Cysteine-rich secretory protein-1 （CRISP-1） is a glycoprotein secreted by the epididym.l epithelium. It is a m.m.er of a large fam.ly of proteins characterized by two conserved dom.ins and a set of 16 conserved cysteine residues. In m.m.als, CRISP-1 inhibits sperm.egg fusion and can suppress sperm.capacitation. The m.lecular m.chanism.of action of the m.m.alian CRISP proteins rem.ins unknown, but certain non-m.m.alian CRISP proteins can block ion channels. In the rat, CRISP-1 com.rises two form. referred to as Proteins D and E. Recent work in our laboratory dem.nstrates that the D form.of CRISP-1 associates transiently with the sperm.surface, whereas the E form.binds tightly. When the sperm.tozoa are washed, the E form.of CRISP-1 persists on the sperm.surface after all D form.has dissociated. Cross-linking studies dem.nstrate different protein-protein interaction patterns for D and E, although no binding partners for either protein have yet been identified. m.ss spectrom.tric analyses revealed a potential post-translational m.dification on the E form.that is not present on the D form. This is the only discernable difference between Proteins D and E, and presum.bly is responsible for the difference in behavior of these two form. of rat CRISP- 1. These studies dem.nstrate that the m.re abundant D form.interacts with sperm.tozoa transiently, possibly with a specific receptor on the sperm.surface, consistent with a capacitation-suppressing function during sperm.transit and storage in the epididym.s, and also confirm.a tightly bound population of the E form.that could act in the fem.le reproductive tract. （Asian J Androl 2007 July; 9： 508-514）

Natural Variation in Tom.to Reveals Differences in the Recognition of AvrPto and AvrPtoB Effectors from.Pseudom.nas syringae

The Pto protein kinase from.Solanum.pim.inellifolium.interacts with Pseudom.nas syringae effectors AvrPto or AvrPtoB to activate effector-triggered im.unity. The previously solved crystal structures of the AvrPto-Pto and AvrPtoB-Pto com.lexes revealed that Pro binds each effector through both a shared and a unique interface. Hera we use natural variation in wild species of tom.to to further investigate Pto recognition of these two effectors. One species, Solanum.chm.elewskU, was found to have m.ny accessions that recognize only AvrPtoB. The Pto ortholog from.one of these accessions was responsible for recognition of AvrPtoB and it differed from.Solanum.pim.inellifolium.Pto by only 14 am.no acids, including two in the AvrPto-specific interface, glutam.te-49/glycine-51. Converting these two residues to those in Pro （histidine-49/valine-51） did not restore recognition of AvrPto. Subsequent experim.nts revealed that a single substitution of a histidine-to-aspartate at position 193 in Pto, which is not near the AvrPto- specific interface, was sufficient for conferring recognRion of AvrPto in plant cells. The reciprocal substi- tution of aspartate-to-histidine-193 in Pto abolished AvrPto recognition, confirm.ng the im.ortance of this residue. Our results reveal new aspects about effector recognition by Pto and dem.nstrate the value of using natural variation to understand the interaction between resistance proteins and pathogen effectors.

Enhanced stabilization of inorganic cesium.lead triiodide （CsPbl3） perovskite quantum.dots with tri-octylphosphine

In recent years, significant attention has been paid to perovskite m.terials. In particular, lead triiodide-based perovskites have exhibited superb optoelectronic properties. Enhancing the stability of these m.terials is an essential step towards large-scale applications. In this stud） by sim.ly adding trioctylphosphine （TOP） as part of the post-synthesis treatm.nt, we significantly enhance the stability of CsPbI3 quantum.dots （QDs） in the solution phase, which otherwise decay rapidly in hours. For CsPbI3 QDs treated with TOP, the absorption and photolum.nescence em.ssion properties are unchanged over the course of weeks, and the quantum.yield rem.ins alm.st constant at 30% even after 1 m.nth. The m.rphologies of both treated and untreated QDs are initially cubic; however, the treated QDs largely m.intain their initial size and shape, while the untreated ones lose size uniform.ty, which is a sign of degradation. Infrared spectroscopy and X-ray photoelectron spectroscopy confirm.the presence of P in the TOP-treated QDs. We hope that the technique of utilizing a ligand post-treatm.nt will lead to new insights that help to resolve the intrinsic instability issue of triiodide perovskite m.terials and devices.

This work focuses on fundam.ntal understanding of m.crostructure evolution of nanostructured ferritic alloy(NFA) and 25 vol.% Cr3C2 coated Si C(Cr3C2@Si C)-NFA com.osite during spark plasm. sintering at950°C and the following therm.l treatm.nt at 1000°C. A unique bi-phase m.crostructure with distinct Cr-rich and Si-rich phases has been observed for the 25 vol.% Cr3C2@Si C-NFA com.osite, while for the NFA sam.le, the traditional large grain m.crostructure rem.ins. Grain sizes are significantly sm.ller for the25 vol.% Cr3C2@Si C-NFA com.osite com.ared to those for the pure NFA, which can be attributed to the presence of grain boundary phases in the com.osite sam.le. During the therm.l treatm.nt, m.crostructure features can be directly correlated with the dissolution kinetics and phase diagram. calculated using Therm.-Calc/DICTRA/PRISm.~?.

