The Vision of Agri-Environmental Sustainability in Bangladesh: How the Policies, Strategies and Institutions Delivered?

Agriculture and environmental change are tightly interconnected in ways that require active roles of policy, strategy, and institution for the promotion of sustainable agriculture development. To explore this issue, this study addresses two questions 1) to what extent is environmental integration reflected in policies, strategies, and institutions? And 2) how do these policies, strategies, and institutions turn ambitions into actions? By adopting two frameworks developed by the OECD and the European Environment Agency as well as critically reviewing policy documents and conducting the key informant interviews, the results revealed that most of the policies and strategies integrated the environmental issue and had stipulated action plans and governance structure for implementation. The results also found that institutions marginally integrated the environmental concerns and faced several problems that hindered turning ambitions into actions. Overall, although policy instruments were good, the vision of sustainability was in difficulty to actualizing into action due to weak institutional setup. This study highlights the interdependence between the dimensions of policy implement tation process. Policy implications should emphasize enhancing institutional capacity, revamping agricultural advisory services, and empowering Bangladesh Agricultural Research Council (BARC) to translate the vision of agri-environmental sustainability into action.

Innate immune reactivity of the liver in rats fed a choline-deficient L-amino-acid-defined diet

AIM： To investigate the innate immune reactivity of tumor necrosis factor-alpha （TNF-α）, Toll-like receptor 4 （TLR4）, and CD14 in the liver of non-alcoholic steatohepatitis （NASH） model rats. METHODS： Male F344 rats were fed a cholinedeficient L-amino-acid-defined （CDAA） diet. The rats were killed after 4 or 8 wk of the diet, and their livers were removed for immunohistochemical investigation and RNA extraction. The liver specimens were immunostained for TNF-α, TLR4, and CD14. The gene expressions of TNF-α, TLR4, and CD14 were determined by reverse-transcriptase polymerase chain reaction （RT-PCR）. Kupffer cells were isolated from the liver by Percoll gradient centrifugation, and were then cultured to measure TNF-α production. RESULTS： The serum and liver levels of TNF-~ in the CDAA-fed rats increased significantly as compared with the control group, as did the immunohistochemical values and gene expressions of TNF-α, TLR4, and CD14 with the progression of steatohepatitis. TNF-α production from the isolated Kupffer cells of the CDAAfed rats was elevated by lipopolysaccharide stimulation. CONCLUSION： The expressions of TNF-α, TLR4, and CD14 increased in the NASH model, suggesting that

Immunotherapy for nonalcoholic steatohepatitis using the multiple cytokine production modulator Y-40138

AIM:To investigate the possible use of the multiple cytokine production modulator,Y-40138,as a novel immunotherapy in the rat nonalcoholic steatohepatitis (NASH) model. METHODS:We allocated 6-wk-old male F344 rats to choline-supplemented,L-amino acid-defined (CSAA) diet (control group),CSAA diet + Y-40138 (control + Y-40138 group),choline-def icient,L-amino acid-def ined (CDAA) diet (NASH group),or CDAA diet + Y-40138 (NASH + Y-40138 group). In each group,we measured the plasma alanine aminotransferase (ALT) levels,and the plasma and liver levels of tumor necrosis factor-α (TNF-α),interferon-γ (IFN-γ),and interleukin-10 (IL-10). Tissue specimens of phosphate buffered saline-perfused liver were subjected to hematoxylin and eosin staining,Azan staining,Sirius red staining,and immunohistochemical staining (for Kupffer cells and TNF-α). We then extracted Kupffer cells from the collagenase-perfused livers using the Percoll gradient centrifugation method,and measured the TNF-α levels in the supernatant (in vitro TNF-α production by Kupffer cells) using an enzyme-linked immunosorbent assay ***:In comparison to the NASH group,serumALT elevation was mild,production of serum and liver TNF-α and IFN-γ was inhibited,and IL-10 production was increased in the NASH + Y-40138 group. Amelioration of liver histology was also noted in the NASH + Y-40138 group. Kupffer cell immunohistochemical staining revealed no differences between groups,whereas TNF-α immunohistochemical staining showed fewer stained cells in the NASH + Y-40138 group than in the NASH group. The TNF-α levels in the in-vitro Kupffer cell culture supernatant were lower in the NASH + Y-40138 group than in the NASH ***:Administration of Y-40138 to NASH model rats reduced hepatic inflammation and suppressed fibrosis. These results indicate that the multiple cytokine production modulator,Y-40138,is promising as a novel treatment for NASH.

