Streamlined human antibody generation and optimization by exploiting designed immunoglobulin loci in a B cell line

Monoclonal antibodies(mAbs)are widely utilized as therapeutic drugs for various diseases,such as cancer,autoimmune diseases,and infectious diseases.Using the avian-derived B cell line DT40,we previously developed an antibody display technology,namely,the ADLib system,which rapidly generates antigen-specific mAbs.Here,we report the development of a human version of the ADLib system and showcase the streamlined generation and optimization of functional human mAbs.Tailored libraries were first constructed by replacing endogenous immunoglobulin genes with designed human counterparts.From these libraries,clones producing full-length human IgGs against distinct antigens can be isolated,as exemplified by the selection of antagonistic mAbs.Taking advantage of avian biology,effective affinity maturation was achieved in a straightforward manner by seamless diversification of the parental clones into secondary libraries followed by single-cell sorting,quickly affording mAbs with improved affinities and functionalities.Collectively,we demonstrate that the human ADLib system could serve as an integrative platform with unique diversity for rapid de novo generation and optimization of therapeutic or diagnostic antibody leads.Furthermore,our results suggest that libraries can be constructed by introducing exogenous genes into DT40 cells,indicating that the ADLib system has the potential to be applied for the rapid and effective directed evolution and optimization of proteins in various fields beyond biomedicine.

With the rapid development of white LEDs,the research of new and efficient white light emitting materials has attracted increasing attention.Zero dimensional(0D)organic–inorganic hybrid metal halide perovskites with superior luminescent property are promising candidates for LED application,due to their abundant and tailorable structure.Herein,[(CH_(3))_(3)S]_(2)SnCl_(6)・H_(2)O is synthesized as a host for dopant ions Bi^(3+)and Sb^(3+).The Sb^(3+)doped,or Bi^(3+)/Sb^(3+)co-doped,[(CH_(3))3S]_(2)SnCl_(6)・H_(2)O has a tunable optical emission spectrum by means of varying dopant ratio and excitation wavelength.As a result,we can achieve single-phase materials suitable for emission ranging from cold white light to warm white light.The intrinsic mechanism is examined in this work,to clarify the dopant effect on the optical properties.The high stability of title crystalline material,against water,oxygen and heat,makes it promising for further application.

Channel Correlation Based User Grouping Algorithm for Nonlinear Precoding Satellite Communication System

Low Earth Orbit(LEO)multibeam satellites will be widely used in the next generation of satellite communication systems,whose inter-beam interference will inevitably limit the performance of the whole system.Nonlinear precoding such as Tomlinson-Harashima precoding(THP)algorithm has been proved to be a promising technology to solve this problem,which has smaller noise amplification effect compared with linear precoding.However,the similarity of different user channels(defined as channel correlation)will degrade the performance of THP algorithm.In this paper,we qualitatively analyze the inter-beam interference in the whole process of LEO satellite over a specific coverage area,and the impact of channel correlation on Signal-to-Noise Ratio(SNR)of receivers when THP is applied.One user grouping algorithm is proposed based on the analysis of channel correlation,which could decrease the number of users with high channel correlation in each precoding group,thus improve the performance of THP.Furthermore,our algorithm is designed under the premise of co-frequency deployment and orthogonal frequency division multiplexing(OFDM),which leads to more users under severe inter-beam interference compared to the existing research on geostationary orbit satellites broadcasting systems.Simulation results show that the proposed user grouping algorithm possesses higher channel capacity and better bit error rate(BER)performance in high SNR conditions relative to existing works.

Interspecific isolation is a common mechanism adopted by plants to prevent gene flow between diverged species,thus maintaining species genetic integrity.However,from a plant breeder's perspective,hybridization is desirable,as it can provide a wider range of agriculturally important traits for crop improvement(Tonosaki et al.,2016).Although significant progress has been made in recent years in our understanding of some key factors controlling pollen-pistil interactions within species,we still know virtually nothing about the genetic and molecular basis underlying interspecific incompatibility.

