Correlative light-electron microscopy(CLEm)requires the availability of robust probes which are visible both in light and electron *** we demonstrate a CLEm approach using small gold nanoparticles as a single *** gold nanoparticles bound to the epidermal growth factor protein were located with nanometric precision background-free in human cancer cells by light microscopy using resonant four-wave mixing(FWm),and were correlatively mapped with high accuracy to the corresponding transmission electron microscopy *** used nanoparticles of 10 nm and 5 nm radius,and show a correlation accuracy below 60 nm over an area larger than 10 pm size,without the need for additional fiducial *** accuracy was improved to below 40 nm by reducing systematic errors,while the localisation precision is below 10 ***-resolved FWm correlates with nanoparticle shapes,promising for multiplexing by shape recognition in future *** to the photostability of gold nanoparticles and the applicability of FWm microscopy to living cells,FWm-CLEm opens up a powerful alternative to fluorescence-based methods.

AIm:To examine factors influencing percutaneous endoscopic gastrostomy(PEG) uptake and outcomes in motor neuron disease(mND) in a tertiary care ***:Case notes from all patients with a confirmed diagnosis of mND who had attended the clinic at the Repatriation General Hospital between January 2007 and January 2011 and who had since died,were *** were extracted for demographics(age and gender),disease characteristics(date of onset,bulbar or peripheral predominance,complications),date and nature of discussion of gastrostomy insertion,nutritional status [weight measurements,body mass index(BmI)],date of gastrostomy insertion and subsequent progress(duration of survival) and quality of life(QoL) [Amyotrophic Lateral Sclerosis Functional Rating Scale-Revised(ALSFRS-R)].In addition,the type of clinician initiating the discussion regarding gastrostomy was recorded as Nutritional Support Team(involved in providing nutrition input viz Gastroenterologist,Speech Pathologist,Dietitian) and other(involved in non-nutritional aspects of patient care).Factors affecting placement and outcomes including length of survival,change in weight and QoL were ***:Case records were available for all 86 patients(49 men,mean age at diagnosis 66.4 years).Thirty-eight patients had bulbar symptoms and 48 had peripheral disease as their presenting ***-six patients reported ***-one patients had undergone gastrostomy *** major indications for PEG placement were dysphagia and weight *** patients required immediate full feeding,whereas 17 patients initially used the gastrostomy to supplement oral intake,4 for medication administration and 1 for *** the PEG regime met 73% ± 31% of the estimated total energy requirements,increasing to 87% ± 32% prior to *** was stabilization of weight in patients undergoing gastrostomy [BmI at 3 mo(22.6 ± 2.2 kg/m 2) and 6 mo(22.5 ± 2.0 kg/m 2) after PEG placement compared to

Pollution control structures may be adapted to enhance the removal of nutrients and pesticides from water. Charcoal-like material is known to sorb organic compounds in solution, including atrazine, a herbicide used to control broadleaf weeds, which persists in soils and can migrate from soils to water bodies, ultimately affecting water quality. The primary goal of this study was to investigate if activated charcoal cloths (single-weave, SW;double-weave, DW;and knitted, KT) are more efficient to remove atrazine from aqueous solutions than a cloth without activated charcoal (control) currently used in pollution control structures. The approach consisted of sorption kinetics, flow-through, and desorption/degradation experiments using 50 and 1000μg L-1 atrazine solutions. Results showed that within 30 min of contact time, the control sorbed from 22.6 to 36.1% of atrazine in solution;whereas the activated charcoal cloths sorbed from 76 to 99% of atrazine in solution (SW > KT > DW). The flow-through ex-periments showed that the SW sorbed 88.2, 76.1, and 52.2%of atrazine at the contact time of 0.75, 5, and 10 min, respectively. After 28 days of incubation, previously sorbed atrazine on the SW cloth did not degrade and mproving atrazine removal in water in pollution control structures.

