Alve.lar macrophage. (AM) are.known to play an e.se.tial role.in lung de.e.se.through the.r ability to re.ove.the.fore.gn matte.s re.ching the.lung alve.li. Cigare.te.smoke.(CS) is a critical risk factor for many lung dise.se.. CS is inhale. into the.lung by re.pire.ion and affe.ts AM. It has be.n pre.iously re.orte. that CS induce. inhibition of cytokine.production, ce.l surface.re.e.tor e.pre.sion and antige. pre.e.tation in AM. Howe.e., the.re.ationship of immune.suppre.sion and DNA damage.cause. by CS in AM is still uncle.r. The.e.ore. in this study, we.inve.tigate. AM immune.function and DNA damage.in CS-e.pose. mice. Mice.we.e.e.pose. to CS of 20 cigare.te./day during 10 days using a HambrugⅡsmoking machine. Afte. e.posure. AM we.e.obtaine. by bronchoalve.lar lavage. De.pite.major advance. in ide.tifying ge.e. that can cause.dise.se.whe. mutate. and mode. the.dise.se.in animals and ce.lular mode.s, it still re.ains uncle.r why motor symptoms sudde.ly appe.r afte. a long pre.symptomatic phase.of appare.tly normal function. One.hypothe.is is that age.re.ate. de.e.ulation of spe.ific prote.ns within ke. ce.l type., e.pe.ially motor ne.rons the.se.ve., initiate. dise.se.symptom appe.rance.and may also drive.progre.sive.de.e.e.ation. Howe.e., the.e.has be.n significant disagre.me.t on using both factor of safe.y(Fo S) and w/h as part of pillar syste. stability crite.ion, as compare. to using Fo S in isolation. This pape. will argue.that the.e.are.valid te.hnical re.sons to bring w/h ratio into syste. stability crite.ia(othe. than its influe.ce.on pillar stre.gth), as it is re.ate. to the.post-failure.stiffne.s of the.pillar, as me.sure. in situ, and its inte.action with ove.burde. stiffne.s.
