AIM: To clarify the relationship between autophagy and lipotoxicity-induced apoptosis, which is termed "lipoapoptosis," in non-alcoholic steatohepatitis. METHODS: Male C57BL/6J mice were fed a high-fat diet(HFD) for 12 wk, after which the liver histology and expression of proteins such as p62 or LC3 were evaluated. Alpha mouse liver 12(AML12) cells treated with palmitate(PA) were used as an in vitro model. RESULTS: LC3-Ⅱ, p62, and Run domain Beclin-1 interacting and cysteine-rich containing(Rubicon) proteins increased in both the HFD mice and in AML12 cells in response to PA treatment. Rubicon expression was decreased upon c-Jun N-terminal kinase(JNK) inhibition at both the m RNA and the protein level in AML12 cells. Rubicon knockdown in AML12 cells with PA decreased the protein levels of both LC3-Ⅱ and p62. Rubicon expression peaked at 4 h of PA treatment in AML12, and then decreased. Treatment with caspase-9 inhibitor ameliorated the decrease in Rubicon protein expression at 10 h of PA and resulted in enlarged AML12 cells under PA treatment. The enlargement of AML12 cells by PA with caspase-9 inhibition was canceled by Rubicon ***: The JNK-Rubicon axis enhanced lipoapoptosis, and caspase-9 inhibition and Rubicon had effects that were cytologically similar to hepatocyte ballooning. As ballooned hepatocytes secrete fibrogenic signals and thus might promote fibrosis in the liver, the inhibition of hepatocyte ballooning might provide antifibrosis in the NASH liver.

AIM: To identify the hepatic vascular architecture of patients with hepatitis C virus (HCV) using superb microvascular imaging (SMI) and investigate the use of SMI in the evaluation of liver ***: SMI was performed in 100 HCV patients. SMI images were classified into five types according to the vascular pattern, and these patterns were compared with the fibrosis stage. Moreover, the images were analyzed to examine vascularity by integrating the number of SMI signals in the region of interest ROI [number of vascular trees (VT)]. The number of VT, fibrosis stage, serum parameters of liver function, and CD34 expression were ***: There was a significant difference between SMI distribution pattern and fibrosis stage (P < 0.001). The mean VT values in each of the fibrosis stages were as follows: 26.69 ± 7.08 in F0, 27.72 ± 9.32 in F1, 36.74 ± 9.23 in F2, 37.36 ± 5.32 in F3, and 58.14 ± 14.08 in F4. The VT showed excellent diagnostic ability for F4 [area under the receiver operator characteristic (AUROC): 0.911]. The VT was significantly correlated with the CD34 labeling index (r = 0.617, P < 0.0001).CONCLUSION: SMI permitted the detailed delineation of the vascular architecture in chronic liver disease. SMI appears to be a reliable tool for noninvasively detecting significant fibrosis or cirrhosis in HCV patients.

AIM:To use leptin-deficient(ob/ob) mice with demonstrated differences in steatosis levels to test a new diagnostic method using the acoustical structure quantification(ASQ) mode and the associated analytical parameter,"focal disturbance ratio"(FD-ratio).METHODS:Nine ob/ob mice,at 5,8,and 12 wk of age(n = 3 in each age group),were used as models for hepatic *** signals obtained from ultrasonography in the mice were analyzed by ASQ,which uses a statistical analysis of echo amplitude to estimate inhomogeneity in the diagnostic ***-ratio,as calculated from this analysis,was the focus of the present ***-ratio and fat droplet areas and sizes were compared between age ***:No fibrosis or inflammation was observed in any of the *** fat droplet area significantly(P < 0.01) increased with age from 1.25% ± 0.28% at 5 wk to 31.07% ± 0.48% at 8 wk to 51.69% ± 3.19% at 12 *** median fat droplet size also significantly(P < 0.01) increased with age,from 1.33(0.55-10.52) m at 5 wk,2.82(0.61-44.13) m at 8 wk and 6.34(0.66-81.83) m at 12 *** mean FD-ratio was 0.42 ± 0.11 at 5 wk,0.11 ± 0.05 at 8 wk,and 0.03 ± 0.02 at 12 *** FD-ratio was significantly lower at 12 wk than at 5 wk and 8 wk(P < 0.01).A significant negative correlation was observed between the FD-ratio and either the fat droplet area(r =-0.7211,P = 0.0017) or fat droplet size(r =-0.9811,P = 0.0052).CONCLUSION:This tool for statistical analysis of signals from ultrasonography using the FD-ratio can be used to accurately quantify fat in vivo in an animal model of hepatic steatosis,and may serve as a quantitative biomarker of hepatic steatosis.

