Impaired suppressive activities of human MUTYH variant proteins against oxidative mutagenesis

Eusocial insects display a caste system in which different castes are morpho-logically and physiologically specialized for different *** studies have revealed that epigenetic modifications,including DNA methylation and histone modification,me-diate caste determination and differentiation,longevity,and polyethism in eusocial *** there has been a growing interest in the relationship between epigenetic mech-anisms and phenotypic plasticity in termites,there is ltte information about differential expression levels among castes and expression sites for these genes in *** we show royaltissuc-specific expression of epigenetic modification genes in the termite Reticulitermes *** RNA-seq,we identified 74 genes,including three DNA methyltransferases,seven sirtuins,48 Trithorax group proteins,and 16 Polycomb group *** these genes,15 showed king-specific expression,and 52 showed age-dependent differential expression in kings and *** real-time PCR revealed that DNA methyltransferase 3 is expressed specifically in the king's testis and fat body,whereas some histone modification genes are remarkably expressed in the king's testis and queen's *** findings imply that epigenetic modification plays important roles in the gamete production process in termite kings and queens.

Development of a validation technique for road surface profile applicable to point cloud data

In recent years,various information and communication technology(ICT)devices measuring three-dimensional(3D)point cloud data have been developed and widely used for the application of pavement surface ***,ICT devices have generally been developed not only for measuring road surface profiles but for various geo-reference point *** this background,the validation of surface profiles acquired with ICT devices fulfils an important role in proving the reliability of measurement result composed by point *** study proposes a wavelet transform agreement(WTA)which involves a normalization factor of profile amplitude for further improvement in the wavelet-based coherence *** WTA analysis allows evaluating similarity/dissimilarity of two profiles considering both the location and wavelength *** this purpose,a terrestrial laser scanner(TLS),a mobile mapping system(MMS),and an unmanned aerial vehicle(UAV)are employed to prove the advantage of WTA in practical *** a result,the advantages of WTA analysis are clearly recognized in the optimization for the measurement interval of TLS,the multi-line measurement of MMS for ride quality improvement of a pavement,and the efficient operation of UAV in terms of the flight *** paper also shows the identification of aging development for surface roughness over time in terms of locations and *** findings help not only to guarantee the accuracy of profile measurements but to realize the sophisticated way of using 3D point clouds acquired with ICT *** outcomes of this study contribute to the increase of productivity for pavement works with improving the quality of surface profile measurement.

Prospective study of differential diagnosis of hepatic tumors by pattern-based classification of contrast-enhanced sonography

AIM: To prospectively evaluate the usefulness of a pattern-based classification of contrast-enhanced sonographic findings for differential diagnosis of hepatic tumors. METHODS: We evaluated the enhancement pattern of the contrast-enhanced sonography images in 586 patients with 586 hepatic lesions, consisting of 383 hepatocellular carcinomas, 89 metastases, and 114 hemangiomas. After injecting a galactose-palmitic acid contrast agent, lesions were scanned by contrast- enhanced harmonic gray-scale sonography in three phases: arterial, portal, and late. The enhancement patterns of the initial 303 lesions were classified retrospectively, and multiple logistic regression analysis was used to identify enhancement patterns that allowed differentiation between hepatic tumors. We then used the pattern-based classification of enhancement we had retrospectively devised to prospectively diagnose 283 liver tumors. RESULTS: Seven enhancement patterns were found to be significant predictors of different hepatic tumors. The presence of homogeneous or heterogeneous enhancement both in the arterial and portal phase was the typical enhancement pattern for hepatocellular carcinoma, while the presence of peritumoral vessels in the arterial phase and ring enhancement or a perfusion defect in the portal phase was the typical enhancement pattern for metastases, and the presence of peripheral nodular enhancement both in the arterial and portal phase was the typical enhancement pattern forhemangioma. The sensitivity, specificity, and accuracy of prospective diagnosis based on the combinations of enhancement patterns, respectively, were 93.2%, 96.2%, and 94.0% for hepatocellular carcinoma, 87.9%, 99.6%, and 98.2% for metastasis, and 95.6%, 94.1%, and 94.3% for hemangioma. CONCLUSION: The pattern-based classification of the contrast-enhanced sonographic findings is useful for differentiating among hepatic tumors.

Diversion colitis and pouchitis: A mini-review

Diversion colitis is characterized by inflammation of the mucosa in the defunctioned segment of the colon after colostomy or ileostomy. Similar to diversion colitis, diversion pouchitis is an inflammatory disorder occurring in the ileal pouch, resulting from the exclusion of the fecal stream and a subsequent lack of nutrients from luminal bacteria. Although the vast majority of patients with surgically-diverted gastrointestinal tracts remain asymptomatic, it has been reported that diversion colitis and pouchitis might occur in almost all patients with diversion. Surgical closure of the stoma, with reestablishment of gut continuity, is the only curative intervention available for patients with diversion disease. Pharmacologic treatments using short-chain fatty acids, mesalamine, or corticosteroids are reportedly effective for those who are not candidates for surgical reestablishment; however, there are no established assessment criteria for determining the severity of diversion colitis, and no management strategies to date. Therefore, in this mini-review, we summarize and review various recently-reported treatments for diversion disease. We are hopeful that the information summarized here will assist physicians who treat patients with diversion colitis and pouchitis, leading to better case management.

