Two principal growth regulators,cytokinins and ethylene,are known to interact in the regulation of plant ***,information about the underlying molecular mechanism and positional specificity of cytokinin/ethylene crosstalk in the control of root growth is *** have identified the spatial specificity of cytokinin-regulated root elongation and root apical meristem(RAM)size,both of which we demonstrate to be dependent on ethylene *** of the cytokinin biosynthetic gene ISOPENTENYLTRANSFERASE(IPT)in proximal and peripheral tissues leads to both root and RAM *** contrast,IPT activation in distal and inner tissues reduces RAM size while leaving the root length comparable to that of mock-treated *** show that cytokinins regulate two steps specific to ethylene biosynthesis:production of the ethylene precursor 1-aminocyclopropane-1-carboxylate(ACC)by ACC SYNTHASEs(ACSs)and its conversion to ethylene by ACC OXIDASEs(ACOs).We describe cytokinin-and ethylene-specific regulation controlling the activity of ACSs and ACOs that are spatially discrete along both proximo/distal and radial root *** direct ethylene measurements,we identify ACO2,ACO3,and ACO4 as being responsible for ethylene biosynthesis and ethylene-regulated root and RAM shortening in cytokinin-treated *** interaction between ARABIDOPSIS RESPONSE REGULATOR 2(ARR2),a member of the multistep phosphorelay cascade,and the C-terminal portion of ETHYLENE INSENSITIVE 2(EIN2-C),a key regulator of canonical ethylene signaling,is involved in the cytokinin-induced,ethylene-mediated control of *** propose tight cooperation between cytokinin and ethylene signaling in the spatially specific regulation of ethylene biosynthesis as a key aspect of the hormonal control of root growth.

Stem cell therapy for the treatment of Leydig cell dysfunction in primary hypogonadism

The production of testosterone occurs within the Leydig cells of the testes. When production fails at this level from either congenital, acquired, or systemic disorders,the result is primary hypogonadism. While numerous testosterone formulations have been developed, none are yet fully capable of replicating the physiological patterns of testosterone secretion. Multiple stem cell therapies to restore androgenic function of the testes are under investigation. Leydig cells derived from bone marrow, adipose tissue, umbilical cord, and the testes have shown promise for future therapy for primary hypogonadism. In particular, the discovery and utilization of a group of progenitor stem cells within the testes, known as stem Leydig cells(SLCs), has led not only to a better understanding of testicular development, but of treatment as well. When combining this with an understanding of the mechanisms that lead to Leydig cell dysfunction, researchers and physicians will be able to develop stem cell therapies that target the specific step in the steroidogenic process that is deficient. The current preclinical studies highlight the complex nature of regenerating this steroidogenic process and the problems remain unresolved. In summary, there appears to be two current directions for stem cell therapy in male primary hypogonadism. The first method involves differentiating adult Leydig cells from stem cells of various origins from bone marrow, adipose, or embryonic sources. The second method involves isolating, identifying, and transplanting stem Leydig cells into testicular tissue. Theoretically, in-vivo re-activation of SLCs in men with primary hypogonadism due to age would be another alternative method to treat hypogonadism while eliminating the need for transplantation.

A hierarchical path-segmentation movement ecology framework

Regulating regulatory T cells to achieve transplant tolerance

BACKGROUND:Regulatory T cells(Tregs) play crucial roles in both induction and maintenance of tolerance. This active immune regulation may contribute not only to the control of immune responses to self-antigens and thereby prevent autoimmune diseases,but also the control of responses to non-self molecules in adaptive immunity. Numerous experimental and clinical studies indicate that manipulating the balance between regulatory and responder T cells is an effective strategy to control immune responsiveness after transplantation. DATA SOURCES:Literature search was conducted using PubMed on the related subjects. Part of the material was based on the most recent work in the authors’ laboratory. RESULTS:We propose some new strategies to achieve transplant tolerance in rodent animals via manipulating Treg function,including using histone deacetylase(HDAC) inhibitor to regulate Foxp3 transcription and enhance Treg suppression,induction of Treg-sparing apoptosis via Nur77,and identification of the co-inhibitory molecule herpes virus entry mediator(HVEM) as an effector molecule for Treg function. CONCLUSION:Regulation of Treg function will definitely provide us very promising tools to achieve clinical tolerance in the future.

