Regulative effect for natural killer cell by hot spring hydrotherapy—Quantitative and qualitative discussion

Along with maintaining immune competent cells, one of the purposes of cancer patient is regulating the first line of defense for survival. Moreover, the factors that influence the acquired immune activity are systemic metabolic disorder in diabetes, malnutrition, extreme exhaustion, stresses, aging and medical side effect such as chemotherapy. So we have to select appropriate menu to regulate immune function through leukocyte storage. Especially, NK cell is first line of defense against virus infected cell and/or tumor cell. In order to recommend the effect of hot spring hydrotherapy on the peripheral white blood cells as a parameter for complementary and alternative medicine (CAM) within a short period, the granulocytes, lymphocytes, lymphocyte subsets in their peripheral blood were monitored. In the young group (35 or less), the number of granulocytes clearly decreased after hydrotherapy. In the older group (over 35 years old), the total number of leukocytes and lymphocytes significantly increased after the therapy. Using a FACScan, we investigated CD2, CD4, CD8, CD16, CD19 and CD56 positive cell as well as the cytokine containing cell, as direct assessment for immunological preparation (IFN-γ, IL-4 and IL-1β). After hydrotherapy, the CD16+ cells also increased outstandingly. However, the CD19+ cells clearly decreased. Moreover, there was a decrease in the cytokine-producing cell count of subjects whose blood cells had a higher number of the cell level before. All of the results indicated that hot spring hydrotherapy could regulate the immune system even in normal human. Like this, blood hormonal level was measured in order to know the mechanism of the quantitative and qualitative, adjustment of the leucocytes becomes possible within a short term. As a result of measuring, 7 kinds of hormone including mainly adrenocortical hormones, adrenalin level reduced in constitution dependent manner. Usefulness of Basic Fibroblast Growth Factor (bFGF) Loaded Dissolving Microneedles for Local Therapy of Skin Wounds

Purpose: The usefulness of dissolving microneedles (DMs) for local skin therapy by basic fibroblast growth factor (bFGF) was studied in rats. Methods: We prepared four kinds of bFGF-loaded DMs, approximately 500 μm length and 300 μm diameter at the bottom. Long-term stability and dissolution studies were performed by HPLC method. Pharmacokinetic and pharmacological evaluations were performed after administration of bFGF loaded DMs to rats. Results: The bFGF contents were 2.15 ± 0.07, 1.07 ± 0.04, 0.56 ± 0.07 and 0.12 ± 0.03 μg. The 100.2 ± 3.4%, 100.2 ± 3.3%, 99.3 ± 1.4% and 100.4 ± 3.0% of bFGF were recovered after 1, 3 and 6 months and 1 year incubation at 40°C. The bFGF was released from DMs within 5 min. In a pharmacokinetic study using 2.0 and 1.0 μg bFGF-loaded DMs, no systemic exposure of bFGF was detected. The initial bFGF concentrations in the rat skin tissue after administration of bFGF-loaded DMs to the hair-removed rat abdominal skin were 510.2 ± 20.1 ng/g wet weight for 2 μg bFGF DMs and 264.2 ± 56.5 ng/g wet weight for 1 μg DMs, declining slowly thereafter to 226.3 ± 33.5 and 105.1 ± 27.4 ng/g wet weight at 6 hr after administration. Good dose-dependency was observed. Pharmacological evaluation of bFGF-loaded DMs of 2.0, 1.0, 0.5, and 0.1 μg, in the wound healing rat model, all used DMs, but 0.1 μg DMs, showed good healing effects. Considered collectively, these results suggest the usefulness of bFGF-loaded DMs for local therapy of skin wound disease.

