环境DNA及其在水生生态系统保护中的应用

环境DNA(environmental DNA,e DNA)是指可以从环境样品(如水、土壤、空气、冰芯等)中直接提取到的DNA片段总和。e DNA技术是在确定调查物种或种群的特异性基因识别片段的基础上,利用各种分子手段检测从环境介质中所提取e DNA包含识别片段的情况,进而确定取样环境中生物的分布状况,包括e DNA获取、e DNA分析和结果分析3个阶段,是近年来新出现的一种生物调查方法。与传统方法相比,e DNA技术具有灵敏度高、省时省力、对调查对象无损伤等优点,不要求调查者具有传统的生物识别及鉴定经验。目前e DNA技术已被应用于目标物种(如入侵物种、濒危物种及其他稀有物种)"有无"的检测、生物量的估测、水体生物多样性的调查等,在水生生态系统的保护中具有广泛的应用前景,但目前仅在少数发达国家展开应用,亟待进一步推广。目前研究者们所用的e DNA方法各不相同,有待对现有方法进行完善并建立技术标准;作为一种调查方法,其时间及空间精确度有待进一步评价;利用e DNA技术估测生物量的准确度还较低,建议首先提高对e DNA产生与降解动力学的认识,再进一步寻求提高其准确度的方法。

对广东南雄盆地、江西池江盆地、安徽潜山盆地、湖南衡阳盆地和内蒙古二连盆地古新世和早始新世沉积物进行的生物地层学、化学地层学和磁性地层学研究,第一次给亚洲早古近纪地层框架提供了严格的年代限定。亚洲古新世和早始新世地层划分为4个生物年代单位(陆生哺乳动物期),从老到新为上湖期、浓山期、格沙头期和伯姆巴期4个亚洲陆生哺乳动物期。南雄盆地古地磁数据显示上湖期的底界约位于C29r的上2/3位置。南雄盆地的古地磁资料及池江盆地最新的古地磁和同位素结果显示上湖-浓山期界线在C27n的上部和C26r的下部之间,接近C27n-C26r转换界面。这一记录支持将上湖-浓山期的界线与北美陆生哺乳动物期中的Torrejonian-Tiffanian期以及海相记录中丹尼-塞兰特期(Danian-Selandi-an)的界线相对比。二连盆地古地磁和同位素研究表明典型的格沙头动物群延续至C24r,并且可能向下延伸至C25r和C26n。南雄盆地的记录指示浓山组和古城村组可能相当于C26r的上部。这些结果表明亚洲哺乳动物分期中的浓山-格沙头期的界线可能在北美哺乳动物分期的Tiffanian期中。衡阳盆地的古地磁和同位素证据指示,以短暂的碳同位素漂移为标志的古新世-始新世界线出现在栗木坪组和岭茶组之间,位于C24r中,与海相记录中的坦尼特-伊普里斯期(Thanetian-Ypresian)界线一致。伯姆巴期岭茶动物群的时代似乎与北美Wa-satchian-0带的动物群可以比较。上湖期和浓山期动物群主要由亚洲特有的动物组成。发生在上湖期和浓山期界线处的动物群更替,与北美Torrejonian-Tiffanian期界线以及丹尼-塞兰特期界线一致。在相隔很远的生态系统上的这种巧合,可能反映出各自独立的对气候变化的生态和/或进化上的响应。格沙头动物群的地方性色彩更弱,有与北美Clarkforkian期动物群共有的属一级分类单元。伯姆巴动物群是全球性的。现生哺乳动物各目在格沙头-伯姆巴期界线处突然出现,与北美和欧洲的记录一致,显示动物群的更替与古新世-始新世界线处短暂出现的全球变暖事件(又称古新世-始新世高温事件)有关。

