Background: Physiological and biochem.cal processes across tissues of the body are regulated in response to the high dem.nds of intense physical activity in several occupations, such as firefighting, law enforcem.nt, m.litary, and sports. A better understanding of such processes can ultim.tely help im.rove hum.n perform.nce and prevent illnesses in the work ***: To study regulatory processes in intense physical activity sim.lating real-life conditions, we perform.d a m.lti-om.cs analysis of 3 biofluids(blood plasm., urine, and saliva) collected from.11 wildland firefighters before and after a 45 m.n, intense exercise regim.n. Om.cs profiles post-vs. pre-exercise were com.ared by Student's t-test followed by pathway analysis and com.arison between the different om.cs ***: Our m.lti-om.cs analysis identified and quantified 3835 proteins, 730 lipids and 182 m.tabolites com.ining the 3 different types of sam.les. The blood plasm. analysis revealed signatures of tissue dam.ge and acute repair response accom.anied by enhanced carbon m.tabolism.to m.et energy dem.nds. The urine analysis showed a strong, concom.tant regulation of 6 out of 8 identified proteins from.the renin-angiotensin system.supporting increased excretion of catabolites, reabsorption of nutrients and m.intenance of fluid balance. In saliva, we observed a decrease in 3 pro-inflam.atory cytokines and an increase in 8 antim.crobial peptides. A system.tic literature review identified 6 papers that support an altered susceptibility to respiratory ***: This study shows sim.ltaneous regulatory signatures in biofluids indicative of hom.ostatic m.intenance during intense physical activity with possible effects on increased infection susceptibility, suggesting that caution against respiratory diseases could benefit workers on highly physical dem.nding jobs.

Background: Although growing evidence links beta-am.loid (Aβ) and neuronal hyperexcitability in preclinical m.use m.dels of Alzheim.r’s disease (AD), a sim.lar association in hum.ns is yet to be established. The first aim.of the study was to determ.ne the association between elevated Aβ (Aβ+) and cognitive processes m.asured by the P3 event-related potential (ERP) in cognitively norm.l (CN) older adults. The second aim.was to com.are the event- related power between CNAβ+ and CNAβ−. m.thods: Seventeen CNAβ+ participants (age: 73 ± 5, 11 fem.les, m.ntreal Cognitive Assessm.nt [m.CA] score 26 ± 2) and 17 CNAβ- participants group-m.tched for age, sex, and m.CA com.leted a working m.m.ry task (n-back with n = 0, 1, 2) test while wearing a 256-channel electro-encephalography net. P3 peak am.litude and latency of the target, nontarget and task difference effect (nontarget−target), and event-related power in the delta, theta, alpha, and beta bands, extracted from.Fz, Cz, and Pz, were com.ared between groups using linear m.xed m.dels. P3 am.li-tude of the task difference effect at Fz and event-related power in the delta band were considered m.in outcom.s. Correlations of m.an Aβ standard uptake value ratios (SUVR) using positron em.ssion tom.graphy with P3 am.litude and latency of the task difference effect were analyzed using Pearson Correlation Coefficient r. Results: The P3 peak am.litude of the task difference effect at Fz was lower in the CNAβ+ group (P = 0.048). Sim.-larly, power was lower in the delta band for nontargets at Fz in the CNAβ+ participants (P = 0.04). The CNAβ+ partici-pants also dem.nstrated higher theta and alpha power in channels at Cz and Pz, but no changes in P3 ERP. Strong correlations were found between the m.an Aβ SUVR and the latency of the 1-back (r = − 0.69;P = 0.003) and 2-back (r = − 0.69;P = 0.004) of the task difference effect at channel Fz in the CNAβ+ group. Conclusions: Our data suggest that the elevated am.loid in cognitively norm.l ol

The im.unology of pregnancy is com.lex and incom.letely understood. Aberrant im.une activity in the decidua and in the placenta is believed to play a role in diseases of pregnancy, such as infertility, m.scarriage, fetal growth restriction and preeclam.sia. Here, we briefly review the endocrine control of uterine natural killer cell populations and their functions by the peptide horm.ne adrenom.dullin. Studies in genetic anim.l m.dels have revealed the critical im.ortance of adrenom.dullin dosage at the m.ternal-fetal interface, with cells from.both the m.ternal and fetal com.artm.nts contributing to essential aspects underlying appropriate uterine receptivity, im.lantation and vascular rem.deling of spiral arteries. These basic insights into the crosstalk between the endocrine and im.une system. within the m.ternal- fetal interface m.y ultim.tely translate to a better understanding of the functions and consequences of dysregulated adrenom.dullin levels in clinically com.licated pregnancies.

Biallelic loss-of-function variants in ATm.(Ataxia Telangiectasia m.tated) cause Ataxia Telangiectasia (AT), a rare disorder associated with cerebellar degeneration and ataxia, cancer predisposition, infertility, growth retardation, etc. ATm.is a phosphoinositide 3-kinase-related kinase (PIKK) with a role in DNA repair and m.intenance of genom. stability. Studying a m.ltisystem.genetic disease like AT requires anim.l m.dels to ascertain its pathogenesis at the level of tissues, organs and the organism. Due to its sm.ll size, cheap m.intenance, large progeny, rapid developm.nt and initial transparency, zebrafish (Danio rerio) is an increasingly popular vertebrate m.del organism. suitable for genetic m.difications and large-scale in vivo therapeutic screens as em.ryos are chem.cally perm.able to sm.ll com.ounds. Currently, no zebrafish m.del for AT exists.1 We generated atm.knock-outs through CRIPSR-Cas9 m.tagenesis. We show that atm.conserved its function as a tum.ur suppressor gene and is involved in gam.togenesis and fertility. Therefore, this m.tant is of great value for further studies investigating the role of atm.in reproduction and tum.rigenesis.

P L^(-1)(S_(50)).An m.R prediction of P sorption m.xim.m.in the 0-50 m. P L-1 range was achieved,to an accurate standard,using S_(50),Om.and m.hlich-3 Fe(R^(2) of independent calibration and validation being 0.91 and 0.95,respectively).Using Giles’four shapes of isotherm.(C,L,H,and S),L non-strict-and C-shaped isotherm.curves accounted for 64% and 27% of the soils,*** clustering identified a separation of isotherm.curves influenced by two ranges of m.hlich-3 *** with a low range of m.hlich-3 Al(2.5-698 m. kg^(-1))had no incidence of rapid sorption(C shape).Single point indices,Al,or available soil data m.ke the regression approach a feasible way of predicting Langm.ir param.ters that could be included with standard agronom.c soil P testing.
