A new system’s geo-referencing from sp.ce is entirely free from any GNSS (Gp. or equivalent) systems. The system addresses to various strategic and economic ap.lications such as in remote clock synchronism, aircraft and balloon navigation, missile and smart bombs tracking, satellite orbital determination and remote target geo-p.sitioning. The new geometry concep. corresp.nds to an “inverted Gp.” configuration, utilizing four ground-based reference stations, synchronized in time, installed at well known geodesic coordinates and a rep.ater in sp.ce, carried by an aircraft, balloon, satellite, etc. Signal transmitted by one of the reference bases is retransmitted by the transp.nder, received back by the four bases, p.oducing four ranging measurements which are corrected for the time delays undergone in every retransmission. A minimization function was derived to comp.re the rep.ater’s p.sitions referred to at least two group. of three reference bases, to correct for the signal transit time at the rep.ater and p.op.gation delays, and consequently to p.ovide the accurate rep.ater p.sition for each time interaction. Once the rep.ater’s coordinates are known, the other determinations and ap.lications become straightforward. The system solving algorithm and p.ocess p.rformance has been demonstrated by simulations adop.ing a p.actical examp.e with the transp.nder carried by an aircraft moving over bases and a target on the ground. Effects p.oduced by reference clock synchronism uncertainties at the four bases on the measurements are reviewed.

Transcrip.ion factors (TFs) are key cellular comp.nents that control gene exp.ession. They recognize sp.cific DNA sequences, the TF binding sites (TFBSs), and thus are targeted to sp.cific regions of the genome where they can recruit transcrip.ional co-factors and/or chromatin regulators to fine-tune sp.tiotemp.ral gene regulation. Therefore, the identification of TFBSs in genomic sequences and their subsequent quantitative modeling is of crucial imp.rtance for understanding and p.edicting gene exp.ession. Here, we review how TFBSs can be determined exp.rimentally, how the TFBS models can be constructed in silico, and how they can be op.imized by taking into account features such as p.sition interdep.ndence within TFBSs, DNA shap., and/or by introducing state-of-the-art comp.tational algorithms such as deep.learning methods. In addition, we discuss the integration of context variables into the TFBS modeling, including nucleosome p.sitioning, chromatin states, methylation p.tterns, 3D genome architectures, and TF coop.rative binding, in order to better p.edict TF binding under cellular contexts. Finally, we exp.ore the p.ssibilities of combining the op.imized TFBS model with technological advances, such as targeted TFBS p.rturbation by CRISp., to better understand gene regulation, evolution, and p.ant diversity.

肾动脉内支架成形术的临床应用

目的：对肾动脉阻塞性病变采用Ｐａｌｍａｚ内支架进行治疗，并对其临床远期疗效进行前瞻性、非随机研究分析。材料与方法：７例（２６％）为经皮肾动脉气囊成形术（ＰＴＲＡ）治疗失败者，１３例（４８％）为ＰＴＲＡ后病变再狭窄，４例（１５％）为ＰＴＲＡ后出现并发症，３例（１１％）为肾动脉开口处狭窄直接作内支架治疗。共有４枚Ｐａｌｍａｚ内支架、５枚Ｐａｌｍａｚ－Ｓｃｈａｔｓ节段性内支架及１８枚Ｐａｌｍａｚ中号内支架被用于本组病例，内支架透视导向下用气囊导管及８Ｆ导引鞘释放到靶血管段。结果：技术成功率为１００％（残余狭窄＜３０％），未发生严重并发症，病变两端平均压差从６．８±１．５ｋＰａ（１ｋＰａ＝７．５ｍｍＨｇ）降至０ｋＰａ（Ｐ＜０．０５），随访结果（３～３２个月，平均１０±６个月）显示４例治愈，２０例缓解，总有效率８９％，平均收缩压从术前２４．８±３．０ｋＰａ降至１８．９±２．０ｋＰａ，舒张压从１４．７±１．３ｋＰａ降至１１．３±１．５ｋＰａ（Ｐ＜０．０５），６例肾功能衰竭病人中有２例术后肾功能改善，另４例稳定不变，２例分别于术后３、１８个月出现病变内再狭窄，但未出现临床症状，病变两端压力差改变不显著。结论：常规ＰＴＲＡ?

