were characterized with significantly higher presence of crown-like structures(cLS) in WAT and higher plasma levels of proinflammatory cytokines(TNF-α, IL-6, leptin, McP-1, PAI-1, and resistin). HFD-fed mice also demonstrated impaired insulin sensitivity(lower QUIcKI, higher HOMA-insulin resistance(HOMA-IR), and lower HOMA-percent sensitivity(HOMA-%S)) index ***: Male c57 BL/6 j mice on 9 weeks HFD providing 60 kcal energy from fat display impaired insulin sensitivity and chronic inflammation, thus making this DIO mouse model appropriate for studies of early stages of obesity-related pathology.

Pancreatic ductal adenocarcinoma is the 4th leading cause of cancer deaths in the United States. The majority of patients are candidates only for palliative chemotherapy, which has proven largely ineffective in halting tumor progression. One proposed mechanism of chemoresistance involves signaling via the mesenchymal-epithelial transition factor protein (MET), a previously established pathway critical to cell proliferation and migration. Here, we review the literature to characterize the role of MET in the development of tumorigenesis, metastasis and chemoresistance, highlighting the potential of MET as a therapeutic target in pancreatic cancer. In this review, we characterize the role of c-Met in the development of tumorigenesis, metastasis and chemoresistance, highlighting the potential of c-Met as a therapeutic target in pancreatic cancer.

Bimatoprost/timolol fixed combination(BTFc) in patients with primary open angle glaucoma or ocular hypertension in Greece

AIM：To evaluate the efficacy and tolerability of the fixed combination of bimatoprost 0.03%and timolol 0.5%（BTFc）in patients in Greece with primary open angle glaucoma（POAG）or ocular hypertension（OHT）whose previous therapy provided insufficient lowering of intraocular pressure（IOP）.·METHODS：A multicenter,prospective,open-label,non-interventional,observational study of the use of BTFc in clinical practice was conducted at 41 sites in *** primary endpoint was the reduction in IOP from baseline at study end,approximately 12wk after initiation of BTFc therapy.·RESULTS：A total of 785 eligible patients were enrolled in the study and 97.6%completed the *** mean±SD IOP reduction from baseline at 12wk after initiation of BTFc was 6.3±2.8 mm Hg（=764;〈0.001）.In patients（=680）who replaced their previous IOP-lowering monotherapy（a single drug,or a fixed combination of 2drugs in a single ophthalmic drop）with once-daily BTFc,the mean±SD IOP reduction from baseline at 12wk was 6.2±2.8 mm Hg（〈0.001）.IOP was reduced from baseline in 99.2%of patients,and 58.0%of patients reached or exceeded their target *** mean IOP reductions were observed regardless of the previous *** was well tolerated,with 96.0%of patients who completed the study rating the tolerability of BTFc as＂good＂or＂very good.＂Adverse events were reported in 8.3%of patients;only 0.6%of patients discontinued the study due to adverse events.·c ONcLUSION：In clinical practice in Greece,BTFc is well tolerated and effectively lower the IOP in patients with POAG or OHT who requires additional IOP lowering on their previous therapy.

Implications of oral biofilms in medically at risk persons

There is the need to understand the composition of oral biofilms so that appropriate preventive and treatment regimens, including using appropriate antimicrobials, can be developed further. Additionally, when the systemic effects from specific microorganisms in oral biofilms are better understood, more targeted preventive treatment options may be recommended for persons at high risk for potential systemic diseases such as cardiovascular dis- ease, and for aspiration pneumonia. Hence, the possible association between periodontopathic microorganisms, and also between cariogenic microorganisms in high caries risk persons, and systemic diseases requires further re- search involving metagenomic and large well-designed clinical studies. Effective preventive oral care is important for reducing potential systemic diseases.

