Loss of GATA6-mediated up-regulation of UTX promotes pancreatic tumorigenesis and progression

Ubiquitously transcribed tetratricopeptide repeat on chromosome X(UTX),also known as lysine(K)-specific demethylase 6A(KDM6A),functions as a tumor suppressor gene or oncogene depending on the tumor type and ***,its tumor-suppressive mechanisms remain largely ***,we investigated the clinical significance and biological effects of UTX expression in pancreatic ductal adenocarcinoma(PDA)and determined the potential mechanisms of its *** expression and its association with clinicopathologic characteristics of PDA patients were analyzed using *** mRNA and protein expression and their regulation in PDA cell lines were measured using quantitative polymerase chain reaction and Western blot *** biological functions of UTX in PDA cell growth,migration,and invasion were determined using gain-and loss-of-function assays with both in vitro and in vivo animal *** expression was reduced in human PDA cell lines and *** UTX expression was associated with poor differentiation and prognosis in *** UTX expression inhibited PDA proliferation,migration,and invasion in vitro and PDA growth and metastasis in vivo,whereas knockdown of UTX expression did the ***,UTX expression was trans-activated by GATA6 ***6-mediated PDA progression could be blocked,at least partially,by silencing UTX *** conclusion,loss of GATA6-mediated UTX expression was evident in human PDA and restored UTX expression suppressed PDA growth and ***,UTX is a tumor suppressor in PDA and may serve as a prognostic biomarker and therapeutic target.

Down-regulation of microRNA-23a promotes pancreatic ductal adenocarcinoma initiation and progression by up-regulation of FOXM1 expression

Transcriptional factor Forkhead box M1(FOXM1)plays an important role in pancreatic ductal adenocarcinoma(PDAC)development and *** molecular mechanisms underlying its dysregulation remain *** identified and functionally validated the microRNAs(miRNAs)that critically regulate FOXM1 expression in *** expression levels of miRNA-23a(miR-23a-3p and-5p)were altered in PDAC cell lines and their effects on FOXM1 signaling and cell proliferation and migration and tumorigenesis were examined in vitro and in vivo using mouse PDAC *** with non-tumor pancreatic tissues,PDAC tissues and cell lines exhibited significantly reduced levels of miR-23a *** miR-23a expression and concomitant increase in FOXM1 expression were also observed in acinar-toductal metaplasia and pancreatic intraepithelial neoplasia,the major premalignant lesions of *** expression of miR-23a reduced the expression of FOXM1 and suppressed cell proliferation and migration in PDAC cells,whereas the inhibitors of miR-23a did the *** or reduced levels of miR-23a increased the levels of FOXM1 expression,while increased expression of FOXM1 down-regulated miR-23a expression,suggesting that miR-23a and FOXM1 were mutual negative regulators of their expression in PDAC ***,the miR-23a/FOXM1 signaling axis is important in PDAC initiation and progression and could serve as an interventional or therapeutic target for patients with early or late stages of PDAC.

Understanding PDF uncertainty in W boson mass measurements

We study the dependence of the transverse mass distribution of charged leptons and the missing energy on parton distributions(PDFs)adapted to W boson mass measurements at the CDF and ATLAS *** compare the shape variations of the distribution induced by different PDFs and find that the spread of predictions from different PDF sets can be significantly larger than the PDF uncertainty predicted by a specific PDF *** suggest analyzing the experimental data using up-to-date PDFs to gain a better understanding of the PDF uncertainties in W boson mass *** also perform a series of Lagrange multiplier scans to identify the constraints on the transverse mass distribution imposed by individual data sets in the CT18 global *** the case of the CDF measurement,the distribution is mostly sensitive to d-quark PDFs in the intermediate x region,which are largely constrained by DIS and Drell-Yan data on deuteron targets and Tevatron lepton charge asymmetry data.

Collective rotations of fission isomers in actinide nuclei

Using the total-Routhian-surface(TRS)method,the rotational behaviors of fission isomers in the second well of actinide nuclei234 242U,236 244Pu and238 246Cm were *** pairing-deformation-frequency self-consistent TRS calculations reproduced reasonably the experimental moments of inertia extracted from spectroscopic *** is calculated that,in these largelyelongated(β2≈0.65 andβ4≈0.03)fission isomers,theν12[981]neutron andπ12+[651]proton align simultaneously at rotational frequencyω≈0.4 0.6 MeV(corresponding to spin I≈80),which leads to clear upbending in moments of inertia(MoI’s).Our calculations have indicated that the hexadecapole deformationβ4influenced significantly the frequency of the rotational alignment of the proton12+[651]orbit.

Unraveling the evolution of oxygen vacancies in TiO/Cu and its role in COhydrogenation

The remarkable contribution of oxygen vacancies has been revealed by the operando spectroscopies for methanol synthesis from CO2hydrogeneration on the reducible metal oxide-supported copper catalysts. However, a challenge remains in the intrinsic understanding of the evolution and advantage of oxygen vacancies for methanol synthesis. Here we prepare the TiO2–x/Cu with different oxygen vacancy densities by adjusting the ball-milling frequency. At the optimal condition, a TiO2–x/Cu with more oxygen vacancies delivers an excellent methanol yield of 26.5 mmol g-1h-1at 300 °C with a selectivity of more than 70%. The combined analysis of experimental characterizations and theoretical calculations reveals that the mutual dispersions of TiO2–xand Cu driven by mechanical energy induce the electron rearrangement in the d orbital of the Ti atom and relax the Ti–O binding at the interface, which facilitates the formation of oxygen vacancies that further reduce the barrier of CO2hydrogenation to*HCOO due to higher nucleophilicity of titanium ions.