Portal vein stenosis after pancreatectomy following neoadjuvant chemoradiation therapy for pancreatic cancer

Extrahepatic portal vein (PV) stenosis has various causes, such as tumor encasement, pancreatitis and as a postsurgical complication. With regard to post-pancreaticoduodenectomy, intraoperative radiation therapy with/ without PV resection is reported to be associated with PV stenosis. However, there has been no report of PV stenosis after pancreatectomy following neoadjuvant chemoradiation therapy (NACRT). Here we report the cases of three patients with PV stenosis after pancreatectomy and PV resection following gemcitabine-based NACRT for pancreatic cancer and their successful treatment with stent placement. We have performed NACRT in 18 patients with borderline resectable pancreatic cancer since 2005. Of the 15 patients who completed NACRT, nine had undergone pancreatectomy. Combined portal resection was performed in eight of the nine patients. We report here three patients with PV stenosis, and thus the ratio of post-operative PV stenosis in patients with PV resection following NACRT is 37.5% in this series. We encountered no case of PV stenosis among 22 patients operated with PV resection for pancreatobiliary cancer without NACRT during the same period. A relationship between PV stenosis and NACRT is suspected, but further investigation is required to determine whether NACRT has relevance to PV stenosis.

The Xiaoxinancha Au-rich copper deposit is one of important Au-Cu deposits along the continental margin in Eastern China. The deposit consists of two sections: the Beishan mine (North), composed of altered rocks with veinlet-dissemination sulfides and melnicovite-dominated sulfide-quartz veins, and the Nanshan mine (South), composed of pyrrhotite-dominated sulfide-quartz veins and pure sulfide veins. The isotope compositions of noble gases extracted from fluid inclusions in ore minerals, i.e. ratios of 3He/4He, 20Ne/22Ne and40Ar/36Ar are in the ranges of 4.45―0.08 Ra, 10.2―8.8 and 306―430, respectively. Fluid inclusions in minerals from the Nanshan mine have higher 3He/4He and 20Ne/22Ne ratios whereas those from the Beishan mine have lower 3He/4He ratios. The analysis of origin, and evolution of the ore fluids and its relations with the ore-forming stages and the ages of mineralization suggests that the initial hydrothermal fluids probably come from the melts generated by partial melting of oceanic crust with the participation of fluids from the mantle (mantle-plume type)/aesthenosphere. This also corresponds to the continental margin settings during the subduction of Izanagi ocaneic plate towards the palaeo-Asian continent (123―102 Ma). The veinlet-dissemination ore bodies of the Beishan mine were formed through replacement and crystallization of the mixed fluids generated by mixing of the ascending high-temperature boiling fluid with young crustal fluid whereas the melnicovite-dominated sulfide-quartz veins were formed subsequently by filling of the high-temperature ore fluid in fissures. Pyrrhotite-dominated sulfide-quartz veins in the Nanshan mine were formed by filling-deposition-crystallization of the moderate-temperature ore fluids and the pure sulfide veins were formed later by filling-deposition-crystallization of ore substance-rich fluids after boiling of the moderate-temperature ore fluids. Electric power generation characteristics of lead zirconate titanate (PZT) piezoelectric ceramic have been examined during cyclic loading with different compressive strain value. A thin PZT ceramic plate attached to a thin brass plate was used. With an increasing compressive strain value in the PZT ceramic plate, the electric voltage increased, and the highest electric voltage was obtained for the sample with the high strain level (0.4%). The electric voltage decreased for the sample with more than 0.45% strain because of the failure in the PZT ceramic. The electric voltage was different depending on the strain condition, where the higher strain and the wider strain range (Δε) made the high electric voltage. Moreover, the electric voltage was attributed to the strain rate, where the higher strain rate made higher electric voltage due to high kinetic energy. The electric generation characteristics could be estimated with an operation of the strain value and strain ratio (εmin/εmax) especially for the sample loaded cyclically under the slow loading speed. This was affected by the linear correlation between the electric voltage and the strain value. Based upon the experimental work, suitable loading condition to make high electric voltage could be proposed.

Ferritic heat resistant steels involving precipitation of intermetallic phases have drawn a growing interest for the enhancement of creep strength, while the brittleness of the intermetallic phases may lower the toughness of the alloy. Therefore, it is necessary to optimize the dispersion characteristics of the intermetallics phase through microstructural control to minimize the trade-off between the strength and toughness. The effects of -Fe matrix substructures on the precipitation sequence, morphology, dispersion characteristics, and the stability of the intermetallic phases are investigated in Fe-Cr-W-Co-Si system. The precipitates of the Si-free Fe-10Cr-l.4W-4.5Co (at%) alloy aged at 873K are the R-phase but those of the Si-added Fe-10Cr-l.4W-4.5Co-0.3Si (at%) alloy are the icosahedral quasicrystalline phase. The precipitates in both the Si-free and Si-added alloys aged at 973K are the Laves phase. Matrix of the alloys is controlled by heat treatments as to provide three types of matrix substructures; ferrite, ferrite/martensite mixture and martensite. The hardening behavior of the alloys depends on the matrix substructures and is independent of the kinds of precipitates. In the alloys with ferrite matrix, the peak of hardness during aging at 873K shifts to longer aging time in comparison with that in the alloys with lath martensite matrix which contain numbers of nucleation sites.
