The human microbiota-associated(HMA)mice model,especially the germ-free(GF)-humanized mice,has been widely used to probe the causal relationships between gut microbiota and human diseases such as type 1 diabetes(T1D).However,most studies have not clarified the extent to which the reconstruction of the human donor microbiota in recipient mice correlates with corresponding phenotypic *** this study,we transplanted fecal microbiota from five patients with T1D and four healthy people into GF mice,and microbiota from each donor were transplanted into 10 *** with similar microbiota structure to the donor exhibited better phenotypic *** characteristics of the microbial community assembly of donors also influenced the phenotypic reproducibility in mice,and individuals with a higher proportion of stochastic processes showed more severe *** enriched in patients with T1D had a stronger colonization potential in mice with impaired glucose metabolism,and microbiota functional features related to T1D were better reproduced in these *** indicates that assembly traits and colonization efficacy of microbiota influence phenotypic reproducibility in GF-humanized *** findings provide important insights for using HMA mice models to explore links between gut microbiota and human diseases.

Based on observed meteorological elements,photolysis rates(J-values)and pollutant concentrations,an automated J-values predicting system by machine learning(J-ML)has been developed to reproduce and predict the J-values of O^(1)D,NO_(2),HONO,H_(2)O_(2),HCHO,and NO_(3),which are the crucial values for the prediction of the atmospheric oxidation capacity(AOC)and secondary pollutant concentrations such as ozone(O_(3)),secondary organic aerosols(SOA).The J-ML can self-select the optimal“Model+Hyperparameters”without human *** evaluated results showed that the J-ML had a good performance to reproduce the J-values wheremost of the correlation(R)coefficients exceed 0.93 and the accuracy(P)values are in the range of 0.68-0.83,comparing with the J-values from observations and from the tropospheric ultraviolet and visible(TUV)radiation model in Beijing,Chengdu,Guangzhou and Shanghai,*** hourly prediction was also well performed with R from 0.78 to 0.81 for next 3-days and from 0.69 to 0.71 for next 7-days,*** with O_(3)concentrations by using J-values from the TUV model,an emission-driven observation-based model(e-OBM)by using the J-values from the J-ML showed a 4%-12%increase in R and 4%-30%decrease in ME,indicating that the J-ML could be used as an excellent supplement to traditional numerical *** feature importance analysis concluded that the key influential parameter was the surface solar downwards radiation for all J-values,and the other dominant factors for all J-values were 2-m mean temperature,O_(3),total cloud cover,boundary layer height,relative humidity and surface pressure.

It is of great significance to accurately and rapidly identify shale lithofacies in relation to the evaluation and prediction of sweet spots for shale oil and gas reservoirs. To address the problem of low resolution in logging curves, this study establishes a grayscale-phase model based on high-resolution grayscale curves using clustering analysis algorithms for shale lithofacies identification, working with the Shahejie Formation, Bohai Bay Basin, China. The grayscale phase is defined as the sum of absolute grayscale and relative amplitude as well as their features. The absolute grayscale is the absolute magnitude of the gray values and is utilized for evaluating the material composition(mineral composition + total organic carbon) of shale, while the relative amplitude is the difference between adjacent gray values and is used to identify the shale structure type. The research results show that the grayscale phase model can identify shale lithofacies well, and the accuracy and applicability of this model were verified by the fitting relationship between absolute grayscale and shale mineral composition, as well as corresponding relationships between relative amplitudes and laminae development in shales. Four lithofacies are identified in the target layer of the study area: massive mixed shale, laminated mixed shale, massive calcareous shale and laminated calcareous shale. This method can not only effectively characterize the material composition of shale, but also numerically characterize the development degree of shale laminae, and solve the problem that difficult to identify millimeter-scale laminae based on logging curves, which can provide technical support for shale lithofacies identification, sweet spot evaluation and prediction of complex continental lacustrine basins.

