Pseudomonas syringae ***(Psa)causes bacterial canker,a devastating disease threatening the Actinidia fruit *** a search for non-host resistance genes against Psa,we find that the nucleotidebinding leucine-rich repeat receptor(NLR)protein ZAR1 from both Arabidopsis and Nicotiana benthamiana(Nb)recognizes Hop Z5 and triggers cell *** recognition requires ZED1 in Arabidopsis and jIM2 in Nb plants,which are members of the ZRK pseudokinases and known components of the ZAR1 ***,Arabidopsis ZAR1 and RPM1,another NLR known to recognize Hop Z5,confer disease resistance to Hop Z5 in a strain-specific ***,ZAR1,but not RPM1,is solely required for resistance to *** ES4326(Psm)carrying hop Z5,whereas RPM1 is primarily required for resistance to *** DC3000(Pst)carrying hop ***,the ZAR1-mediated resistance to Psm hop Z5 in Arabidopsis is insensitive to SOBER1,which encodes a deacetylase known to suppress the RPM1-mediated resistance to Pst hop *** addition,hop Z5 enhances *** virulence in the absence of ZAR1 or RPM1 and that SOBER1 abolishes such virulence *** the study suggests that ZAR1 may be used for improving Psa resistance in Actinidia and uncovers previously unknown complexity of effectortriggered immunity and effector-triggered virulence.

The phospholipase A2 （PLA2） family members arc critical regulators of membrane structure and lipid composition and have been implicated in neuroinflammation, oxidative stress and neurodegeneration. Here, we review the published data describing regulation of cPLA2 and iPLA2 gene expression. Based on promoter sequence, cPLA2 expression can be regulated by glucocorticoid and pro-inflammatory cytokines, whereas transcription of iPLA2 can be controlled in response to sterol level. RNA degradation in 3＇ UTR and epigenetic mechanisms may be involved in the regulation of cPLA2 and iPLA2 expression, respectively. MicroRNA target sequences lie within cPLA2 and iPLA2 mRNAs. Together, these findings indicate differential regulation of cPLA2 and iPLA2 expression. It is hoped that determination of diverse regulatory mechanisms of the PLA2 family may open new doors for development of novel therapeutic compounds that modulate PLA2 expression and function in the treatment of brain diseases.

Cervical spine fractures in osteopetrosis:a case report and review of the literature

While management of appendicular fractures has been well described in the setting of osteopetrosis, there is limited information on managing fractures of the axial spine. Here we present an osteopetrotic patient with multiple traumatic multiple, comminuted, unstable cervical spinal fractures managed with non-operative stabilization, and provide a review of the pathophysiology, genetic characteristics, and special considerations that must be explored when determining operative versus non-operative management of spinal injury in osteopetrosis. A PubMed query was performed for English articles in the literature published up to june 2016, and used the following search terms alone and in combination：＂osteopetrosis＂, ＂spine＂, ＂fractures＂, ＂osteoclasts＂, and ＂operative management＂. Within four months after initial injury, treatment with halo vest allowed for adequate healing. The patient was asymptomatic with cervical spine dynamic radiographs confirming stability at four months. On four-year follow up examination, the patient remained without neck pain, and CT scan demonstrated partially sclerotic fracture lines with appropriate anatomical alignment. In conclusion, external halo stabilization may be an effective option for treatment of multiple unstable acute traumatic cervical spine fractures in patients with osteopetrosis. Given the challenge of surgical stabilization in osteopetrosis, further research is necessary to elucidate the optimal form of treatment in this select patient population.

Withania sominifera （Indian ginseng） was transformed by Agrobacterium rhizogenes. Explants from seedling roots, stems, hypocotyls, cotyledonary nodal segments, cotyledons and young leaves were inoculated with A. rhizogenes strain R1601. Hairy （transformed） roots were induced from cotyledons and leaf explants. The transgenic status of hairy roots was confirmed by polymerase chain reaction using nptll and rolB specific primers and, subsequently, by Southern analysis for the presence of nptll and rolB genes in the genomes of transformed roots. Four clones of hairy roots were established; these differed in their morphology. The doubling time of faster growing cultures was 8-14 d with a fivefold increase in biomass after 28 d compared with cultured, non-transformed seedling roots. MS-based liquid medium was superior for the growth of transformed roots compared with other culture media evaluated （SH, LS and N6）, with MS-based medium supplemented with 40 g/L sucrose being optimal for biomass production. Cultured hairy roots synthesized withanolide A, a steroidal lactone of medicinal and therapeutic value. The concentration of withanolide A in transformed roots （157.4 μg/g dry weight） was 2.7-fold more than in non-transformed cultured roots （57.9 μg/g dry weight）.

Arthrodesis of the sacroiliac joint（SIj） for surgical treatment of SIj dysfunction has regained interest among spine *** techniques described in the literature most often utilize intraoperative fluoroscopy to aid in implant placement;however,image guidance for SIj fusion may allow for minimally invasive percutaneous instrumentation with more precise implant *** the following cases,we performed percutaneous stereotactic navigated sacroiliac instrumentation using O-arm^（？）multidimensional surgical imaging with StealthStation^（？）navigation（Medtronic,***,MN）.Patients were positioned prone and an image-guidance reference frame was placed contralateral to the surgical site.O-arm^（？） integrated with StealthStation^（？） allowed immediate *** skin incision was planned with an image-guidance *** image-guided awl,drill and tap were utilized to choose a starting point and *** titanium cage（s） packed with autograft and/or allograft were then placed.O-arm^（？） image-guidance allowed for implant placement in the SIj with a small skin ***,we could not track the cage depth position with our current system,and in one patient,the SIj cage had to be revised secondary to the anterior breach of sacrum.

Distribution of pericellular matrix molecules in the temporomandibular joint and their chondroprotective effects against inflammation

The objectives of this study were to （1） determine the distribution and synthesis of pericellular matrix （PCM） molecules （collagen VI, collagen IV and laminin） in rat temporomandibular joint （TMj） and （2） investigate the effects of PCM molecules on chondrocytes against inflammation in osteoarthritis. Four zones （fibrous, proliferating, mature and hypertrophic） of condylar cartilage and three bands （anterior, intermediate and posterior） of disc were analysed by immunohistochemistry for the presence of PCM molecules in rat TMjs. Isolated chondrocytes were pre-treated with PCM molecules before being subjected to interleukin （IL）-II~ treatment to stimulate inflammation. The responses of the chondrocytes were analysed using gene expression, nitric oxide release and matrix metalloproteinase （MMP）-13 production measures. Histomorphometric analyses revealed that the highest areal deposition of collagen VI （67.4%）, collagen IV （45.7%） and laminin （52.4%） was in the proliferating zone of TMj condylar cartilage. No significant difference in the distribution of PCM molecules was noted among the three bands of the TMj disc. All three PCM molecules were expressed intracellularly by chondrocytes cultured in the monolayer. Among the PCM molecules, pre-treatment with collagen VI enhanced cellular proliferation, ameliorated IL-lp-induced MMP-3, MMP-9, MMP-13 and inducible nitric oxide synthase gene expression, and attenuated the downregulation of cartilage matrix genes, including collagen I, aggrecan and cartilage oligomeric matrix protein （COMP）. Concurrently, collagen VI pretreatment inhibited nitric oxide and MMP-13 production. Our study demonstrates for the first time the distribution and role of PCM molecules, particularly collagen VI, in the protection of chondrocytes against inflammation.