From the branches of Microtropis japonica (Celastraceae), seven phenolic alcohol glucosides, named microtropins J-P (1-7), were isolated. The 6-position of glucose was esterified with 2-ethyl-2,3-dihydroxybutyric acid. Microtropin K (2) was hydrolyzed under a mild basic condition to give methyl (2S,3R)-2-ethyl-2,3-dihydroxybutyrate, whose absolute structure was determined by the comparison of NMR data and the optical rotation value with that reported.

The advent of tyrosine kinase inhibitor(TKI)therapy markedly improved the outcome of patients with chronic-phase chronic myeloid leukemia(CML).However,the poor prognosis of patients with advanced-phase CML and the lifelong dependency on TKIs are remaining challenges;therefore,an effective therapeutic has been *** BCR–ABL p210 fusion protein’s junction region represents a leukemia-specific neoantigen and is thus an attractive target for antigen-specific T-cell ***–ABL p210 fusion-region-specific CD4+T-helper(Th)cells possess antileukemic potential,but their function remains *** this study,we established a BCR–ABL p210 b3a2 fusion-region-specific CD4+Th-cell clone(b3a2-specific Th clone)and examined its dendritic cell(DC)-mediated antileukemic *** b3a2-specific Th clone recognized the b3a2 peptide in the context of HLA-DRB1*09:01 and exhibited a Th1 *** of this clone through T-cell antigen receptor stimulation triggered DC maturation,as indicated by upregulated production of CD86 and IL-12p70 by DCs,which depended on CD40 ligation by CD40L expressed on b3a2-specific Th ***,in the presence of HLA-A*24:02-restricted Wilms tumor 1(WT1)235–243 peptide,DCs conditioned by b3a2-specific Th cells efficiently stimulated the primary expansion of WTI-specific cytotoxic T lymphocytes(CTLs).The expanded CTLs were cytotoxic toward WT1235–243-peptide-loaded HLA-A*24:02-positive cell lines and exerted a potent antileukemic effect in ***,the b3a2-specific Th-clone-mediated antileukemic CTL responses were strongly inhibited by both TKIs and interferon-α.Our findings indicate a crucial role of b3a2-specific Th cells in leukemia antigen-specific CTL-mediated immunity and provide an experimental basis for establishing novel CML immunotherapies.

Bisphenol A(BPA)is a monomer used in manufacturing a wide range of chemical products,including epoxy resins and ***,an important endocrine disrupting chemical that exerts estrogen-like activities,is detectable at nanomolar levels in human serum *** pregnancy associated doses of 17b-estradiol(E2)plus tumor-necrosis factor-a(TNF-a)induce distorted maturation of human dendritic cells(DCs)that result in an increased capacity to induce T helper(Th)2 *** current study demonstrated that the presence of BPA during DC maturation influences the function of human DCs,thereby polarizing the subsequent Th *** the presence of TNF-a,BPA treatment enhanced the expression of CC chemokine ligand 1(CCL1)in *** addition,DCs exposed to BPA/TNF-a produced higher levels of IL-10 relative to those of IL-12p70 on CD40 ligation,and preferentially induced Th2 *** exerts the same effect with E2 at the same dose(0.01–0.1 mM)with regard to DC-mediated Th2 *** findings imply that DCs exposed to BPA will provide one of the initial signals driving the development and perpetuation of Th2-dominated immune response in allergic reactions.

Automatic Anomaly Detection of Respiratory Motion Based on Singular Spectrum Analysis

The realization of automatic anomaly detection of respiratory motion could be very useful to prevent accidental damage during radiation therapy. In this paper, we proposed an automatic anomaly detection method using singular value decomposition analysis. Before applying this method, the investigator needs a normal respiratory motion data of a patient. From these data, a trajectory matrix representing normal time-series feature is created. Decomposing the matrix, we obtained the feature of normal time series. Then, we applied the same procedure to real-time data and obtained real-time features. Calculating the similarity of those feature matrixes, an anomaly score was obtained. Patient motion was observed by a depth camera. In our simulation, two types of motion e.g. cough and sudden stop of breathing were successfully detected, while gradual change of respiratory cycle frequency was not detected clearly.

