Single-molecule localization microscopy(SMLM)enables three-dimensional(3D)investigation of nanoscale structures in biological samples,offering unique insights into their organization.However,traditional 3D super-resolution microscopy using high numerical aperture(NA)objectives is limited by imaging depth of field(DOF),restricting their practical application to relatively thin biological samples.Here,we developed a unified solution for thick sample super-resolution imaging using a deformable mirror(DM)which served for fast remote focusing,optimized point spread function(PSF)engineering,and accurate aberration correction.By effectively correcting the system aberrations introduced during remote focusing and sample aberrations at different imaging depths,we achieved high-accuracy,large DOF imaging(~8μm)of the whole-cell organelles[i.e.,nuclear pore complex(NPC),microtubules,and mitochondria]with a nearly uniform resolution of approximately 35 nm across the entire cellular volume.

The ability to discriminate species and recognize individuals is crucial for reproductive success and/or survival in most animals.However,the temporal order and neural localization of these decision-making processes has remained unclear.In this study,event-related potentials(ERPs)were measured in the telencephalon,diencephalon,and mesencephalon of the music frog Nidirana daunchina.These ERPs were elicited by calls from 1 group of heterospecifics(recorded from a sympatric anuran species)and 2 groups of conspecifics that differed in their fundamental frequencies.In terms of the polarity and position within the ERP waveform,auditory ERPs generally consist of 4 main components that link to selective attention(N1),stimulus evaluation(P2),identification(N2),and classification(P3).These occur around 100,200,250,and 300 ms after stimulus onset,respectively.Our results show that the N1 amplitudes differed significantly between the heterospecific and conspecific calls,but not between the 2 groups of conspecific calls that differed in fundamental frequency.On the other hand,the N2 amplitudes were significantly different between the 2 groups of conspecific calls,suggesting that the music frogs discriminated the species first,followed by individual identification,since N1 and N2 relate to selective attention and stimuli identification,respectively.Moreover,the P2 amplitudes evoked in females were significantly greater than those in males,indicating the existence of sexual dimorphism in auditory discrimination.In addition,both the N1 amplitudes in the left diencephalon and the P2 amplitudes in the left telencephalon were greater than in other brain areas,suggesting left hemispheric dominance in auditory perception.Taken together,our results support the hypothesis that species discrimination and identification of individual characteristics are accomplished sequentially,and that auditory perception exhibits differences between sexes and in spatial dominance.

BACKGROUND Gastric cancer(GC)is associated with high mortality rates.Bile acids(BAs)reflux is a well-known risk factor for GC,but the specific mechanism remains unclear.During GC development in both humans and animals,BAs serve as signaling molecules that induce metabolic reprogramming.This confers additional cancer phenotypes,including ferroptosis sensitivity.Ferroptosis is a novel mode of cell death characterized by lipid peroxidation that contributes universally to malignant progression.However,it is not fully defined if BAs can influence GC progression by modulating ferroptosis.AIM To reveal the mechanism of BAs regulation in ferroptosis of GC cells.METHODS In this study,we treated GC cells with various stimuli and evaluated the effect of BAs on the sensitivity to ferroptosis.We used gain and loss of function assays to examine the impacts of farnesoid X receptor(FXR)and BTB and CNC homology 1(BACH1)overexpression and knockdown to obtain further insights into the molecular mechanism involved.RESULTS Our data suggested that BAs could reverse erastin-induced ferroptosis in GC cells.This effect correlated with increased glutathione(GSH)concentrations,a reduced GSH to oxidized GSH ratio,and higher GSH peroxidase 4(GPX4)expression levels.Subsequently,we confirmed that BAs exerted these effects by activating FXR,which markedly increased the expression of GSH synthetase and GPX4.Notably,BACH1 was detected as an essential intermediate molecule in the promotion of GSH synthesis by BAs and FXR.Finally,our results suggested that FXR could significantly promote GC cell proliferation,which may be closely related to its anti-ferroptosis effect.CONCLUSION This study revealed for the first time that BAs could inhibit ferroptosis sensitivity through the FXR-BACH1-GSHGPX4 axis in GC cells.This work provided new insights into the mechanism associated with BA-mediated promotion of GC and may help identify potential therapeutic targets for GC patients with BAs reflux.

The super elongation complex(SEC)containing positive transcription elongation factor b plays a critical role in regulating transcription elongation.AFF1 and AFF4,two members of the AF4/FMR2 family,act as central scaffold proteins of SEC and are associated with various human diseases.However,their precise roles in transcriptional control remain unclear.Here,we investigate differences in the genomic distribution patterns of AFF1 and AFF4 around transcription start sites(TSSs).AFF1 mainly binds upstream of the TSS,while AFF4 is enriched downstream of the TSS.Notably,disruption of AFF4 results in slow elongation and early termination in a subset of AFF4-bound active genes,whereas AFF1 deletion leads to fast elongation and transcriptional readthrough in the same subset of genes.Additionally,AFF1 knockdown increases AFF4 levels at chromatin,and vice versa.In summary,these findings demonstrate that AFF1 and AFF4 function antagonistically to regulate RNA polymerase Ⅱ transcription.

