Tumor vaccines are a promising avenue in cancer *** the progress in targeting specific immune epitopes,tumor cells lacking these epitopes can evade the ***,we aimed to construct an efficient in situ tumor vaccine called Vac-SM,utilizing shikonin(SKN)to induce immunogenic cell death(ICD)and Mycobacterium smegmatis as an immune adjuvant to enhance in situ tumor vaccine *** showed a dose-dependent and time-dependent cytotoxic effect on the tumor cell line and induced ICD in tumor cells as evidenced by the CCK-8 assay and the detection of the expression of relevant indicators,*** with the control group,the in situ Vac-SM injection in mouse subcutaneous metastatic tumors significantly inhibited tumor growth and distant tumor metastasis,while also improving survival *** smegmatis effectively induced maturation and activation of bone marrow-derived dendritic cells(DCs),and in vivo tumor-draining lymph nodes showed an increased maturation of DCs and a higher proportion of effector memory T-cell subsets with the Vac-SM treatment,based on flow cytometry analysis ***,the Vac-SM vaccine effectively induces ICD,improves antigen presentation by DCs,activates a specific systemic antitumor T-cell immune response,exhibits a favorable safety profile,and holds the promise for clinical translation for local tumor immunotherapy.

Polydatin ameliorates hepatic ischemia-reperfusion injury by modulating macrophage polarization

Association between glaucoma and stroke:A bidirectional mendelian randomization study

Purpose:Observational studies have reported positive associations between glaucoma and stroke;however,controversial results ***,the nature of the relationship remains unknown since previous studies were not designed to test ***,we aimed to investigate the possible causal relationships between glaucoma and ***:Our two-sample Mendelian randomization(MR)encompassed multi-ethnic large-scale genome-wide association studies with more than 2.000 cases and 2.0000 controls for glaucoma,and more than 80000 cases and 630000 controls for *** effect estimates for each SNP were combined using the inverse-variance weighted(IVW)*** avoid potential pleiotropic effects,we adjusted the main results by excluding genetic variants associated with metabolic *** weighted median and MR-Egger methods were also used for the sensitivity ***:Our MR analysis revealed that glaucoma and its subtypes,including primary open-angle glaucoma and primary angle-closure glaucoma,exhibited no causal role in relation to any stroke(AS),any ischemic stroke(AIS),large-artery atherosclerotic stroke(LAS),small-vessel stroke(SVS),or cardioembolic stroke(CES)across MR analyses(all P>0.05).The null associations remained robust even after adjusting for metabolic-related traits and were consistent in both the European and Asian ***,reverse MR analyses also did not indicate any significant causal effects of AS,AIS,LAS,or CES on glaucoma ***:Evidence from our series of causal inference approaches using large-scale population-based MR an-alyses did not support causal effects between glaucoma and *** findings suggest that the relationship of glaucoma management and stroke risk prevention should be carefully evaluated in future *** turn,stroke diagnosis should not be simply applied to glaucoma risk prediction.

Increasing root-lower characteristics improves drought tolerance in cotton cultivars at the seedling stage

Drought is an important abiotic stress factor in cotton *** root system architecture(RSA)of cotton shows high plasticity which can alleviate drought-related stress under drought stress(DS)conditions;however,this alleviation is cultivar ***,this study estimated the genetic variability of RSA in cotton under *** the paper-based growth system,we assessed the RSA variability in 80 cotton cultivars at the seedling stage,with 0 and10%polyethylene glycol 6000(PEG6000)as the control(CK)and DS treatment,*** analysis of 2. aboveground and root traits in the 80 cotton cultivars revealed different responses to *** the 10th day after DS treatment,the degree of variation in the RSA traits under DS(5–55%)was greater than that of CK(5–49%).The 80 cultivars were divided into drought-tolerant cultivars(group 1),intermediate drought-tolerant cultivars(group 2.,and drought-sensitive cultivars(group 3)based on their comprehensive evaluation values of drought *** DS,the root lengthlower,root area-lower,root volume-lower,and root length density-lower were significantly reduced by 63,71,76,and 4%in the drought-sensitive cultivars compared to ***,the drought-tolerant cultivars maintained their root lengthlower,root area-lower,root volume-lower,and root length density–lower *** to CK,the root diameter(0–2.mm)-lower increased by 2.%in group 1 but decreased by 3 and 64%in groups 2.and 3,respectively,under ***,the drought-tolerant cultivars displayed a plastic response under DS that was characterized by an increase in the root-lower *** resistance was positively correlated with the root area-lower and root length ***,the RSA of the different cotton cultivars varied greatly under ***,important root traits,such as the root-lower traits,provide great insights for exploring whether drought-tolerant cotton cultivars can effectively withstand adverse environment

