Single-cell transcriptome analysis reveals widespread monoallelic gene expression in individual rice mesophyll cells

Monoallelic gene expression refers to the phenomenon that all transcripts of a gene in a cell are expressed from only one of the two alleles in a diploid organism. Although monoallelic gene expression has been occasionally reported with bulk transcriptome analysis in plants, how prevalent it is in individual plant cells remains unknown. Here, we developed a single-cell RNA-seq protocol in rice and investigated allelic expression patterns in mesophyll cells of indica （93-11 ） and japonica （Nipponbare） inbred lines, as well as their F1 reciprocal hybrids. We observed pervasive monoallelic gene expression in individual mesophyll cells, which could be largely explained by stochastic and independent transcription of two alleles. By con- trast, two mechanisms that were proposed previously based on bulk transcriptome analyses, parent-of- origin effects and allelic repression, were not well supported by our data. Furthermore, monoallelically expressed genes exhibited a number of characteristics, such as lower expression levels, narrower H3K4me3/H3K9acJH3K27me3 peaks, and larger expression divergences between 93-11 and Nipponbare. Taken together, the development of a single-cell RNA-seq protocol in this study offers us an excellent opportunity to investigate the origins and prevalence of monoallelic gene expression in plant cells.

We present the design of a dual-band left-handed metamaterial with fishnet structure in the terahertz regime. Its left-handed properties are described by the retrieved effective electromagnetic parameters. We introduce an equivalent circuit which offers a theoretical explanation for the left-handed behavior of the dual-band fishnet metamaterial, and investigate its losses receiving- higher figure of mcrit, The design is beneficial to the development of frequency agile and broadband THz materials and devices＇. The dual-band fishnet metamaterial can be extended to infrared and optical frequency ranges by regulating the structural parameters.

The ultra-low permeability reservoir is regarded as an important energy source for oil and gas resource development and is attracting more and more *** this work,the active silica nanofuids were prepared by modifed active silica nanoparticles and surfactant *** dispersion stability tests showed that the hydraulic radius of nanofuids was 58.59 nm and the zeta potential was−48.39 *** active nanofuids can simultaneously regulate liquid-liquid interface and solid-liquid *** nanofuids can reduce the oil/water interfacial tension(IFT)from 23.5 to 6.7 mN/m,and the oil/water/solid contact angle was altered from 42°to 145°.The spontaneous imbibition tests showed that the oil recovery of 0.1 wt%active nanofuids was 20.5%and 8.5%higher than that of 3 wt%NaCl solution and 0.1 wt%BSSB-12 ***,the efects of nanofuids on dynamic contact angle,dynamic interfacial tension and moduli were studied from the adsorption behavior of nanofuids at solid-liquid and liquid-liquid *** oil detaching and transporting are completed by synergistic efect of wettability alteration and interfacial tension *** fndings of this study can help in better understanding of active nanofuids for EOR in ultra-low permeability reservoirs.

The effective removal and selective detection of explosive and toxic pollutant trinitrophenol (TNP) is an attractive but challenging ***,a double-cavity nor-seco-cucurbit[10]uril (ns-Q[10])-based supramolecular assembly 8-HQ@ns-Q[10]was fabricated and its structure was characterized by X-ray single crystal *** this assembly,the stoichiometric ratio of ns-Q[10]and 8-hydroxyquinoline (8-HQ) is 1:2,which is also attributed to the special double-cavity structure of ns-Q[10].The luminescence sensing experiments showed that 8-HQ@ns-Q[10]can be used as a good fluorescence-enhanced sensing material (enhanced 27-fold) for the rapid detection of explosives and the aqueous contaminant TNP,with a limit of detection (LOD) of 2.07×10^(-5)mol/L,without interference from other phenolic ***,TNP can be efficiently removed in the presence of assembly 8-HQ@ns-Q[10],and the removal efficiency is more than 89%.Therefore,the supramolecular assembly 8-HQ@ns-Q[10],as a fluorescenceenhanced luminescence sensor and adsorption material,has rich research value and potential application prospect when applied to the detection and removal of TNP in aqueous environment.

The effect of sodium pyrophosphate （SPH） on the separation of chalcopyrite from galena was examined through flotation, adsorption, electrokinetic studies and infrared spectral analysis. Differential flotation tests indicate that satisfactory separation can be achieved within the pH range from 2.5 to 6 using SPH to depress the galena, but not the chalcopyrite when O-isopropyl-N-ethyl thionocarbamate （IPETC） is used as the collector. The electrophoretic mohilities of both the minerals dramatically become negatively charged following SPH adsorption in the pH range from 2.5 to 12, The infrared spectral analysis suggests that chemical adsorption occurs on galena surface treated by SPH, indicating that a chelate complex has formed. At weakly acidic pH values, the adsorption density of IPETC onto galena is significantly reduced in the presence of SPH. However, the amount of IPETC adsorbed onto chalcopyrite almost remains at the same level. Since the observed adsorption density of IEPTC onto chalcopyrite is quite high compared to galena, the observed flotation results are explained. A possible mechanism for the interaction between the two sulphide minerals and SPH is discussed.
