CD21-independent Infection of Epstein-Barr Virus in Human Signet Ring Gastric Carcinoma Cell Line

To study the mechanism of infection of Epstein-Barr virus (EBV) in gastric carcinoma cells, the Akata and P3HR-1 strains of EBV were used as the test strains of viruses, and the signet ring cell line HSC-39 of gastric carcinoma cells was used as the target cells of infection. The virus-infected cell clones were isolated by limited dilution method. It was found that the EBV-encoded small RNA (EBER) could be detected in the infected cells. The Akata and P3HR-1 EBV infected parental cells and most of clones expressed EBNA1, but not EBNA2. Latent membrane protein (LMP-1) and LMP-2, and the Q promoter (p), but not the Cp/Wp for EBNA gene transcription was active in the infected parental cells as well as all the clones. Uninfected HSC-39 cells did not express CD21, however, Akata but not P3HR-1 EBV-infected clones expressed low level of CD21 mRNA. These results demonstrate that HSC-39 cells are susceptible to both EBV strains and EBV infects HSC-39 cells through the CD21-independent pathway. This study defines a signet ring type of gastric carcinoma cells line as a unique target cells for the study of EBV infection mechanism.

Peritoneal dissemination(PD)is the most common cause of metastasis in gastric cancer(GC).Because there are no standard treatments for PD,it is associated with a poor prognosis.Although clinicians have performed intraperitoneal chemotherapy for GC with PD,the outcome remains unsatisfactory.Therefore,the development of novel treatments and diagnostic tools for PD is expected to improve the prognosis of GC patients with PD.Notably,it is essential to elucidate the molecular mechanisms involved in the development of PD in GC.In this review,the molecular mechanisms of PD(three steps:detachment from the primary tumor,adaptation to the microenvironment of the peritoneal cavity,and attachment to peritoneal mesothelial cells)and new topics in GC are highlighted.

In this study, urban environmental condition using Terra ASTER (advanced spaceborne thermal emission and reflection radiometer) remotely sensed satellite imagery was estimated and compared to statistically derived socioeconomic condition data from Semarang Municipality in Central Java Province: Indonesia. From satellite imagery: UI (urban index) and NDVI (normalized difference vegetation index) were calculated and used that information to construct urban density and vegetation distribution maps of the study area. Urban density and vegetation distribution were combined to estimate urban environmental condition; then compared to socioeconomic conditions. The results revealed that the Semarang Municipality had a higher percentage of developed or urbanized areas (49%) compared to the percentage of area covered by vegetation (42%). In general, southern Semarang had a higher quality urban environment and socioeconomic condition compared to northern Semarang, which had a higher urban density, lower quality urban environment and lower socioeconomic condition. The indicate that socioeconomic condition can be determined using satellite-derived urban and vegetation indices even in places like Semarang Municipality, where the population settlement was sparse and unorganized.

Reaction products of 2,4,6-tris(4-phenyl-phenoxy)-1,3,5-triazine derived from 4-phenylphenol cyanate ester and phenyl glycidyl ether were analyzed. In addition to an isocyanurate compound and an oxazolidone compound which were well known as reaction products of cyanate esters and epoxy resins, compounds with hybrid ring structure of cyanurate/isocyanurate were determined. Gibbs free energies of the compound having hybrid ring structure of cyanurate/isocyanurate with two isocyanurate moiety were found to be lower than that of the compound with cyanurate ring structure through calculations. Calculation data supported the existence of hybrid ring structure of cy-anurate/isocyanurate. It was revealed that isomerization from cyanurate to isocyanurate occurs via hybrid ring structure of cyanurate/isocyanurate in the reaction of aryl cyanurate and epoxy.

