Objective Kidney renal clear cell carcinoma(KIRC)is a common renal malignancy that has a poor prognosis.As a member of the F box family,cyclin F(CCNF)plays an important regulatory role in normal tissues and tumors.However,the underlying mechanism by which CCNF promotes KIRC proliferation still remains unclear.Methods Bioinformatics methods were used to analyze The Cancer Genome Atlas(TCGA)database to obtain gene expression and clinical prognosis data.The CCK8 assay,EdU assay,and xenograft assay were used to detect cell proliferation.The cell senescence and potential mechanism were assessed by SA-β-gal staining,Western blotting,as well as ELISA.Results our data showed that CCNF was highly expressed in KIRC patients.Meanwhile,downregulation of CCNF inhibited cell proliferation in vivo and in vitro.Further studies showed that the reduction of CCNF promoted cell senescence by decreasing cyclin-dependent kinase 1(CDK1),increasing the proinflammatory factors interleukin(IL)-6 and IL-8,and then enhancing the expression of p21 and p53.Conclusion We propose that the high expression of CCNF in KIRC may play a key role in tumorigenesis by regulating cell senescence.Therefore,CCNF shows promise as a new biomarker to predict the clinical prognosis of KIRC patients and as an effective therapeutic target.

河马(Hippo)信号通路最初作为一种抑制果蝇组织生长的通路被发现,主要由MST1/2、LATS1/2和Yap/TAZ三种激酶级联组成。随着研究的进展,在哺乳动物中Hippo信号通路的同源基因也得到了证实,并且在控制器官大小和其他生理功能当中也发挥着关键的作用。Hippo信号通路主要通过调控Yap的核移位发生相应的作用。当Hippo信号通路的上游激酶失活时,Yap/TAZ被去磷酸化并且能够作为转录共激活因子进入细胞核,与相应的转录因子结合发挥作用。既往Hippo-Yap信号通路研究主要集中于细胞命运、新陈代谢、肿瘤发生和免疫系统等方面。随着研究的逐步深入以及肝脏疾病的发生率逐步升高,Hippo信号通路在肝脏疾病的发生与发展过程中的相关研究取得一定的进展。笔者从多种临床常见相关肝脏疾病(胆汁淤积性肝损伤、肝缺血再灌注损伤、非酒精性脂肪性肝病、酒精性肝病、对乙酰氨基酚诱导肝损伤、肝纤维化以及肝癌)阐述Hippo-Yap信号通路在其中所发挥作用。

In 2020 and 2021,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),a novel coronavirus,caused a global pandemic.Vaccines are expected to reduce the pressure of prevention and control,and have become the most effective strategy to solve the pandemic crisis.SARS-CoV-2 infects the host by binding to the cellular receptor angiotensin converting enzyme 2(ACE2)via the receptor-binding domain(RBD)of the surface spike(S)glycoprotein.In this study,a candidate vaccine based on a RBD recombinant subunit was prepared by means of a novel glycoengineered yeast Pichia pastoris expression system with characteristics of glycosylation modification similar to those of mammalian cells.The candidate vaccine effectively stimulated mice to produce high-titer anti-RBD specific antibody.Furthermore,the specific antibody titer and virus-neutralizing antibody(NAb)titer induced by the vaccine were increased significantly by the combination of the double adjuvants Al(OH)_(3) and CpG.our results showed that the virus-NAb lasted for more than six months in mice.To summarize,we have obtained a SARS-CoV-2 vaccine based on the RBD of the S glycoprotein expressed in glycoengineered Pichia pastoris,which stimulates neutralizing and protective antibody responses.A technical route for fucose-free complex-type N-glycosylation modified recombinant subunit vaccine preparation has been established.

On Chevalley Restriction Theorem for Semi-reductive Algebraic Groups and Its Applications

An algebraic group is called semi-reductive if it is a semi-direct product of a reductive subgroup and the unipotent radical.Such a semi-reductive algebraic group naturally arises and also plays a key role in the study of modular representations of non-classical finite-dimensional simple Lie algebras in positive characteristic,and some other cases.Let G be a connected semi-reductive algebraic group over an algebraically closed field F and g=Lie(G).It turns out that G has many same properties as reductive groups,such as the Bruhat decomposition.In this note,we obtain an analogue of classical Chevalley restriction theorem for g,which says that the G-invariant ring F[g]~G is a polynomial ring if g satisfies a certain“positivity”condition suited for lots of cases we are interested in.As applications,we further investigate the nilpotent cones and resolutions of singularities for semi-reductive Lie algebras.

