人类脂肪组织抽吸物中脂肪细胞、血管间质成分和脂肪来源干细胞凋亡和坏死的分析

背景与目的脂肪来源干细胞（adipose-derived stem cells，ADSCs）已成为研究最多的成体干细胞。笔者对人类脂肪组织抽吸物中脂肪细胞、血管间质成分（stromal vascular fraction，SVF）和ADSCs凋亡和坏死进行分析。方法通过标准的吸脂术获取人类脂肪抽吸物（n=8）。将SVF从脂肪细胞中分离出来，并通过培养纯化ADSCs。通过一组干细胞标记物来确认培养的ADSCs的表面表型。从SVF中可分离出3种干细胞亚群（CD90^＋／CIM5^-，CDl05^＋／CD45^-及CD34^＋／CD31^-）。利用流式细胞术分析，通过膜联蛋白V／碘化丙啶检验确定细胞凋亡和细胞死亡。结果由倍增时间以及油红O染色和碱性磷酸酶染色均为阳性证明，培养的ADSCs具有长期增殖和分化的能力。从脂肪抽吸物中获取的各部分细胞中，脂肪细胞出现（19．7±3．7）％凋亡和（1．1±0．3）％坏死；SVF（22．0±6．3）％凋亡和（11．2±1．9）％坏死；SVF的坏死率较脂肪细胞高（P〈0．05）。在SVF细胞中，（51．1±3．7）％细胞表达CD90^＋／CD45^-，（7．5±1．0）％细胞表达CDl05^＋／CD45^-，而（26．4±3．8）％细胞表达CD34^＋／CD31^-。表达CD34^＋／CD31^-与表达CDl05^＋／CD45^-的ADSCs相比，其细胞凋亡和细胞坏死的概率均较低（P〈0．05）。结论与脂肪细胞相比，ADSCs有较高的细胞凋亡和细胞坏死发生概率。而在ADSCs亚群之间，细胞凋亡和细胞坏死的程度也明显不同。

Research on the underlying mechanism behind abrasive flow machining on micro-slit structures and simulation of viscoelastic media

In this study,the machining mechanism of abrasive flow machining(AFM)microstructures was analyzed in depth according to the transmission morphology and rheological behaviors of the abrasive *** transmission morphology demonstrated the excellent combination of the polymer melt with abrasive grains at the interface,indicating that the polymer melt,combined with the uniform distribution of the polymer chains,could exert a harmonious axial force on the abrasive *** on the rheological behavior analysis of the abrasive media,for example,the stress relaxation and moduli of storage and loss,a machining mechanism model was established incorporating the effect of microplastic deformation and continuous viscous flow,which was further verified by the grooves along the flow *** addition,the Phanthien-tanner(Ptt)model combined with a wall slipping model was employed to simulate the machining process for the first time *** value of the simulated pressure(1.3 MPa)was similar to the measured pressure(1.45 MPa),as well as the simulated volumetric rate(0.0114 mL/s)to the measured volumetric rate(0.067 mL/s),which further proved the validity of the simulation *** flow duration(21 s)derived from a velocity of 1.2 mm/s further confirmed the residual stretched state of the polymer chains,which favored the elasticity of the abrasive media on the ***,the roughly uniform distribution of the shear rate at the main machining region exhibited the advantages of evenly spread storage and loss moduli,contributing to the even extension of indentation caused by the grains on the target surface,which agreed with the mechanism model and machined surface morphology.

Surface and structure characteristics of commercial K-Feldspar powders:Effects of temperature and leaching media

this study presents morphological and structural variations of K-Feldspar mineral after acid treatment. Both organic and inorganic acids such as C2H2O4, HCl, HNO3 and H2SO4 were employed for this purpose. Another aim of this study was to find an optimum experimental condition for iron(Fe) removal with a minimum damage on the structure of K-Feldspar in which high whiteness index is obtained. the effect of different parameters such as concentration, pH and temperature on the final structure of this mineral was investigated. to find out the chemical composition of powder, XRF was utilized. FtIR, XRD and SEM were employed to study the structure of mineral. Spectrophotometry was chosen to analyze whiteness index of powder after acid treatment. It was found that O—Al—O bond at 647 cm^-1 for H2SO4 and HNO3 treated sample disappeared. However, HCl and C2H2O4 were ineffective at this band. In addition, the results revealed an increase in K-Feldspar content, a decrease in Fe content, an increase in whiteness index and no significant structural change for C2H2O4 leached sample. Whiteness index of 91% was obtained for C2H2O4 leached sample with the pH of 2.5 to 3 at temperature of 50 ℃ and during 1 h.

