A novel animal model for in vivo study of liver cancer metastasis

AIM:To establish an animal model with human hepatocyte-repopulated liver for the study of liver cancer ***:Cell transplantation into mouse livers was conducted using alpha-fetoprotein(AFP)-producing hu-man gastric cancer cells(h-GCCs) and h-hepatocytes as donor cells in a transgenic mouse line expressing urokinase-type plasminogen activator(uPA) driven by the albumin enhancer/promoter crossed with a severe combined immunodeficient(SCID) mouse line(uPA/SCID mice).Host mice were divided into two groups(A and B).Group A mice were transplanted with h-GCCs alone,and group B mice were transplanted with h-GCCs and h-hepatocytes *** replacement index(RI),which is the ratio of transplanted h-GCCs and h-hepatocytes that occupy the examined area of a histological section,was estimated by measuring h-AFP and h-albumin concentrations in sera,respectively,as well as by immunohistochemical analyses of h-AFP and human cytokeratin 18 in histological ***:The h-GCCs successfully engrafted,repopulated,and colonized the livers of mice in group A(RI = 22.0% ± 2.6%).These mice had moderately differentiated adenocarcinomatous lesions with disrupted glandular structures,which is a characteristics feature of gastric *** serum h-AFP level reached 211.0 ± 142.2 g/mL(range,7.1-324.2 g/mL).In group B mice,the h-GCCs and h-hepatocytes independently engrafted,repopulated the host liver,and developed colonies(RI = 12.0% ± 6.8% and 66.0% ± 12.3%,respectively).h-GCC colonies also showed typical adenocarcinomatous glandular structures around the *** mice survived for the full 56 day-study and did not exhibit any metastasis of h-GCCs in the extrahepatic regions during the observational *** mice with an h-hepatocyte-repopulated liver possessed metastasized h-GCCs and therefore could be a useful humanized liver animal model for studying liver cancer metastasis in ***:A novel animal model of human liver cancer metastas

Cell surface protein engineering for high-performance whole-cell catalysts

Cell surface protein engineering facilitated by accumulation of information on genome and protein structure involves heterologous production and modifica- tion of cell surface proteins using genetic engineering, and is important for the development of high-performance whole-cell catalysts. In this field, cell surface display is a major technology by exposing target proteins, such as enzymes, on the cell surface using a cartier protein. The target proteins are fused to the carrier proteins that transport and tether them to the cell surface, as well as to a secretion signal. This paper reviews cell surface display systems for prokaryotic and eukaryotic cells from the perspective of carrier proteins, which determine the number of displayed molecules, and the localization, size, and direction （N- or C-terminal anchoring） of the passengers. We also discuss advanced methods for displaying multiple enzymes and a new method for the immobilization of whole-cell catalysts using adhesive surface proteins.

Artificial and bioartificial liver support:A review of perfusion treatment for hepatic failure patients

Liver transplantation and blood purification therapy,including plasmapheresis,hemodiafiltration,and bioartificial liver support,are the available treatments for patients with severe hepatic *** liver support,in which living liver tissue is used to support hepatic function,has been anticipated as an effective treatment for hepatic *** two mainstream systems developed for bioartificial liver support are extracorporeal whole liver perfusion(ECLP)and bioreactor *** various types of bioartificial liver in view of function,safety,and operability,we concluded that the best efficacy can be provided by the ECLP ***,in our subsequent experiments comparing ECLP and apheresis therapy,ECLP offers more ammonia metabolism than HD and *** addition,ECLP can compensate amino acid imbalance and can secret bile.A controversial point with ECLP is the procedure is labor intensive,resulting in high ***,ECLP has the potential to reduce elevated serum ammonia levels of hepatic coma patients in a short *** these problems are solved,bioartificial liver support,especially ECLP,can be adopted as an option in ordinary clinical therapy to treat patients with hepatic failure.

Removal of fluoride from aqueous solution onto Zr-loaded garlic peel (Zr-GP) particles

Garlic peel,as the raw material,was modified by loading with zirconium(IV),exhibiting quite good uptaking behaviour for fluoride *** adsorption experiments were carried out in batch shaking vessels,and the process was strongly dependent on the pH *** adsorption fits Langmuir model well,and the maximum adsorption capacities at equilibrium pH 2 and 6 are evaluated to be 1.10 and 0.89 mol(fluoride)/kg of Zr-loaded garlic peel gel,*** evaluation of effects of coexisting anions such as nitrate,sulfate and phosphate shows that nitrate and sulfate have no negative effect on the adsorption of fluoride,while phosphate has a little *** kinetics of fluoride is well described by pseudo-second-order rate equation,and the corresponding adsorption rate constant is calculated to be 3.25×10?3 g/(mg·min).