对妊娠期瘦素的分泌及其调节机制进行探讨.将100只3月龄雌性金黄仓鼠分成16组,每组6-7只,每天采血样用于测定其妊娠期的瘦素分泌.实验结果表明,瘦素于妊娠第9天开始显著升高,于妊娠第12天达到高峰.为验证不同生殖阶段的血清是否对瘦素分泌有不同影响,将不同生殖阶段抽提与未抽提类固醇激素的血清加入培养液中进行培养,发现动情期及妊娠期的血清均可刺激体外培养的脂肪细胞瘦素分泌,动情期抽提与未抽提类固醇激素的血清对脂肪细胞瘦素分泌的刺激作用没有显著差异(P>0.05).与动情期血清相比,妊娠中后期未抽提类固醇激素的血清显著刺激脂肪细胞瘦素分泌(P0.05).妊娠不同阶段未抽提类固醇激素的血清对瘦素分泌的刺激作用有很大差异,妊娠第15天的血清对瘦素分泌的刺激作用显著高于妊娠第8天(P<0.0001)和妊娠第12天(P<0.001)的血清.体外研究进一步表明,肾上腺皮质素、孕酮以及胰岛素均能刺激瘦素分泌,而雌激素却能抑制体外瘦素分泌.研究结果提示,妊娠期血液类固醇激素水平的上升是引起妊娠期瘦素峰出现的重要因素.

The purpose of this study is to make clear the behavior of swirling pipe flows with non-circular section. We have investigated experimentally and numerically the swirling flows in a rectangular channel with aspect ratio of 1.3, which is one of the typical pipe flow with non-circular section. In this paper, experimental apparatus with water circulation system is produced, and LDV measurement of the swirling flows is performed in order to clarify the mean velocities and turbulent quantity with higher accuracy. In addition, flow visualization for movement of vortex core region in the swirling flows is carried out using tracer method. Moreover, large eddy simulation (LES) of the flow field is made and compared with the experimental results. As results of these investigations, the behavior of the swirling flows in the rectangular channel is totally discussed.

Study of pruritus in chronic hepatitis C patients

AIM:To investigate the occurrence and severity of pruritus in chronic hepatitis C patients treated with or without interferon(IFN)therapy.METHODS:A total of 89 patients with chronic hepatitis C and 55 control(non-hepatitis)patients were asked to rate their experience of diurnal and nocturnal pruritus in the preceding week using a visual analogue scale(VAS)and a five-point scale,respectively.Blood samples were taken and serum thymus and activationregulated chemokine(TARC)levels were measured by enzyme-linked immunosorbent assay.RESULTS:A significantly greater proportion of chronic hepatitis C patients experienced nocturnal pruritus compared with control(58.4%vs 5.5%,P<0.0001).Chronic hepatitis C patients also had more severe pruritus compared with control patients,indicated by the higher mean VAS scores in both the IFN-treated and non-IFN-treated groups.In particular,patients who received combined peginterferon alfa-2b and ribavirin had significantly higher mean VAS scores than those receiving peginterferon alfa-2a or no IFN treatment.SerumTARC levels did not correlate with pruritus scores,and no significant differences in TARC levels were observed between the IFN-treated and non-IFN-treated groups.CONCLUSION:Patients with chronic hepatitis C experience pruritus more than those without.Serum TARC levels do not correlate with pruritus severity in chronic hepatitis C patients.

This study established a method for easily and quickly estimating the specific activity produced in the concrete walls and floors of accelerator rooms during long-term operation of accelerator,for advanced zoning of activated/nonactivated areas in planning the decommissioning of an accelerator.We propose a new,highly sensitive method for nondestructively estimating the specific activity in concrete that can be applied to activation zoning.In this method,instead of direct determination of the specific activities of important long-half-life radionuclides for decommissioning,such as 152 Eu and 60 Co,we determine the specific activities of short-half-life radionuclides,24 Na and 56 Mn,in situ to obtain neutron flux.The obtained neutron flux and accelerator operation history yield the specific activities of 152 Eu and 60 Co for the advance zoning of activated/non-activated concrete.This method is a powerful long-term prediction tool for concrete activation.

Stainability and Fine Structure of Intracytoplasmic Inclusion Bodies in the Locus Ceruleus of Mouse

The present study compared intracytoplasmic inclusion bodies in the locus ceruleus (LC) of mouse with protein bodies in the LC of human. Phosphotungstic acid-hematoxylin (PTAH) and the Mallory method were used as the anionic stains. The inclusion bodies and protein bodies, which stained with PTAH and the Mallory method in the brain, contain proteins that appear to belong to the same family. Although both inclusion and protein bodies were formed with the same arginine composition, their distribution in the brain was similar and similar physiological changes by morbidity were observed, and the fine structure of the inclusion bodies and protein bodies appeared to be different. The present findings suggest that mouse inclusion bodies and human protein bodies are different.