A fully-customized dataflow engine for 3D earthquake simulation with a complex topography

With HPC(high performance computing) evolving into the exascale era, improvements in computing performance and power efficiency have become increasingly more important. Based on our previous work on enabling earthquake simulations on a large scale on Sunway Taihu Light, we further explore other possibilities to improve the application through a fully-customized hardware design on reconfigurable FPGA(field programmable gate array) devices. We investigate the feasibility and the potential benefits of a complete fixed-point design. We first perform a coarse-resolution-based simulation to analyze the representation range and precision needed to capture both the total energy and the energy distribution of variables over space and time. We then derive a complete fixed-point design that identifies the suitable bitwidth for major categories of variables and dynamically represents the range through a dynamic scaling scheme. Finally, we use the optimized fixed-point design to run a case of the Wenchuan earthquake to demonstrate the potential of supporting large-scale scientific simulations on FPGA devices. The results demonstrate that an 18-bit fixed-point design already provides an almost identical description of the seismic events in the Wenchuan scenario down to a single-precision floating-point version and provides sustainable performance equivalent to 13.1 Intel Xeon Gold 6154 18-core CPUs or 2.10 Sunway 260-core processors, with performance per watt(power efficiency) improved by 15.3 and 3.72 times compared with the Intel Xeon Gold 6154 18-core CPUs and the Sunway 260-core processors, respectively.

Carcinoid of the ampulla of Vater: Morphologic features and clinical implications

Carcinoids involving the ampulla of Vater are rare le- sions that may produce painless jaundice. The published data indicate that these tumors, in contrast to their midgut counterparts, metastasize in approximately half of cases irrespective of primary tumor size. Therefore, radical excision in the form of pancreaticoduodenectomy is recommended regardless of tumor size. As with other gastrointestinal carcinoid tumors, biological treatment with octreotide analogues can be applied to symptomatic patients. Tumor-targeted radioactive therapy is a newly emerging treatment option. We here report case of a carcinoid tumor of the ampulla of Vater presenting as painless jaundice in a 65-year old man and review the relevant literature, giving special attention to the mor- phologic features, clinical characteristics, and treatment modalities associated with this disease process.

Current updates on laboratory techniques for the diagnosis of male reproductive failure

The incidence of male reproductive failure leading to infertility, whether due to delayed parenthood, environmental issues, genetic factors, drugs, etc., is increasing throughout the world. The diagnosis and prognosis of male subfertility have become a challenge. While the basic semen assessment has been performed for many years, a number of studies question the value of the traditional semen characteristics. This is partly due to inadequate methods and standardization, limited knowledge of technical requirements for quality assurance, and an incomplete understanding of what clinical information a semen assessment can provide. Laboratories currently performing semen and endocrine assessment show great variability. The World Health Organization （WHO） manual for the evaluation of semen has been the core of andrology and fertility evaluation that has helped in further development of this field over many years. These include the physical appearance of the ejaculate, assessments of sperm count, motility, vitality, morphology, and functional aspects of the sperm and semen sample. These tests also include male endocrine profile, biochemical evaluation of the semen, detection of antisperm antibodies in serum, the use of computer-aided sperm analysis （CASA）, sperm DNA integrity, and its damage due to oxidative stress. Assisted reproductive techniques （e.g., IVF, ICSI） have shown great success but are too expensive. Further development in this field with newer techniques and extensive training/instructions can improve accuracy and reduce variability, thus maintaining the quality and standards of such an evaluation. There is an urgent need to have standardized training centers and increased awareness in this area of men＇s health for reproductive success.

Forage and breed effects on behavior and temperament of pregnant beef heifers