Synthesis of Mesoporous, Nanocrystalline Lanthanum Phosphate in the Presence of Citric Acid and Stearic Acid

Lanthanum phosphate was prepared in the presence of citric acid and stearic acid under methanolic conditions at pH 4.5 and pH 7, respectively. The samples obtained were intensively characterized using X-ray diffraction, nitrogen adsorption-desorption isotherm study, transmission electron microscopy （TEM）, thermal gravimetric and differential thermal analysis, and Fourier transform infrared （FTIR） analysis . The as-synthesized samples prepared at pH 4.5 showed lamellar mesostruroned form with high crystallinity. Results showed that the pore size and pore volume changed when the materials were prepared under different pH conditions. Morphology of the samples was observed by using TEM, which showed that the samples possessed relatively small particles closely packed together. The as-synthesized samples were investigated using FTIR, and the mesopore formation mechanism was discussed.

Synchrotron X-ray diffraction techniques for in situ measurement of hydride formation under several gigapascals of hydrogen pressure

The high-pressure technique is a fundamental tool for realizing novel phase transitions, chemical reactions, and other exotic phenomena. Hydrogenation is one example of a high-pressure reaction; at high pressures of several gigapascals, hydrogen becomes chemically active and reacts with metals and alloys to form hydrides. This paper covers a high-pressure study of the hydrogenation process and the synthesis of hydrides using a cubic-type multi-anvil apparatus. The experimental details of a hydrogenation cell assembly, high-temperature and highpressure generation, and an in situ observation technique are presented. These experiments are conducted with the aid of in situ synchrotron radiation X-ray diffraction measurements operated in an energy-dispersive mode in the conventional manner for time-resolved measurements and a newly developed angle-dispersive mode for observation of the crystal growth process during formation of metal hydrides. Two successful cases of high-pressure hydrogenation are presented: aluminum hydride, Al H3, and an aluminum-based alloy hydride, Al2 Cu Hx, which are potential candidates for hydrogen storage materials.

Leaching of phosphorus from incinerated sewage sludge ash by means of acid extraction followed by adsorption on orange waste gel

Ashes from sewage sludge incineration have a high phosphorus content, approximately 8% （W/W）, which indicates a potential resource of the limiting nutrient. Incineration of sewage sludge with subsequent recovery of phosphorus is a relatively new sludge treatment technique. In this article, the leaching of phosphorus by using sulfuric acid as well as hydrochloric acid by means of several batch experiments was presented. At the same time a selective recovery of phosphorus by adsorption was also discussed. The effects of acid concentration, temperature and time on extraction were studied. The phosphorus leaching increased with the increase in acid concentration and temperature. Kinetic studies showed that the complete leaching of phosphorus took place in less than 4 h. Selective adsorption of phosphorus by using orange waste gel provided a hint for recovery of this natural resource, which eventually could meet the ever-increasing requirement for phosphorus. The overall results indicated that the incinerated sewage sludge ash can be treated with acid to efficiently recover phosphorus and thus can be considered a potentially renewable source of phosphorus.

Fluorescence quantum yield of Gd_(0.9-x)R_xEu_(0.1)OOH(R=Y, La) crystals

*** （R=Y, La） crystals were synthesized by a flux method using a mixture of NaOH and KOH as the flux. X-ray diffraction data were well refined using the Rietveld method assuming that the crystals had a monoclinic structre belonging to the P21/m space group. The unit cell volumes of the Gdo.9 *** crystals increased with increasing x, whereas those of *** decreased with increasing x. The Commission Internationale de IEclairage （CIE） chromaticity coordinates of Gdo.9 *** （R=Y, La） crystals were located in the red region and were nearly constant over the range ofx values considered. The fluorescence quantum yield Dr monotonically decreased with increasing x regardless of the ionic radii of the substitute ions. The fact that the Dr value of Lao.9EuoAOOH was greater than that of *** indicated that the observed decrease in the values of the Gdo.9-,LaxEuo.1OOH crystals up to x=0.3 could not be attributed to an increase in the distance between Eu3＋ ions or to a decrease in the Gd3＋ content. The decrease might be due to an increase of killer centers introduced by elemental substitutions with the larger ion.

Theoretical Propulsion System for Fuel Cell Vehicles with Infinite Cruising Range

The realization of a clean automobile society would require electrically-powered propulsion systems in *** recent years,electric vehicles have attracted considerable attention from the perspective of utilizing electricity generated from natural sources,such as solar and wind *** propulsion method considered in the present investigation differs from the conventional off-board energy scheme in a manner such that pure stoichiometric H2/O2 fuels for fuel cells are generated on-board during vehicle *** this method,energy conversion occurs by means of ESI-PSE(electrostatic-induction potential-superposed electrolysis).If a quasi-static process is assumed,the theoretical power requirement to produce pure stoichiometric H2/O2 fuels is only 17%of the total energy required owing to a new method for supplying power to the EC(electrolytic cell).If an ESI-PSE EC is combined with a fuel cell(FC)to form an energy cycle,a HREG(hydrogen redox electric power generator)that uses solid PEMs(polymer electrolyte membranes)for the EC as well as the FC can be *** to calculations based on data from the operational conditions of commercially available ECs and FCs,more than 70%of the power delivered from the FC can be extracted for driving a motor constantly while a car is in *** of energy self-sustainability on the HREG side,the power control system should not have any power *** propulsion system will realize tough vehicles that can continue running at a top speed at long unlimited cruising range.