TransCell:In Silico Characterization of Genomic Landscape and Cellular Responses by Deep Transfer Learning

Gene expression profiling of new or modified cell lines becomes routine today;however,obtaining comprehensive molecular characterization and cellular responses for a variety of cell lines,including those derived from underrepresented groups,is not trivial when resources are *** gene expression to predict other measurements has been actively explored;however,systematic investigation of its predictive power in various measurements has not been well ***,we evaluated commonly used machine learning methods and presented TransCell,a two-step deep transfer learning framework that utilized the knowledge derived from pan-cancer tumor samples to predict molecular features and *** these models,TransCell had the best performance in predicting metabolite,gene effect score(or genetic dependency),and drug sensitivity,and had comparable performance in predicting mutation,copy number variation,and protein ***,TransCell improved the performance by over 50%in drug sensitivity prediction and achieved a correlation of 0.7 in gene effect score ***,predicted drug sensitivities revealed potential repurposing candidates for new 100 pediatric cancer cell lines,and predicted gene effect scores reflected BRAF resistance in melanoma cell ***,we investigated the predictive power of gene expression in six molecular measurement types and developed a web portal(http://***/transcell/)that enables the prediction of 352.000 genomic and cellular response features solely from gene expression profiles.

Lead sulfide(PbS)colloidal quantum dot(CQD)photodiodes integrated with silicon-based readout integrated circuits(ROICs)offer a promising solution for the next-generation short-wave infrared(SWIR)imaging *** their potential,large-size CQD photodiodes pose a challenge due to high dark currents resulting from surface states on nonpassivated(100)facets and trap states generated by CQD *** this work,we present a novel approach to address this issue by introducing double-ended ligands that supplementally passivate(100)facets of halidecapped large-size CQDs,leading to suppressed bandtail states and reduced defect *** results demonstrate that the dark current density is highly suppressed by about an order of magnitude to 9.6 nA cm^(2. at -10 mV,which is among the lowest reported for PbS CQD ***,the performance of the photodiodes is exemplary,yielding an external quantum efficiency of 50.8%(which corresponds to a responsivity of 0.532.A W^(-1))and a specific detectivity of 2.5×10^(12. Jones at 1300 *** integrating CQD photodiodes with CMOS ROICs,the CQD imager provides high-resolution(640×512.SWIR imaging for infrared penetration and material discrimination.

TPpred-SC: multi-functional therapeutic peptide prediction based on multi-label supervised contrastive learning

Therapeutic peptides contribute significantly to human health and have the potential for personalized medicine. The prediction for the therapeutic peptides is beneficial and emerging for the discovery of drugs. Although several computational approaches have emerged to discern the functions of therapeutic peptides, predicting multi-functional therapeutic peptide types is challenging. In this research, a novel approach termed TPpred-SC has been introduced. This method leverages a pretrained protein language model alongside multi-label supervised contrastive learning to predict multi-functional therapeutic *** framework incorporates sequential semantic information directly from large-scale protein sequences in TAPE. Then, TPpred-SC exploits multi-label supervised contrastive learning to enhance the representation of peptide sequences for imbalanced multi-label therapeutic peptide prediction. The experimental findings demonstrate that TPpred-SC achieves superior performance compared to existing related methods. To serve our work more efficiently, the web server of TPpred-SC can be accessed at http://***/TPpred-SC.