A soy glycinin derived octapeptide protects against MCD diet induced non-alcoholic fatty liver disease in mice

Soy glycinin derived octapeptide(SGP8)is a peptide obtained from degradation of the soy glycinin,whose amino acid sequence is IAVPGEVA.To determine the effect of SGP8 on non-alcoholic fatty liver disease(NAFLD),steatosis Hep G2 cells were induced by 1 mmol/L free fatty acid(FFA)and C57 BL/6 J mice were fed with methionine-choline defi cient(MCD)diet for 3 weeks to establish NAFLD model.The results of oil red O staining and total cholesterol(TC)/triglyceride(TG)contents showed that SGP8 could signifi cantly reduce the lipid content of steatosis Hep G2 cells.In vivo,SGP8 lowered plasma alanine aminotransferase(ALT)and low density lipoprotein(LDL)content,normalized hepatic superoxide dismutase(SOD)and malondialdehyde(MDA)production,and reduced the severity of liver infl ammation.The results of Western blotting showed that SGP8 increased expression of Sirtuin-1(SIRT1)and phosphorylation level of AMP activated protein kinase(AMPK)in hepatocytes.Through activation of SIRT1/AMPK pathway,SGP8 downregulated the expression of sterol regulatory element binding protein 1 c(SREBP-1 c)and its target genes ACC and FAS expression levels,and increased the phosphorylation level of acetyl Co A carboxylase(ACC).Furthermore,SGP8 also upregulated the expression of transcription factor peroxisome proliferator activated receptorα(PPARα),which was regulated by SIRT1/AMPK pathway,and its target gene CPT1 level.In conclusion,SGP8 might improve NAFLD by activating the SIRT1/AMPK pathway.our data suggest that SGP8 may act as a novel and potent therapeutic agent against NAFLD.

本文设计、制备了一种基于硅基光电子技术的超紧凑混合表面等离激元光场窄化器件,并验证了器件的纳米聚焦性能.实验结果表明,该光场窄化器件利用了长度约为1.23μm的锥形渐变结构,将硅条形波导中的光场聚焦到硅基混合表面等离激元波导中,在1550 nm的近红外波段最高可实现约20倍的非谐振光场增强效应.结构简单,性能优异的硅基混合表面等离激元光场窄化器件,在光场操控、光传感、非线性光学器件、光相变存储等领域中具有潜在的应用价值.

肝缺血再灌注损伤(hepatic ischemia reperfusion injure,HIRI)是在肝移植、肝切除等外科手术中的常见并发症,是引起肝损伤的一个重要原因,可能会导致肝功能衰竭等。目前HIRI相关信号通路与防治措施的实验性研究虽然较多,但临床疗效仍存在争议,因此HIRI的防治依然是巨大挑战。HIRI的相关信号通路作用机制不完全明确,且不断有新的信号通路被发现。笔者从损伤性作用、保护性作用、双向调节、相互串扰四个方面对将HIRI中的信号通路进行归纳阐述,为今后的临床治疗提供新思路。

Laplacian Speech Model and Soft Decision Based MMSE Estimator for Noise Power Spectral Density in Speech Enhancement

The estimation of noise Power spectral density(PSD) is a very crucial issue for speech enhancement as a result of its significant effect on the quality and intelligibility of the enhanced speech. Most of the existing estimators for noise PSD try to employ Gaussian speech priors, which, however, have been proven inconsistent with the reality. We derived an effective solution to this problem of estimating noise PSD in the Minimum mean square error(MMSE) sense when the speech component is modeled by a Laplacian distribution. Meanwhile, the soft decision technique instead of the hard Voice activity detection(VAD) is evolved into our algorithm, which can automatically makes the estimation unbiased without requiring a bias compensation. The performance of the proposed method is tested by several objective and subjective measures under various stationary and nonstationary noise environments. The results confirm that our method achieves good performance for all the noise conditions and Signalnoise-ratio(SNR) settings.

Osimertinib,a third-generation epidermal growth factor receptor tyrosine kinase inhibitor(EGFR-TKIs),is approved globally as the first-line treatment for patients with EGFR mutations(EGFRm:Ex19del/L858R)and T790M resistance mutations in advanced non-small cell lung cancer(NSCLC).[1−3]With its widespread use,the cardiotoxicity of osimertinib has been of great concern.According to the U.S.Food and Drug Administration Adverse Events Reporting System(FAERS),the main cardiac-related adverse events with EGFR-TKIs are heart failure,QT prolongation,atrial fibrillation,acute myocardial infarction and pericardial effusion,and osimertinib has a higher incidence of QT prolongation,heart failure and atrial fibrillation than other EGFR-TKIs.