Cardiac arrest can lead to severe neurological impairment as a result of inflammation,mitochondrial dysfunction,and post-cardiopulmonary resuscitation neurological *** preconditioning has been shown to improve migration and survival of bone marrow–derived mesenchymal stem cells and reduce pyroptosis after cardiac arrest,but the specific mechanisms by which hypoxia-preconditioned bone marrow–derived mesenchymal stem cells protect against brain injury after cardiac arrest are *** this end,we established an in vitro co-culture model of bone marrow–derived mesenchymal stem cells and oxygen–glucose deprived primary neurons and found that hypoxic preconditioning enhanced the protective effect of bone marrow stromal stem cells against neuronal pyroptosis,possibly through inhibition of the MAPK and nuclear factor κB ***,we transplanted hypoxia-preconditioned bone marrow–derived mesenchymal stem cells into the lateral ventricle after the return of spontaneous circulation in an 8-minute cardiac arrest rat model induced by *** results showed that hypoxia-preconditioned bone marrow–derived mesenchymal stem cells significantly reduced cardiac arrest–induced neuronal pyroptosis,oxidative stress,and mitochondrial damage,whereas knockdown of the liver isoform of phosphofructokinase in bone marrow–derived mesenchymal stem cells inhibited these *** conclude,hypoxia-preconditioned bone marrow–derived mesenchymal stem cells offer a promising therapeutic approach for neuronal injury following cardiac arrest,and their beneficial effects are potentially associated with increased expression of the liver isoform of phosphofructokinase following hypoxic preconditioning.

This study describes the use of the weighted multiplicative algebraic reconstruction technique(WMART)to obtain vertical ozone profiles from limb observations performed by the scanning imaging absorption spectrometer for atmospheric chartography(SCIAMACHY).This technique is based on SaskMART(the combination of the multiplicative algebraic reconstruction technique and SaskTRAN radiative transfer model),which was originally developed for optical spectrometer and infrared imaging system(OSIRIS)*** of the objectives of this study was to obtain consistent ozone profiles from the two *** this study,the WMART algorithm is combined with a radiative transfer model(SCIATRAN),as well as a set of measurement vectors comprising five Hartley pairing vectors(HPVs)and one Chappuis triplet vector(CTV),to retrieve ozone profiles in the altitude range of 10–69 *** that the weighting factors in WMART have a significant effect on the retrievals,we propose a novel approach to calculate the pair/triplet weighting factors using wavelength weighting *** results of the application of the proposed ozone retrieval scheme are compared with the SCIAMACHY v3.5 ozone product by University of Bremen and validated against profiles derived from other passive satellite observations or measured by *** 18 and 55 km,the retrieved ozone profiles typically agree with data from the SCIAMACHY ozone product within 5%for tropics and middle latitudes,whereas a negative deviation exists between 35 and 50 km for northern high latitudes,with a deviation of less than 10%above 50 *** of the retrieved profiles with microwave limb sounder(MLS)v5.0 indicates that the difference is within±5%between 18 and 55 km,and an agreement within 10%is achieved in other altitudes for tropics and middle *** of the retrieved profiles with OSIRIS v7.1 indicates that the average deviation is within±5%between 20 and 59 km,and difference of approximately 10%is

It is of great significance to find safe and effective radiosensitizers. A primary investigation has been made on fisetin's modification of radiation effect, but its radiosensitization and related mechanisms still need to be deeply clarified. Furthermore, fisetin with high hydrophobicity is difficult to dissolve in water,severely limiting its research and application. In this study, we fabricated a safe and soluble radiosensitizer fisetin micelle for precisely enhancing radiotherapy by inhibiting platelet-derived growth factor receptor-β(PDGFRβ)/signal transducer and activator of transcription 1(STAT1)/signal transducer and activator of transcription 3(STAT3)/B cell lymphoma 2(Bcl-2) signaling pathway in the tumor. Systematic and detailed studies were performed to verify its radiosensitization effect in vitro and in vivo. On the cellular level, fisetin micelles selectively increased the radiosensitivity of tumor cells(CT26 and 4T1 cells)and had little effect on the sensitivity of normal mouse cells(L929 cells) to radiation. In the mouse models of colon and breast cancers, fisetin micelles showed an efficient radiosensitization capacity without apparent toxicity. Additionally, we first found that fisetin micelles played a radiotherapy sensitization role by inhibiting the PDGFRβ/STAT1/STAT3/Bcl-2 pathway activity. In general, this work not only confirmed that fisetin micelles precisely exhibit a radiosensitization effect in vitro and in vivo, but also profoundly explored its mechanisms underlying, to provide a theoretical and experimental basis for the clinical application of fisetin micelles.