BACKGROUND The study on predicting the differentiation grade of colorectal cancer(CRC)based on magnetic resonance imaging(MRI)has not been reported yet.Developing a non-invasive model to predict the differentiation grade of CRC is of great value.AIM To develop and validate machine learning-based models for predicting the differ-entiation grade of CRC based on T2-weighted images(T2WI).METHODS We retrospectively collected the preoperative imaging and clinical data of 315 patients with CRC who underwent surgery from March 2018 to July 2023.Patients were randomly assigned to a training cohort(n=220)or a validation cohort(n=95)at a 7:3 ratio.Lesions were delineated layer by layer on high-resolution T2WI.Least absolute shrinkage and selection operator regression was applied to screen for radiomic features.Radiomics and clinical models were constructed using the multilayer perceptron(MLP)algorithm.These radiomic features and clinically relevant variables(selected based on a significance level of Plinical models.The performance of the three models(clinical,radiomic,and radiomic-clinical model)were evaluated using the area under the curve(AUC),calibration curve and decision curve analysis(DCA).RESULTS After feature selection,eight radiomic features were retained from the initial 1781 features to construct the radiomic model.Eight different classifiers,including logistic regression,support vector machine,k-nearest neighbours,random forest,extreme trees,extreme gradient boosting,light gradient boosting machine,and MLP,were used to construct the model,with MLP demonstrating the best diagnostic performance.The AUC of the radiomic-clinical model was 0.862(95%CI:0.796-0.927)in the training cohort and 0.761(95%CI:0.635-0.887)in the validation cohort.The AUC for the radiomic model was 0.796(95%CI:0.723-0.869)in the training cohort and 0.735(95%CI:0.604-0.866)in the validation cohort.The clinical model achieved an AUC of 0.751(95%CI:0.661-0.842)in the training cohort and

Spatial identification and evaluation of rural vitality from a function-element-flow perspective: Evidence of lin'an district in Hangzhou, China

Rural vitality is the life force expressed by a combination of endogenous dynamics and external influences. Exploring the complex relationship between rural functions, elements and flows could achieve sustainable rural development. This study constructed a theoretical framework guided by the four functions of production, living, ecology and culture with the support of mobile big data. Furthermore, the network centrality of villages was estimated to reflect the intensity of urban-rural social mobility ties. The results indicated marked spatial disparities in rural vitality, and the coupling of ecological-cultural vitality has a high degree of coherence. Four rural vitality grades were identified: high level(38, 14.08%), medium-high level(66, 24.44%), medium-low level(110, 40.74%) and low level(56, 20.74%), covering 270 administrative village units. The flow intensity of social linkage elements is consistent with rural vitality and the socioeconomic spillover effect of urban centers on neighboring villages was noticeable. Topographic complexity negatively affected the living function, mainly in the T1 and T2 terrain gradients;the rural ecological function was not fully correlated with urban adjacency, and proximity could lead to adverse effects such as urban sprawl and resource destruction. The application of this study is to explore the importance of “flow” by utilizing mobile big data to refine the evaluation unit to the village scale. Accelerating the construction of network coverage and information interconnection and promoting the elemental flow of people, transportation and information between urban and rural areas are important ways to enhance rural vitality.

Epilepsy can be defined as a dysfunction of the brain network,and each type of epilepsy involves different brain-network changes that are implicated diffe rently in the control and propagation of interictal or ictal discharges.Gaining more detailed information on brain network alterations can help us to further understand the mechanisms of epilepsy and pave the way for brain network-based precise therapeutic approaches in clinical practice.An increasing number of advanced neuroimaging techniques and electrophysiological techniques such as diffusion tensor imaging-based fiber tra ctography,diffusion kurtosis imaging-based fiber tractography,fiber ball imagingbased tra ctography,electroencephalography,functional magnetic resonance imaging,magnetoencephalography,positron emission tomography,molecular imaging,and functional ultrasound imaging have been extensively used to delineate epileptic networks.In this review,we summarize the relevant neuroimaging and neuroelectrophysiological techniques for assessing structural and functional brain networks in patients with epilepsy,and extensively analyze the imaging mechanisms,advantages,limitations,and clinical application ranges of each technique.A greater focus on emerging advanced technologies,new data analysis software,a combination of multiple techniques,and the construction of personalized virtual epilepsy models can provide a theoretical basis to better understand the brain network mechanisms of epilepsy and make surgical decisions.