The Shift of the Ischial Region during Maneuvering the Standard Wheelchair and the Electric Wheelchair in Healthy Adults

Pressure injuries are frequent secondary complications that occur in patients with Spinal Cord Injury (SCI), and the recurrence rate of ischial pressure injury is highest in SCI patients. Most SCI patients use wheelchairs, and some studies have shown a relationship between buttock pressure and sitting posture. However, the pressure distribution during wheelchair maneuvering is unclear. We measured and compared the shift of the ischial region when maneuvering a standard wheelchair or electric wheelchair in healthy adults. The subjects drove wheelchairs on a flat ground of 10 m and the shift of ischial region was measured with a pressure distribution-measuring device. The ischial region shifted forward or backward while maneuvering both the standard wheelchair and the electric wheelchair, and the shift rate of the ischial region was not significantly different between the standard wheelchair and the electric wheelchair. The ischial region shift occurred when the forward head moved by the video while maneuvering both a standard wheelchair and an electric wheelchair. Therefore, the shift might occurred because the pelvis shifted forward in conjunction with head movement during maneuvering of both the standard wheelchair and the electric wheelchair. This result showed that it is important to reduce head and trunk movements when maneuvering a wheelchair to reduce the shear force on the buttocks and prevent pressure injuries.

A histologically proven case of progressive liver sarcoidosis with variceal rupture

Sarcoidosis is a chronic multi-systemic granulomatous disease,and liver involvement frequently *** most cases,no evidence of liver dysfunction is ob-served,and portal hypertension due to sarcoid liver diseases is a ***,no case of liver sarcoidosis has ever been reported with confirma-tion of the disease *** we describe a patient having hepatic sarcoidosis with severe portal hypertension and liver *** diagnosis was histologically confirmed from granulomatous status to established liver cirrhosis over 10 years.A 46-year-old woman developed massive hematemesis due to the rupture of gastric cardial *** underwent emer-gency endoscopic injection sclerotherapy,and clear evi-dence of chronic hepatic *** years ago,she was diagnosed as having sarcoidosis with *** biopsy revealed asymptomatic incidental granulomas without fibrosis *** a couple of years,features of liver dysfunction were manifest and *** years after the first biopsy,a second liver biopsy was performed,and well established dense fibrosis was *** significant liver dysfunction with portal hypertension is rarely seen in sarcoidosis,this case indicates that we have to consider the possibility that sarcoidosis may cause end-stage liver cirrhosis.

Perivascular Fat Attenuation Index on Non-Contrast-Enhanced Cardiac Computed Tomography: Comparison with Coronary Computed Tomography Angiography

Objective: Perivascular fat attenuation index (FAI) measurement on non-contrast-enhanced cardiac computed tomography (NCCT) has not been rigorously validated in previous studies. Herein, we compared perivascular FAI values between NCCT and coronary computed tomography angiography (CCTA). We also investigated the variability and reproducibility of perivascular FAI measurement. Methods: A total of 44 patients who underwent NCCT and CCTA were included in this study. For NCCT, perivascular FAI was measured using three threshold settings: from −30 to −190 Hounsfield Units (HU), −20 to −180 HU, and −10 to −170 HU. For CCTA, perivascular FAI was measured using one threshold setting: from −30 to −190 HU. Perivascular FAI measurements by NCCT were compared with those by CCTA using the paired t-test, and correlations were assessed using Pearson’s correlation coefficient. The intra- and inter-observer variabilities for the measurements with NCCT and CCTA were evaluated with the intraclass correlation coefficient. Results: Perivascular FAI measurements with the threshold setting of −30 to −190 HU were significantly lower on NCCT than on CCTA. There were no significant differences between the perivascular FAI measurements at the remaining thresholds on NCCT and those on CCTA. The perivascular FAI at all thresholds on NCCT correlated significantly with those on CCTA. The intra- and inter-observer agreements were excellent for the measurements on NCCT and CCTA.
