The influence of adiponectin G276T gene polymorphism on changes in total and high molecular weight adiponectin levels by exercise training

Adiponectin is an adipocyte-secreted hormone and regulates the metabolism of lipid and glucose. We examined the influence of adiponectin G276T gene polymorphism on changes in total or high molecular weight adiponectin levels by exercise training. A randomized parallel-design study (n = 53;40 women and 13 men;age range, 32 - 65 years) was conducted. Participants were randomly assigned to the exercise (n = 26) or control (n = 27) group and received exercise training for 70 min 2 times per week for 12 weeks and exercise advice at the baseline, respectively. Blood sample were obtained before and after the intervention. The primary outcomes were changes in total adiponectin and high molecular weight adiponectin levels. At the baseline, the participants with and without 276G allele did not differ with the total and high molecular weight adiponectin levels in the entire study population. No significant difference in the change in the total and high molecular weight adiponectin levels between the subjects with the G276G genotype and 276T allele carriers were found. This study provides evidence that the G276T polymorphism of the adiponectin gene is not associated with the magnitude of the effect of twice-weekly exercise training on the total and high molecular weight adiponectin levels.

Characterization of a novel variant of the second domain of bikunin with increased leukocyte elastase inhibitory activity

The light chain of inter-α inhibitor, also known as bikunin or urinary trypsin inhibitor, is composed of two tandemly arranged Kunitz-type protease inhibitor domains. The second domain of bikunin has factor Xa inhibitory activity which previously was enhanced by mutating two amino acids, glutamine 19 and tyrosine 46 to lysine and aspartate, respectively. In this study, we tried to potentiate its inhibitory activity against leukocyte elastase. A molecular docking model of the second domain of bikunin with leukocyte elastase revealed that P5 arginine 11 was a candidate residue for a third substitution. We generated six triple point mutants using site-directed mutagenesis, compared their leukocyte elastase-inhibitory activities, and selected the most potent variant with arginine 11 substituted to serine. The IC50 values for factor XIa, factor Xa, and leukocyte elastase were 182, 302, and 273 nM, respectively. Moreover, this triple point mutant prolonged the activated partial thromboplastin time and moderately reduced leukocyte elastase-induced endothelial injury. Additionally, favorable conformations created by these mutations were speculated using the structure of the Kunitz protease inhibitor domain of protease nexin 2 complexed with factor XIa as a reference. We discovered a novel triple point mutant of the second domain of bikunin that has potent inhibitory activities against factor XIa, factor Xa, and leukocyte elastase. This variant exhibited anticoagulant activity in plasma and suppressed endothelial cell injury.

Association of depressive symptoms with dietary habits among Japanese physicians

The aim of this study was to determine the association of depressive symptoms with dietary habits among physicians working in hospitals in Japan. We mailed an anonymous questionnaire with the Quick Inventory of Depressive Symptomatology and items about dietary habits to 10,000 randomly selected physicians. Logistic regression analysis was used to explore the association of depressive symptoms with dietary habits. The participants comprised 3862 physicians (3025 men and 837 women). Among the respondents, 252 (8.3%) men and 88 (10.5%) women were determined to be in depressive symptoms. For men, “never eat a balanced diet” (Odds ratio;95% confidence interval, 2.54;1.70 - 3.80), and for women “eat quite small meals” (3.81;1.14 - 13.1), “always eat until full” (4.40;1.48 - 13.1), and “never eat a balanced diet” (3.05;1.49 - 6.23) were associated with depressive symptoms. For men, “do not eat too much” (0.57;0.39 - 0.83) was associated with prevention of depression. Physicians should recognize the important role of healthy dietary habits in good mental health. Hospitals should intervene to help physicians address depressive symptoms and dietary habits.

Atmospheric dust aerosol impacts the Earth’s climate by affecting the radiation *** influences the Earth’s energy budget not only by scattering and absorbing the radiation when it is floating in the atmosphere,but also by decreasing the surface albedo when it is deposited on the snow *** the snow albedo is one of the most important parameter to control the radiation budget on snow surface,the impact of dust deposition on snow has potentially ***,the effect is uncertain because of the lack of the understanding of the relationship between dust deposition and snow *** on long-term observations of snow albedo and snow impurities,we developed a physically based snow albedo model,and incorporated it into a global aerosol transport model called the Model of Aerosol Species IN the Global Atmosphere (MASINGAR).The model calculates the visible and infrared albedos of snow surface as functions of snow impurities,snow grain size,and solar zenith *** snow impurities in the model consist of deposited black carbon and dust aerosols,which have light absorbing *** concentrations of the black carbon and dust aerosols in the snowpack are calculated from wet and dry deposition fluxes of these aerosols using *** performed 3-year global simulations and estimated the global and annual mean radiation budget difference at the top of the atmosphere with and without dust deposition is +0.2 W m for all-sky condition and +0.04 W m for clear sky *** difference in radiation budget is significant over northern Siberia and the Himalayas,where the snow cover remains until early *** result implies that the dust aerosol deposition on snow surface has a significant climatic role.

We investigated whether long-term glimepiride (GP) monotherapy improves insulin resistance and exerts a beneficial effect on beta cell function, as compared with glibenclamide (GC). One hundred Japanese Type 2 diabetic patients were randomly assigned to the GP (n = 50) or the GC (n = 50) group. During a 5-year monitoring period, patients received the indicated SU monotherapy, while changes in SU doses were allowed as needed to maintain HbA1C below 7.0%. The GC group, in parallel with fasting insulin, showed a rapid homeostatic model assessment (HOMA)-R increase and maintained a high HOMA-R level. In contrast, HOMA-R in the GP group decreased continuously, from 2.9 at baseline to 1.8 at study completion. In the GC group, HOMA-b was markedly increased in the first 6 months, then gradually decreased through 18 months. While the HOMA-β elevation in the GP group was more moderate than that in the GC group, HOMA-β levels were maintained with a slight decrease. The cumulative macrovascular disease outcome was 1 for the GP and 7 for the GC group, showing a significant difference. These results suggest that glimepiride monotherapy markedly improved HOMA-R with moderate insulin stimulation, which may account for the difference in macrovascular disease development as compared with the group receiving glibenclamide.