Effects of Image Presentation Using a Head-Mounted Display on Muscle and Brain Activity during Swallowing

Introduction: It is well known that humans have an increased sense of taste and consume more food when eating with others. This effect is called social facilitation of eating. We previously reported that self-images also have the effect of social facilitation of eating. In the present study, we investigated the effect of a head-mounted display (HMD) as a means of presenting self-images. Methods: Fourteen healthy adults (7 females and 7 males) were included in the study. The mean age of the participants was 20.4 years. The experimental conditions were as follows: the presentation conditions include the SG condition, in which an image was projected on the smart glasses, and the MT condition, in which an image was projected on the monitor. The stimulation conditions include the self-image condition (Self condition), in which the participant’s own image was projected, and the background image condition (Background condition), in which the background image was projected. Participants were asked to consume green tea jelly three times under the four conditions, which consisted of combinations of the presentation and stimulation conditions. Electromyography (EMG) and electroencephalography (EEG) were used to measure the muscle activity of the suprahyoid muscle group and brain activity, respectively. Results: The EMG of the “SG-Self” condition was significantly higher than that of “SG-Background” and “MT-Background” conditions. The EMG of the “MT-Self” condition was significantly higher than that of the “MT-Background” condition. In the EEG before the start of swallowing, “SG-Self” was significantly lower in the alpha band and higher in the beta band than “MT-Background”. The beta-alpha ratio was significantly higher for “SG-Self” than for “SG-Background” and “MT-Background” and higher for “MT-Self” than for “MT-Background”. SINGLE AND DOUBLE EDGE INTERFACE CRACK SOLUTIONS FOR ARBITRARY FORMS OF MATERIAL COMBINATION

In this paper interfacial edge crack problems are considered by the application of the finite element method. The stress intensity factors are accurately determined from the ratio of crack-tip-stress value between the target given unknown and reference problems. The reference problem is chosen to produce the singular stress fields proportional to those of the given unknown problem. Here the original proportional method is improved through utilizing very refined meshes and post-processing technique of linear extrapolation. The results for a double-edge interface crack in a bonded strip are newly obtained and compared with those of a single-edge interface crack for different forms of combination of material. It is found that the stress intensity factors should be compared in the three different zones of relative crack lengths. Different from the case of a cracked homogeneous strip, the results for the double edge interface cracks are found to possibly be bigger than those for a single edge interface crack under the same relative crack length.

Generation of spectrally stable 6.5-fs visible pulses via filamentation in krypton

We produced 5-μJ, 6.5-fs visible pulses at a repetition rate of 1 kHz using filamentation in a gas cell filled with krypton followed by spectral selection and phase compensation by a combination of dielectric mirrors. The visible pulses have a smooth spectrum from 520 to 650 nm with a shot-to-shot stability in each spectral component of better than 2%(standard deviation). This pulse compression scheme is simple and robust, and can be easily integrated into intense ultrashort-pulse laser systems.

Characterization of Three Homoeologous cDNAs Encoding Chloroplast-targeted Aminolevulinic Acid Dehydratase in Common Wheat

In the tetrapyrrole biosynthetic pathway of higher plants, 5-aminolevulinic acid （ALA） is metabolized by ALA dehydratase （ALAD）. Here, we isolated ALAD1 cDNA from common wheat （Triticum aestivum L.） and its diploid progenitors, and produced transgenic tobacco plants expressing the wheat ALAD1 gene. The ALAD1 genes were highly conserved among wheat relatives, and three homoeologous loci of wheat ALAD1 （TaALAD1） were equally transcribed in common wheat. A transient expression assay of a TaALAD1-GFP （green fluorescent protein） fusion protein suggested that TaALAD1 is localized in chloroplasts. Overexpression of TaALAD1 in transgenic tobacco resulted in a significant increase in ALAD activity in leaves. Moreover, the transgenic tobacco showed vigorous growth and increased survival rate on medium containing ALA at herbicidal concentrations. These results indicate that wheat ALAD1 has catalytic activity in metabolizing ALA in plastids, and that ectopic expression of TaALAD1 in transgenic plants increases their tolerance to ALA application at high concentrations.
