Glutathione as a Prognostic Biomarker and a Potential Therapeutic Target for Ovarian Cancer

Aim: Glutathione (GSH) is an antioxidant, protecting cell against toxicity of reactive oxygen species (ROS). Data showed that GSH might play roles in malignancy including ovarian cancer (OC), and, thus, we attempted to determine the clinical significance of GSH and effects of erastin (an inhibitor of GSH synthesis) in OC. Methods: OC tissues were taken from 41 OC patients, and cancer-tissue GSH level was measured with GSH Assay Kit. Survival curves were carried out by the Kaplan-Meier method and evaluated using the log-rank test. Multivariable Cox proportional hazard risk regression model was performed to screen the independent factor affecting the prognosis of OC patients. In vitro effect of erastin was studied using OC cell lines. Cell viability, GSH levels and whole (cytosolic and lipid) ROS production were assessed. Results: Patients with high OC-tissue-GSH levels had an apparently lower progression free survival (PFS) and overall survival (OS) compared with those with low GSH levels. The GSH levels were independent factors for predicting the PFS and OS. The basal ROS level was inversely proportional to GSH levels in OC cell lines. The basal GSH levels were important for estimating the sensitivity to erastin. Reduction of intracellular GSH levels increased whole ROS, which caused cell deaths. Conclusions: Data suggested that the GSH levels could be a candidate of prognostic biomarkers and that erastin might be worth studying as a new therapeutic drug in OC.

Conservative Medroxyprogesterone Acetate Therapy in Early Stage of Endometrial Carcinoma Associated with Phosphatase and Tensin Homolog Expression

Young patients with the endometrial cancer IA who desire to preserve fertility, can select the conservative therapy with progestin. However, the therapy involves risks of progression and relapse. We examined immunohistochemical analyses of phosphatase and tension homolog (PTEN) and p53 expressions to predict the early relapse, and pregnancy and delivery. Twenty women with endometrial cancer, FIGO IA (1988) (FIGO staging was essentially defined post-surgically), instead of the pathogical specimen before surgery without myometrial invasion were estimated by MRI under 40 years at Gifu University Hospital, Japan from 1988 to 2009. Patients were treated with medroxyprogesterone acetate (MPA, 400 - 600 mg/day) for 4 - 10 months, with whole wall endometrial curettage performed every four weeks. Response to the therapy, pregnancy, delivery and relapse of disease during follow-up over a 72-month period. Immunohistochemical expression of PTEN and p53 was also evaluated with pregnancy, delivery and relapse rate. All patients had pathological complete remissions within 4 - 10 months. Relapse rate was high (60%) in more than 72 months. Immunohistochemical PTEN retain in tumor cells before MPA treatment (8/10) was significant better correlation with pregnancy and delivery rate than of lost cases (1/5) in non-obese women (P < 0.05). Conservative therapy is feasible in carefully selected young women with endometrial cancer without myometrial invasion. However, the relapse rate was high. In cases who desire to be a pregnant, an earlier infertility treatment may be considered especially for PTEN loss especially in nonobese cases.

Genomic Recombination Enhances Pathogenic Factors in the Periodontopathogenic Bacterium <i>Eikenella corrodens</i>

We reported previously that plasmid-mediated genomic recombination at the pilin gene locus increased hemagglutination activity, growth rate, biofilm formation, hemolytic activity, and adherence to epithelial cells in Eikenella corrodens 23834. To determine whether these enhancements were common in this bacterium, we introduced the recombinase gene ORF4 into seven clinically isolated strains. Genomic recombination at the type IV pilin gene locus was observed in strains 1080, L9B6, L8Ao3, and RV2 (group A), but not in strains 261-2, 612-L, and 257-4 (group B). Similarly, group A strains displayed changed colony morphology following loss of type IV pili, which was not observed in group B. Group A strains showed also enhanced hemagglutination activity, growth rate, hemolytic, activity and biofilm formation. These results suggest that ORF4-induced genomic recombination at the pilin gene locus is a general phenomenon in a part of E. corrodens, which likely stimulates patho-genicity and virulence.

＜正＞Background Psoriasis represents one of the Th17-mediated autoimmune diseases,which has been already shown to be associated with metabolic *** adipokines and Th17-related cytokines have been reported to be altered in patients with *** aim of the present study is to investigate the correlations of adipokines with Th17-related cytokines in psoriasis patients. Methods Serum adipokines and Th17-related cytokines were measured by enzyme-linked immunosorbent assays （Th-17 related cytokines:TNF-alpha,IL-6,IL-17,IL-21,IL-22,IL-23,adipokines:resistin,retinol-binding protein-4 （RBP-4）,chemerin,leptin,total adiponectin,high molecular weight adiponectin） from 30 psoriasis patients and 30 normal *** between each adipokine and each cytokine was determined using Pearson correlation analysis. Results Pearson correlation analysis showed that chemerin levels were highly positively associated with resistin levels in normal ***,adiponectin and high molecular weight adiponectin showed a strong positive correlation in normal *** psoriasis patients,resistin levels were significantly positively associated with TNF-alpha,whereas RBP4 and IL-6 levels showed a highly negative ***,a remarkably positive correlation between IL-22 and adiponectin was found in psoriasis patients. Conclusion These data suggest that distinct interaction between adipokines and Th17 cytokines is involved in psoriasis.

Purpose:To investigate whether corneal epithelial cells of individuals with lattice corneal dystrophy(LCD) possess an intrinsic ***:Retrospective case-control ***:The medical charts of nine individuals with LCD and those of 14 patients with keratoconus and 11 patients with corneal leukoma(controls) ,all of whom underwent penetrating keratoplasty(PKP) in one eye at Yamaguchi University Hospital between February 1998 and November 2001,were examined for the time for epithelial resurfacing after ***:The time required for resurfacing of the corneal epithelium after PKP was significantly greater in LCD patients(8.56± 4.95 days,mean± SD) than in patients with either keratoconus(1.71± 0.91 days,P=.006) or corneal leukoma(3.00± 1.95 days,P=.03) .Conclusions:Corneal epithelial wound healing was delayed in LCD patients after PKP,suggesting that the keratoepithelin gene mutations responsible for this condition affect corneal epithelial cells.