Real-world emission factors for caterpillar 797B heavy haulers during mining operations

Real-world fuel-based emission factors （EFs） from caterpillar 797B diesel heavy haulers when used for oil sands mining operations were measured using an on-hoard portable emissions monitoring system. Average EFs （in g/kg fuel） for 16 separate tests on four trucks were 3150 ± 4 for cO2, 2.51 ± 1.30 for cH4, 10.28 ± 3.21 for cO, 0.61 ± 0.32 for non-methane hydrocarbons, 55.49 ± 9,75 for NOx （reported as NO2）, 0.70 ± 0.17 for PM2.s （mass of particles with aerodynamic diameter 〈2.5 μm）, and 0.34 ± 0.05 for black carbon （Bc）. Ultrafine particle numbers averaged （4.7 ± 4.1 ） × 10^15 particleslkg fuel. Fuel-based EFs of NO, NO2, and particle numbers were higher during idling, whereas PM2.5 and Bc EFs were higher during traveling without a load. A comparison of emissions with those reported by oil sands facilities showed large variations, both among the facilities and between the reported emissions and the real-world measurements.

Diagnostics are presented from an ensemble of high-resolution forecasts that differed markedly in their predictions of the rapid intensification(RI)of Typhoon *** show that the basic difference stems from subtle differences in initializations of(a)500-850-h Pa environmental winds,and(b)midlevel moisture and *** then describe how these differences impact on the evolving convective organization,storm structure,and the timing of *** expected,ascent,diabatic heating and the secondary circulation near the inner-core are much stronger in the member that best forecasts the *** evolution of vortex cloudiness from this member is similar to the actual imagery,with the development of an inner cloud band wrapping inwards to form the *** present evidence that this structure,and hence the enhanced diabatic heating,is related to the tilt and associated dynamics of the developing inner-core in *** the most accurate ensemble member:(a)inhibition of ascent and a reduction in convection over the up-shear sector allow moistening of the boundary-layer air,which is transported to the down-shear sector to feed a developing convective asymmetry;(b)with minimal ventilation,undiluted clouds and moisture from the down-shear left quadrant are then wrapped inwards to the up-shear left quadrant to form the eyewall cloud;and(c)this process seems related to a critical down-shear tilt of the vortex from midlevels,and the vertical phase-locking of the circulation over up-shear *** the member that forecasts a much-delayed RI,these processes are inhibited by stronger vertical wind shear,initially resulting in poor vertical coherence of the circulation,lesser moisture and larger *** analysis suggests that ensemble prediction is needed to account for the sensitivity of forecasts to a relatively narrow range of environmental wind shear,moisture and vortex inner-structure.

Pediatric autoimmune neuropsychiatric disorders associated with or without streptococcal and other bacterial infections (PANDAS/cANS) are emerging as a featured pediatric disorder. Although there is some controversy regarding treatment approaches, especially related to the behavioral sequelae, we have hypothesized in other published work that it is characterized by the rapid onset of Reward Deficiency Syndrome (RDS) in children. We propose utilizing a multi-systems biological approach involving the coupling of genetic addiction risk testing and pro-dopamine regulation (KB220/POLYGEN®) to help induce “dopamine homeostasis” in patients with PANDAS, especially those with known DNA-induced hypodopaminergia. This case study examines a 12-year-old caucasian male with no prior psychiatric issues who presented with a sudden onset of severe anxiety, depression, emotional liability, and suicidal ideation. The patient underwent genotyping and the genetic addiction risk score (GARS) testing, which revealed risk polymorphisms in the dopamine D2 (-DRD2/ANKK (Taq1A), OPRM1 (A/G), DRD3 (c/T), and MAOA (4R) genes. These polymorphisms have been linked to hypodopaminergia. The patient was subsequently placed on research ID-KB220ZPBMPOLY (POLYGEN®), and albeit the possibility of bias, based upon self and parental assessment, a marked rapid improvement in psychiatric symptoms was observed. In the second phase of treatment (102 days utilizing KB220), the patient received standard antibody testing, which was positive for Lyme. Antibacterial therapy started immediately, and KB220z was discontinued to provide a wash-out period. A monotonic trend analysis was performed on each outcome measure, and a consistently decreasing trend was observed utilizing antibacterial therapy. Our recommendation, albeit only one case, is to utilize and further research a combined therapeutic approach, involving precision-guided DNA testing and pro-dopamine regulation along with antibacterial therapy, as well as

Aged populations have remarkable variability in recent memory and cognitive mapping. Although some individuals may have substantial age-related impairments, others perform almost as well as young individuals. This paper reviews the relevant data on aged rats and indicates two challenges for biomarkers of aging. The first is to provide an appropriate quantitative description of these individual differences. The second is to use them effectively as markers for age-related changes in psychological functions and their neural substrates.