Design of Portable Shell and tube Heat Exchanger for a Solar Powered Water Distiller

this study presents theoretical considerations and results of a portable shell and tube heat exchanger in a solar water distiller system. the device is composed of a glass heat exchanger, which served as a condenser for vapor condensing which were produced in black paint solar absorber. It was also composed of a tank for water source and a tank for produced distilled water. Shell and tube was designed and simulated using an implicit numerical scheme. Simulation results showed that accumulated mass water greatly depended on the inlet vapor temperature and volume, heat exchanger material, coolant water temperature and volume. thus, changing the material from stainless steel to glass in the same condition （vapor temperature, vapor volume, coolant temperature and coolant volume）. these inexpensive shell and tube heat exchangers permitted to produce 40 litre/day, distilled water from vapor with 378 K inlet temperature in atmosphere pressure. If inlet pressure increases, vapor temperature will decline and thereupon, heat exchanger＇s efficiency tangibility will increase.

介绍了美国加州洛杉矶港口停靠设施两种支挡结构的岩土工程勘探和设计成果,为了在码头提供两个驳船停靠点,要对现有堤坝边坡进行开挖,开挖后的边坡需要有支挡结构支撑。根据现场勘察和室内试验数据,项目地基为砂、泥质砂和黏土层,场地在运行地震水平下(震级5.5,峰值加速度0.28g)和偶发性地震水平下(震级6.5,峰值加速度0.52g)将会发生液化现象。建议支挡结构采用AZ型中心桩和AZ型辅助桩结合的钢板桩,并采用静力和运行震动水平共同作用下的侧压力分布图和极限平衡法分析确定桩长和截面尺寸。

背景与目的脂肪来源干细胞已成为研究最多的成体干细胞。我们对新鲜的人类脂肪抽吸物中脂肪细胞、基质血管细胞群（stromalvascularfraction，SVF）和脂肪来源干细胞（adipose—derivedstemcells，ADSCs）的凋亡和坏死概率进行了检测。方法利用标准的吸脂术方法获取人类脂肪抽吸物（n=8）。将SVF细胞从脂肪细胞中分离出来，并经培养获取纯化的ADSCs。利用一组干细胞标记物来鉴定培养的ADSCs的表面表型。将独特的干细胞亚群（CD90^＋CD45^-、CD105^＋／CD45^-、CD34^＋／CD31^-）从SVF中挑选出来。利用膜联蛋白V／碘化丙啶实验和流式细胞术检测分析细胞凋亡和坏死情况。结果通过细胞倍增时间及油红O染色和碱性磷酸酶染色阳性证实，培养的ADSCs显示出长效增殖和分化的潜能。对脂肪抽吸物进行分离，脂肪细胞的凋亡率为19．7％±3．7％，坏死率为1．1％±0．3％；SVF细胞的凋亡率为22．0％±6．3％，坏死率为11．2％±1．9％；与脂肪细胞相比，SVF细胞有较高的坏死率（P〈0．05）。在SVF细胞中，51．1％±3．7％的细胞表达CD90^＋／CD45^-，7．5％±1．0％的细胞表达CD105^＋／CD45^-，而26．4％±3．8％的细胞表达CD34^＋／CD31^-。与表达CD105^＋／CIM5^-的AD—SCs相比，表达CD34^＋／CD31^-的ADSCs其凋亡和坏死概率更低（P〈0．05）。结论虽然ADSCs比脂肪细胞有更高的凋亡和坏死率。但在各种ADSCs亚群之间，其凋亡和坏死的程度存在明显差异。