This work focuses on irradiation behaviors of a novel silicon carbide and carbon coated nanostructured ferritic alloy(SiC-C@NFA)com.osite for potential applications as a cladding and structural m.terial for next generation nuclear *** SiC-C@NFA sam.les were irradiated with 1 m.V Kr ions up to 10 dpa at 300 and 4500 *** and defect evolution were studied in-situ at the IVEm.Tandem.facility at Argonne National *** effects of ion irradiation on various phases such asα-ferrite m.trix,(Fe,Cr)_(7)C_(3),and(Ti,W)C precipitates were ***α-ferrite m.trix showed a continuous increase in dislocation density along with spatial ordering of dislocation loops(or loop strings)at>5 *** size of the dislocation loops at 450℃was larger than that at 300℃.The nucleation and growth of new(Ti,W)C precipitates inα-ferrite grains were enhanced with the ion dose at 450℃.This study provides new insight into the irradiation resistance of the SiC-C@NFA system.

Neuronal nicotinic acetylcholine receptors （nAChRs） containing Gt4 and 132 subunits are the principal receptors in the m.m.alian central nervous system.that bind nicotine with high affin- ity. These nAChRs are involved in nicotine dependence, m.od disorders, neurodegeneration and neuroprotection. However, our understanding of the interactions between a4β2-containing （a4β2） nAChRs and other proteins rem.ins lim.ted. In this study, we identified proteins that inter- act with ct4β2 nAChRs in a gene-dose dependent pattern by im.unopurifying β2 nAChRs from.m.ce that differ in ct4 and β2 subunit expression and perform.ng proteom.c analysis using isobaric tags for relative and absolute quantitation （iTRAQ）. Reduced expression of either the a4 or the β2 subunit results in a correlated decline in the expression of a num.er of putative interacting proteins. We identified 208 proteins co-im.nunoprecipitated with these nAChRs. Furtherm.re, stratified lin- ear regression analysis indicated that levels of 17 proteins was correlated significantly with expres- sion of at4β2 nAChRs, including proteins involved in cytoskeletal rearrangem.nt and calcium.signaling. These findings represent the first application of quantitative proteom.cs to produce a β2 nAChR interactom. and describe a novel technique used to discover potential targets for pharm.- cological m.nipulation of a4β2 nAChRs and their downstream.signaling m.chanism..

Bone m.rphogenetic proteins （Bm.s） have m.ltiple roles in skeletal developm.nt, hom.ostasis and regeneration. Bm.s signal via type I and type II serine/threonine kinase receptors （Bm.RI and Bm.RII）. In recent decades, genetic studies in hum.ns and m.ce have dem.nstrated that perturbations in Bm. signaling via Bm.RI resulted in various diseases in bone, cartilage, and m.scles. In this review, we focus on all three types of Bm.RI, which consist of activin-like kinase 2 （ALK2, also called type IA activin receptor）, activin- llke kinase 3 （ALK3, also called Bm.RIA）, and activin-like kinase 6 （ALK6, also called Bm.RIB）. The research areas covered include the current progress regarding the roles of these receptors during m.ogenesis, chondrogenesis, and osteogenesis. Understanding the physiological and pathological functions of these receptors at the cellular and m.lecular levels will advance drug developm.nt and tissue regeneration for treating m.sculoskeletal diseases and bone defects in the future.

Caring for the breast cancer survivor's health and wellbeing

The breast cancer care continuum.entails detection, diagnosis, treatm.nt, and survivorship. During this tim., focus on the whole wom.n and m.dical concerns beyond the breast cancer diagnosis itself is essential. In this com.rehensive review, we critically review and evaluate recent evidence regarding several topics pertinent to and specific for the wom.n living with a prior history of breast cancer. m.re specifically, we discuss the m.st recent recom.endations for contraceptive options including long-acting reversible contraception and em.rgency contraception, fertility and pregnancy considerations during and after breast cancer treatm.nt, m.nagem.nt of m.nopausal vasom.tors sym.tom.and vulvovaginal atrophy which often occurs even in young wom.n during treatm.nt for breast cancer. The need to directly query the patient about these concerns is em.hasized. Our focus is on non-system.c horm.nes and non-horm.nal options. Our holistic approach to the care of the breast cancer survivor includes such preventive health issues as sexual and bone health,which are im.ortant in optim.zing quality of life. We also discuss strategies for breast cancer recurrence surveillance in the setting of a prior breast cancer diagnosis. This review is intended for prim.ry care practitioners as well as specialists caring for fem.le breast cancer survivors and includes key points for evidence-based best practice recom.endations.

Effect of an Environm.ntal Contam.nant, Diisopropyl m.thylphosphonate, on the Blood Pressure of the m.llard

During a toxicological test using diisopropyl m.thylphosphonate (DIm.) on m.llards, an anaesthesia-like response was noted after oral dosing. In order to further elucidate these effects, arterial blood pressures were determ.ned both pre-and post-dosing on adit m.le and fem.le m.llards by cannulation of the left carotid artery. A significant decrease in systolic, diastolic, and m.an blood pressure was noted after the oral adm.nistration of DIm.. There was no significant difference in response between m.les and fem.les. During the first 30 m.n after dosing, systolic pressure fell from.an average of 158 m.Hg, to 94 m.Hg, diastolic decreased from.127 m.Hg to 63 m.Hg, and m.an blood pressure dropped from.138 m.Hg to 75 m.Hg. However, there was no significant effect on pulse pressure, heart rate, or respiratory rate. These results are com.ared to physiological data cited in the literature for various CNS-acting drugs on m.llards.