Full use of residues from industrial p.ocesses is a fundamental necessity of contemp.rary society, since it avoids imp.cts to the environment by using residues as inp.ts for other p.oducts of high economic and social imp.rtance. In this study, lip.se p.oduction of the crude enzymatic extracts obtained by Asp.rgillus niger using cassava p.el as substrate and sunflower oil as an inductor was investigated. The op.imized cultivation temp.rature and concentration of inductor were determined using the resp.nse surface methodology. The two variables studied exercised influence in the p.oduction of lip.se in the 95% level of confidence. The resp.nse surface obtained indicated that the conditions that maximize lip.se activity p.oduction were 30.5 ~C and initial concentration of sunflower oil was 2.5% （w/w）. Through this analysis, it is evident that extremes in temp.rature and concentration of inductor tend to decrease lip.se p.oduction, since low temp.ratures decrease metabolism and high temp.ratures may inactivate the lip.se. Op.imum lip.se yield was 59.8 U/g of dry p.el which was fermented for 60 h. Lip.se p.oduction p.esents a p.ak of 61.3 U/g, at 72 h of fermentation. However, this value is statistically equal （p.〉 0.05） of the value of lip.se activity obtained for 60 h and 84 h of fermentation.

OBJECTIVES: We studied the acute effect of caffeine on myocardial blood flow(MBF) at rest and exercise in healthy volunteers at normoxia and during acute exp.sure to simulated altitude. BACKGROUND: Caffeine is a widely consumed stimulant, although its cardiovascular safety remains controversial and its effect on MBF is unknown. METHODS: 15O-labeled H2O and p.sitron emission tomograp.y(p.T) were used to measure regional MBF at rest and immediately after sup.ne bicycle exercise in healthy volunteers at normoxia(n=10; mean workload, 175 W; 98% p.edicted; mean age, 27± 6 years) as well as during hyp.xia, simulating an altitude of 4,500 m by inhalation of a mixture of 12.5% oxygen(n=8; 148 W; 78% p.edicted; mean age, 29± 4 years). Measurements were rep.ated 50 min after oral ingestion of caffeine(200 mg). Myocardial flow reserve(MFR) was calculated as the ratio of hyp.remic to resting MBF. RESULTS: Resting MBF was not affected by caffeine at normoxia(1.05± 0.36 ml/min/g vs. 1.17± 0.27 ml/min/g; p.NS), although it was significantly increased at hyp.xia(1.71± 0.41 ml/min/g vs. 2.22± 0.49 ml/min/g; p.p.remic MBF decreased significantly at normoxia(2.51± 0.58 ml/min/g vs. 2.15± 0.47 ml/min/g; p. 0.05) and hyp.xia(5.15± 0.79 ml/min/g vs. 3.98± 0.83 ml/min/g; p. 0.005 vs. baseline; p. 0.005 vs. normoxia). The MFR decreased by 22% at normoxia(2.53± 0.69 to 1.90± 0.49; p. 0.01) and by 39% at hyp.xia(3.13± 0.60 to 1.87± 0.45, p. 0.005; p. 0.05 vs. normoxia). CONCLUSIONS: In healthy volunteers, a caffeine dose corresp.nding to two cup. of coffee(200 mg)signif icantly decreased exercise-induced MFR at normoxia and was even more p.onounced during exp.sure to altitude.

Objective Chest p.in is a common finding in p.tients with hyp.rtrop.ic cardiom yop.thy and can be observed in 40%to 50%of all p.tients. However, the p.thogen esis of these ischemia-like symp.oms is still unclear. Methods Twenty-two p.ti ents with hyp.rtrop.ic cardiomyop.thy and 15 controls underwent p.sitron emissio n tomograp.y for evaluation of regional myocardial p.rfusion and coronary flow r eserve (hyp.remic/baseline myocardial blood flow). Myocardial p.rfusion (mL/min/ g) was measured using >ammonia at rest and during hyp.remia with dip.ridamol e (0.56 mg/kg intravenously). Regional coronary flow reserve was assessed in 3 p.lanes (ap.cal, midventricular, basal) in 4 regions (sep.al, anterior, lateral, i nferior). p.tients were divided into 2 group.: group.1 consisted of 11 p.tients treated with surgical myectomy (age 56 ±10 years) and group.2 consisted of 11 p.atients treated medically (age 53 ±13 years). Results Mean global coronary flow reserve was 3.87 ±0.92 in controls but 2.31 ±0.40 in op.rated (p.p.tients (p.p.p.rated). Similarly, sep.al coronary flow reserve was 4.19 ±1.22 in co ntrols but significantly reduced in op.rated p.tients (2.26±0.48, p.p.tients (1.76 ±0.58; p.p.al flow reserve was significantly higher in op.rated p.tients than in p.tients with medically treated hyp.rtrop.ic cardiomyop.thy (+37%, p.p.rfusion. Conclusions Global and r egional myocardial p.rfusion is reduced in p.tients with hyp.rtrop.ic cardiomyop.athy. However, myectomy may have a beneficial effect on sep.al p.rfusion and flo w reserve. Thus, ischemia seems to p.ay an imp.rtant role in the symp.omatology and p.thop.ysiology of hyp.rtrop.ic cardiomyop.thy.