Measuring and evaluating of road roughness conditions with a compact road profiler and ArcGIS

With a wide range of requirements throughout the world, high-quality road management is subject to increasing demand from a perspective of customer-oriented levels of service. In recent years, road administrators are requested to create a visual map of a road network to monitor conditions. To fulfill these requirements, this study conducted as follows. Firstly, this paper introduces a new compact road profiler to collect the profile data at ease. Using the international roughness index （IRI） to assess public roads in local cities of Japan＇s Hokkaido prefecture, this study also provides real-time monitoring of pavement roughness conditions. Moreover, this study deals with an effective method for visualizing collected IRI data as an attribute in a geographic information system （GIS） and the database of Japan digital road map （DRM）. Secondly, the authors present the measurement results of IRI at two different cities during different seasons by using GIS to compare the road conditions. According to the results clarified on different statistical characteristics of road profiles, this study recommends that it is necessary to establish pavement management system （PMS） in consideration of road class, network of local city, and evaluation and management of road conditions in winter quantitatively. Finally, the authors measure and evaluate ride quality by assessing differences between the inner wheel path （IWP） and outer wheel path （OWP） of the vehicle into account, using the previously mentioned profilers and the driving simulator, which is called KITDS. Results show that information from both wheel paths contributes to improve current monitoring process regarding pavement surface, and expects to construct a high level of PMS for road administration in the future.

GaN metal-oxide-semiconductor field-effect transistors （MOSFETs） on AIGaN/GaN heterostructure with a recess gate were fabricated and characterized. The device showed good pinch-off characteristics and a maximum field-effect mobility of 145.2 cm2.V-1.s-1. The effects of etching gas of CI2 and SiCI4 were investigated in the gate recess process. SiCI4-etched devices showed higher channel mobility and lower threshold voltage. Atomic force microscope measurement was done to investigate the etching profile with different etching protection mask. Compared with photoresist, SiO2- masked sample showed lower surface roughness and better profile with stepper sidewall and weaker trenching effect resulting in higher channel mobility in the MOSFET.

Prognostic factors associated with mortality in patients with gastric fundal variceal bleeding

AIM To determine the prognostic factors associated with mortality in patients with gastric fundal variceal (GFV) *** In total, 42 patients were endoscopically diagnosed with GFV bleeding from January 2000 to March 2014. We retrospectively reviewed the patients' medical records and assessed their history, etiology of liver cirrhosis, disease conditions, treatment options for GFV bleeding, medications administered before and after onset of GFV bleeding, blood test results(hemoglobin, albumin, and bilirubin concentrations), and imaging results(including computed tomography and abdominal ultrasonography). We also assessed the prognostic factors associated with short-term mortality(up to 90 d) and long-term mortality in all *** Multivariate analysis showed that prophylactic administration of antibiotics was an independent prognostic factor associated with decreases in short-term mortality (OR = 0.08, 95%CI: 0.01-0.52) and longterm mortality (OR = 0.27, 95%CI: 0.08-0.91) in patients with GFV bleeding. In contrast, concurrent hepatocellular carcinoma (HCC) and regular use of proton pump inhibitors (PPI) were independent prognostic factors associated with increases in shortterm mortality(HCC: OR = 15.4, 95%CI: 2.08-114.75; PPI: OR = 12.76, 95%CI: 2.13-76.52) and long-term mortality (HCC: OR = 7.89, 95%CI: 1.98-31.58; PPI: OR = 10.91, 95%CI: 2.86-41.65) in patients with GFV bleeding. The long-term overall survival rate was significantly lower in patients who regularly used PPI than in those who did not use PPI(P = 0.0074).CONCLUSION Administration of antibiotics is associated with decreased short- and long-term mortality, while concurrent HCC and regular PPI administration are associated with increased short- and long-term mortality.

PI-3 kinase pathway can mediate the effect of TGF-β1 in inducing the expression of SHARP-2 in LLC-PK1 cells

We aim to investigate the effect of transforming growth factor （TGF）-β1 on the expression of enhancer of split- and hairy-related protein-2 （SHARP-2） messenger RNA （mRNA） and its signaling pathway. In this study, several cell lines including LLC-PK1 （a porcine kidney tubular epithelial cell line）, MDCK （Madin-Darby canine kidney） and CTLL-2 （cytotoxic T-lymphocyte line） were treated with recombinant human TGF-131, and a series of experiments were carried out, involving Northern blot analysis of total RNA from these cells. Further, several specific chemical inhibitors were applied before TGF-β1 treatment to probe the signaling pathway. The results showed that TGF-β1 can significadtly up-regulate SHARP-2 mRNA expression in the LLC-PK1 cell line. The peak level of induction was found 2 h after TGF-β1 stimulation. While one phospho- inositide 3-kinases （PI-3） kinase inhibitor, LY294002, completely blocked the effect of TGF-131 on SHARP-2 mRNA expression in LLC-PK1 cells at a low concentration, other inhibitors, including PD98059, staurosporine, AG490, wortmannin, okadaic acid and rapamycin, had no effect. The effect of LY294002 was dose-dependent. We conclude that, in LLC-PK1 cells at least, TGF-β1 can effectively induce the SHARP-2 mRNA expression and that the PI-3 kinase pathway can mediate this effect.