全国统一大市场建设中跨省流动人口的迅速增长,对建立健全面向流动人口的社会保障制度体系提出现实需求。“属地管理”的社会救助制度如何满足流动人口的救助需求,是实现“共同富裕”和“社会中国”愿景的重要议题。本研究基于实地调研,对流动人口社会救助的已有探索、现实挑战和未来前景展开分析。研究发现,流动人口社会救助的已有探索包括流动人口申请常住地社会救助和在常住地发起低保异地申办两个方面,前者包括面向常住人口的急难社会救助、专项救助和局部地区试点的基本生活救助,后者则包括低保跨省异地核对以及部分省份开放的辖域内的低保异地申办渠道。但是,依然存在地方财政压力以及跨地区政策待遇差异、家庭经济状况核对平台的跨区域联网程度以及低保异地核对的责任认定与分担的现实挑战。拓展流动人口救助政策可以沿着全面放开常住地急难社会救助、向流动人口逐步有条件开放专项救助和推进基本生活救助的跨省核对和异地申办的方向逐步推进。

CONSPECTUS:Heterogeneous catalysts are widely used in a variety of industrial fields,including environmental protection,energy conversion,and chemical production.Their performance during reactions is usually determined by a small fraction of sites at the catalyst surfaces/interfaces,namely,the active sites.Actually,since the concept of the“active sites”was proposed by Hugh Taylor in the 1920s,determining the active site at the atomic level and understanding the molecular processes that happened at the active site have become the top priority in catalysis research.Researchers tried different methods to acquire various information related to the surface/interface active sites,pursuing their rational design at the atomic level.Although great achievements have been made in catalyst surface study,in situ atomistic design of active sites remains challenging,due to the lack of direct information and effective manipulation means concerning the active sites.Specifically,many critical issues regarding the active sites under reaction conditions remain to be solved:(1)precisely identifying the active sites,which is the foundation for understanding the catalytical mechanism and rational design of the catalyst;(2)accurately manipulating the surface/interface active sites at the atomic scale,which is the basis for realizing the desired performance;(3)maintaining these designed active sites operating in a long-term with high efficiency without deactivation,which is extremely important to the practical applications of the catalysts.All these aspects rely on the fundamental understanding of the interactions between different surface/interface configurations of the catalyst and external environments(gas,pressure,temperature,etc.).In this Account,we present the recent progress in our group on the studies of surface/interface active sites via controlled atmosphere transmission electron microscopy(CATEM).We first briefly introduce the advances in CATEM technologies,including the window(closed)approach and aperture(open)approaches.Then

Since the coronavirus disease 2019(COVID-19)began to spread,it remains pandemic worldwide.The European Medicines Agency’s human medicines committee and Food and Drug Administration have only granted a conditional marketing authorization for remdesivir to treat COVID-19.It is essential to apply other valuable treatments.Convalescent plasma(CP),donated by persons who have recovered from COVID-19,is the cellular component of blood that contains specific antibodies.Therefore,to determine the feasibility of CP for COVID-19,the effectiveness and controversy are discussed in depth here.It is suggested that CP plays a certain role in the treatment of COVID-19.As a treatment,it may have its own indications and contraindications,which need to be further discussed.Meanwhile,it is critical to establish a standard procedure for treatment from CP collection,preservation,transport,to transfusion,and conduct some large sample randomized controlled trials to confirm the transfusion dosage,appropriate time,frequency,and actively prevent adverse outcomes that may occur.

针对两个细则考核风功率上报准确率的问题,提出一种可有效降低风-储场站风险考核电量的日前-日内两阶段功率优化上报策略。首先,提出面向两个细则的风-储场站日前-日内功率协同上报框架;然后,以期望考核电量最小化为目标,建立计及储能调节的日前-日内两阶段风-储功率优化上报模型,模型中,日前、日内风功率场景集通过基于预测误差分箱的Cornish-Fisher级数与Cholesky分解法生成;最后,针对模型的多重非线性特征与不同时间尺度的求解效率要求,制定基于小步长线性化迭代的求解算法。算例仿真结果表明,相较其余上报策略与求解算法,该文提出的日前-日内协同上报策略及相应算法有效地提升了上报准确率及求解性能。

Efficient experiment design is of great significance for the validation of simulation model with high nonlinearity and large input space.Excessive validation experiment raises the cost while insufficient test increases the risks of accepting an invalid model.In this paper,an adaptive sequential experiment design method combining global exploration criterion and local exploitation criterion is proposed.The exploration criterion utilizes discrepancy metric to improve the space-filling property of the design points while the exploitation criterion employs the leave one out error to discover informative points.To avoid the clustering of samples in the local region,an adaptive weight updating approach is provided to maintain the balance between exploration and exploitation.Besides,the credibility distribution function characterizing the relationship between the input and result credibility is introduced to support the model validation experiment design.Finally,six benchmark problems and an engineering case are applied to examine the performance of the proposed method.The experiments indicate that the proposed method achieves satisfactory performance for function approximation in accuracy and convergence.