BACKGROUND Despite continuous changes in treatment methods,the survival rate for advanced hepatocellular carcinoma(HCC)patients remains low,highlighting the importance of diagnostic methods for HCC.AIM To explore the efficacy of texture analysis based on multi-parametric magnetic resonance(MR)imaging(MRI)in predicting microvascular invasion(MVI)in preoperative HCC.METHODS This study included 105 patients with pathologically confirmed HCC,categorized into MVI-positive and MVI-negative groups.We employed Original Data Analysis,Principal Component Analysis,linear Discriminant Analysis(LDA),and Non-LDA(NDA)for texture analysis using multi-parametric MR images to predict preoperative MVI.The effectiveness of texture analysis was determined using the B11 program of the MaZda4.6 software,with results expressed as the misjudgment rate(MCR).RESULTS Texture analysis using multi-parametric MRI,particularly the MI+PA+F dimensionality reduction method combined with NDA discrimination,demonstrated the most effective prediction of MVI in HCC.Prediction accuracy in the pulse and equilibrium phases was 83.81%.MCRs for the combination of T2-weighted imaging(T2WI),arterial phase,portal venous phase,and equilibrium phase were 22.86%,16.19%,20.95%,and 20.95%,respectively.The area under the curve for predicting MVI positivity was 0.844,with a sensitivity of 77.19%and specificity of 91.67%.CONCLUSION Texture analysis of arterial phase images demonstrated superior predictive efficacy for MVI in HCC compared to T2WI,portal venous,and equilibrium phases.This study provides an objective,non-invasive method for preoperative prediction of MVI,offering a theoretical foundation for the selection of clinical therapy.

Mitochondria-targeted nanoparticles overcome chemoresistance via downregulating BACH1/CD47 axis in ovarian carcinoma

The platinum-based chemotherapy is a routine strategy for the treatment of ovarian cancer,while it is prone to chemoresistance in clinical,which hinders the treatment.Therefore,it is urgently needed to elucidate the underlying mechanism of drug resistance and form the appropriate strategy.The sequencing results showed that cisplatin(DDP)resistant ovarian cancer overexpressed BTB and CNC homology 1(BACH1),and up-regulated the“don’t eat me”signal CD47.We identified that hemin,a BACH1 inhibitor,could effectively down-regulate BACH1 and simultaneously inhibit CD47.Moreover,hemin has a synergistic effect with DDP.We designed a pH-responsive nanoparticle(H/D@FA-CaP-NPs)in which folic acid(FA)ensured targeting of ovarian cancer cells,while hemin inhibited BACH1 as well as down-regulated CD47,achieving the promotion of apoptosis of tumor cells and inducing phagocytosis of tumors by macrophages.Moreover,hemin has a synergistic effect with DDP to promote apoptosis of tumor cells.Structurally,hemin and DDP was encapsulated within hydrophobic 1,2-distearoyl-sn-glycero-3-phosphoethanolamine(DSPE)to form a tight core,and hydrophilic polyethylene glycol 2000(PEG2000)and calcium phosphate(CaP)formed the outside shell,and FA was modified on the surface of nanoparticles.In terms of function,(a)FA enhanced the active targeting of nanoparticles to tumors;(b)NPs targeted mitochondria to induce reactive oxygen species(ROS)production;(c)hemin encapsulated in nanoparticles could specifically target BACH1,thereby down regulating CD47;(d)hemin had a synergistic effect with DDP,thus augmenting the chemotherapy.Altogether,mitochondria-targeted nanoparticles H/D@FA-CaP-NPs promoted tumor apoptosis and mobilized phagocytosis to treat tumor,providing a novel scheme for clinical treatment of cisplatin-resistant ovarian carcinoma.

Machine Learning Security Defense Algorithms Based on Metadata Correlation Features

With the popularization of the Internet and the development of technology,cyber threats are increasing day by day.Threats such as malware,hacking,and data breaches have had a serious impact on cybersecurity.The network security environment in the era of big data presents the characteristics of large amounts of data,high diversity,and high real-time requirements.Traditional security defense methods and tools have been unable to cope with the complex and changing network security threats.This paper proposes a machine-learning security defense algorithm based on metadata association features.Emphasize control over unauthorized users through privacy,integrity,and availability.The user model is established and the mapping between the user model and the metadata of the data source is generated.By analyzing the user model and its corresponding mapping relationship,the query of the user model can be decomposed into the query of various heterogeneous data sources,and the integration of heterogeneous data sources based on the metadata association characteristics can be realized.Define and classify customer information,automatically identify and perceive sensitive data,build a behavior audit and analysis platform,analyze user behavior trajectories,and complete the construction of a machine learning customer information security defense system.The experimental results show that when the data volume is 5×103 bit,the data storage integrity of the proposed method is 92%.The data accuracy is 98%,and the success rate of data intrusion is only 2.6%.It can be concluded that the data storage method in this paper is safe,the data accuracy is always at a high level,and the data disaster recovery performance is good.This method can effectively resist data intrusion and has high air traffic control security.It can not only detect all viruses in user data storage,but also realize integrated virus processing,and further optimize the security defense effect of user big data.
