While peptide separations with nano LC have historically been conducted with fairly long gradient elution and re-equilibration times, the demands for instrumentation that will deliver increased throughput while maintaining the sensitivity benefits of nano ESI has increased. Fast gradient analysis of peptide samples are presented that take advantage of precise gradient control with small volume mixing and low gradient delays. Combined with parallel column approaches, these fast chromatography runs can be used to provide significant increases in sample throughput and data collection. Higher pressure LC operation along with external gradient mixing provide additional tools for further increases in speed, resolving power, and sensitivity to meet the demands of higher throughput experiments in areas such as biomarker analysis and peptide quantification.