遗传性骨软骨瘤软骨细胞的体外培养

目的:通过对人遗传性骨软骨瘤软骨细胞的体外培养,观察肿瘤软骨细胞的生长特性及相关生物学特点,为进一步研究骨软骨瘤细胞学起因奠定基础。方法:原代培养遗传性骨软骨瘤组织中的软骨细胞并进行传代,进行活细胞形态观察,同时测定细胞增殖能力及贴壁率,与正常人关节软骨细胞做对照。结果:原代细胞在经过完全消化后,细胞活力仍达到95%。传代后骨软骨瘤细胞体积增大,细胞呈现星形,树突增多,细胞内颗粒多且明显。骨软骨瘤细胞增殖随着传代的增加减慢,第2代细胞增殖能力已经与原代细胞产生明显差别(P<0.01,t=3.203)。原代骨软骨瘤细胞在24h内大部分贴壁,随着传代的增加,细胞贴附能力下降,第4代细胞贴壁率与原代细胞产生明显区别(P<0.01,t=4.611)。结论:体外培养的骨软骨瘤细胞从形态,生物学特性(细胞增殖情况、细胞贴附能力)均不同于正常软骨细胞,为今后更加深入的研究遗传骨软骨瘤的病因学奠定一定的体外培养基础。

A battery of tests was established to determine the oestrogenic, mutagenic and genotoxic potential of two categories of endocrine disrupting chemicals (eDCs), phthalates and alkylphenols. Diisononylphthalate (DINP), diethylhexylphthalate (DeHP), dibutylphthalate (DBP), diisododecylphthalate (DIDP) and 4-nonylphenol (4-NP) were oestrogenic in the yeast estrogen screen (YeS) assay and potently oestrogenic in the MVLN and e-SCReeN assays at environmentally relevant concentrations. DINP and 4-NP were mutagenic in the Ames assay and also induced significant levels of unscheduled DNA synthesis and DNA strand breakage. Significant induction in the percentage of cells containing micronuclei was observed after treatment with DINP, DeHP and 4-NP. In addition, sewage effluents from sewage treatment plants (STPs) in the Border, Midlands and Western (BMW) region of Ireland were significantly oestrogenic in the YeS assay. Moreover, analysis of levels of phthalates and alkylphenol identified in Irish rivers receiving treated effluent showed potent oestrogenicity in the YeS assay. The proliferative and genotoxic ability of the phthalates and alkylphenol, and the oestrogenicity of the treated effluents reported here, is significant as these eDCs and eDCs within the effluent may play a role in the etiology of human abnormalities.

Plugs are crucial for initiating crop production in greenhouses, soil, and controlled environment agriculture (CeA). Vegetable, fruiting, ornamental, and other horticultural crops that utilize plugs for production have demonstrated superior transplant establishment rate, plant health, and total yield. The APS Laboratory for Sustainable Food at Florida Gulf Coast University investigated the quality of plugs grown based on different concentrations and fertigation sources using synthetic and organic sources. We carried out the growth of “Rex Butterhead” Lettuce (Latuca sativa) plugs with five different fertigation treatments, 1) full-strength synthetic starter fertilizer solution;2) half-strength synthetic starter fertilizer solution;3) full-strength organic starter fertilizer solution;4) half-strength organic starter fertilizer solution, and 5) no fertilizer for control. Fertilizer treatments were formulated following manufacturer recommendations. The seeds were sown in Oasis® Horticubes and saturated every day with the different fertilizer treatments. The plugs were cultivated for 15 days in a controlled environment until two leaves after the cotyledons had developed. After 15 days, we collected data which included wet weight (g), dry weight (g), leaf area (cm2), and chlorophyll concentration (mg/cm2). In addition, we derived data including the Leaf Area Index (LAI, cm2/cm2) and Specific Leaf Area (SLA, cm2/g). Descriptive statistics were used to describe the biomass data. A Tukey’s HSD test was carried out to understand the differences between the fertilizer sources. We determined there was a statistically significant difference (P = 7.34e−29) in the measured plug growth parameters due to the various fertigation sources.