Background Heme-oxygenase 1 （HO-1） is a rate-limiting enzyme in the degradation of heme to bilirubin, ferritin and carbon monoxide （CO） and may have significant anti-inflammatory function. The HO-1 gene promoter region shows microsatellite polymorphism with different （GT）n repeats, reported to differently induce gene expression, with the short allele associated with higher gene expression. We measured the acute inflammatory response using coronary artery bypass surgery （CABG） as a well-characterized and uniform stimulus and examined the correlation between levels of IL-6, C-reactive protein （CRP） and fibrinogen and their relationship to HO-1 genotype. methods Two hundred and seventy-five consecutive patients undergoing CABG were genotyped for the HO-1 promoter polymorphism using PCR and automated DNA capillary sequencer. IL-6, CRP and fibrinogen were measured at baseline and 6, 24, 48, 72, 96 and 120 hours after CABG. Results Complete IL-6, CRP and fibrinogen measures were available in 220 patients. Before surgery IL-6 levels showed a strong correlation with CRP and fibrinogen （ r = 0. 48, P 〈 0. 0001 ; r = 0. 41, P 〈 0. 0001 respectively）, with a significant correlation between CRP and fibrinogen （r=0. 61, P 〈 0. 0001 ）. All three acute phase reactants showed a significant increase after CABG. After surgery, peak IL-6 was strongly correlated with peak CRP （r=0. 34, P =0.0009） but not with peak fibrinogen （r=0. 15, P =0. 13 ）, while peak CRP and peak fibrinogen were significantly correlated （r = 0. 15, P 〈 0. 0001 ）. HO-1 allelic repeats ranged from 22- 42, with （GT）25 and （GT）32 being the two most common alleles, and subsequently divided into three groups according to previous published work：〈 30 （GT） n were designated as S （short） , 30 - 37 （GT） n as m （middle） and long repeats with 〉 37 （GT） n as L （long） ; allele frequency 0. 35, 0. 58 and 0.07 respectively. Aims In this study,we examined the effects of Solidago altissima(hereafter Solidago)and two species in the genus Verbesina,Verbesina virginica and Verbesina occidentalis(hereafter Verbesina),on the structure of an old-field plant community and establishment by an invasive plant species,Lespedeza cuneata(hereafter Lespedeza).methods We removed Solidago,Verbesina and both Solidago and Verbesina from 4m^(-2)plots in an intact old-field community during two growing *** then quantified the effects of these removals on richness,evenness,diversity and composition of the subdominant plant *** also measured the total aboveground biomass and the aboveground biomass of the subdominant *** assess how these removals affected establishment by Lespedeza,we planted 20 seeds in each plot and tracked seedling emergence and survival for one growing *** Findings Subdominant community evenness and Shannon diversity were higher in plots from which Solidago and Verbesina were removed relative to control ***,there were no effects of dominant species removal on species richness or composition of the subdominant *** aboveground biomass was not affected by dominant species removal,suggesting that the community of subdominant species exhibited *** fact,subdominant community biomass was greater when Solidago,but not Verbesina,was *** availability was also greater in plots where Solidago was removed relative to control plots throughout the growing *** addition,removal of dominant species,in particular Solidago,indirectly reduced the emergence,but not survival,of Lespedeza seedlings by directly promoting subdominant community *** together,our results suggest that dominant old-field plant species affect subdominant community structure and indirectly promote establishment by Lespedeza.

Numerical study of crude oil batch mixing in a long channel

The main objective of this work is to predict the mixing of two different miscible oils in a very long channel. The background to this problem relates to the mixing of heavy and light oil in a pipeline. As a first step, a 2D channel with an aspect ratio of 250 is considered. The batch-mixing of two miscible crude oils with different viscosities and densities is modeled using an unsteady laminar model and unsteady RANS model available in the commercial CFD solver ANSYSFluent. For a comparison, a LES model was used for a 3D version of the 2D channel. The distinguishing feature of this work is the Lagrangian coordinate system utilized to set no-slip wall boundary conditions. The global CFD model has been validated against classical analytical solutions. Excellent agreement has been achieved. Simulations were carried out for a Reynolds number of 6300(calculated using light oil properties) and a Schmidt number of 10~4. The results show that, in contrast to the unsteady RANS model, the LES and unsteady laminar models produce comparable mixing dynamics for two oils in the channel. Analysis of simulations also shows that, for a channel length of 100 m and a height of 0.4 m, the complete mixing of two oils across the channel has not been achieved. We showed that the mixing zone consists of the three different mixing sub-zones, which have been identified using the averaged mass fraction of the heavy oil along the flow direction. The first sub-zone corresponds to the main front propagation area with a length of several heights of the channel. The second and third sub-zones are characterized by so-called shear-flow-driven mixing due to the Kelvin–Helmholtz vortices occurring between oils in the axial direction. It was observed that the third sub-zone has a steeper mass fraction gradient of the heavy oil in the axial direction in comparison with the second sub-zone, which corresponds to the flow-averaged mass fraction of 0.5 for the heavy oil.
