Therapeutic temperature modulation is associated with pulmonary complications in patients with severe traumatic brain injury

AIM: To examine complications associated with the use of therapeutic temperature modulation(mild hypothermia and normothermia) in patients with severe traumatic brain injury(TBI). METHoDS: one hundred and fourteen charts were reviewed. Inclusion criteria were: severe TBI with Glasgow Coma Scale(GCS) 24 h and non-penetrating TBI. Patients were divided into two cohorts: the treatment group received therapeutic temperature modulation(TTM) with continuous surface cooling and indwelling bladder temperature probes. The control group received standard treatment with intermittent acetaminophen for fever. Information regarding complications during the time in the ICU was collected as follows: Pneumonia was identified using a combination of clinical and laboratory data. Pulmonary embolism, pneumothorax and deep venous thrombosis were identified based onimaging results. Cardiac arrhythmias and renal failure were extracted from the clinical documentation. acute respiratory distress syndrome and acute lung injury were determined based on chest imaging and arterial blood gas results. A logistic regression was conducted to predict hospital mortality and a multiple regression was used to assess number and type of clinical complications. RESULTS: one hundred and fourteen patients were included in the analysis(mean age = 41.4, SD = 19.1, 93 males), admitted to the Jackson Memorial Hospital Neuroscience ICU and Ryder Trauma Center(mean GCS = 4.67, range 3-9), were identified and included in the analysis. Method of injury included motor vehicle accident(n = 29), motor cycle crash(n = 220), blunt head trauma(n = 212), fall(n = 229), pedestrian hit by car(n = 216), and gunshot wound to the head(n = 27). Ethnicity was primarily Caucasian(n = 260), as well as Hispanic(n = 227) and African American(n = 223); four patients had unknown ethnicity. Patients received either TTM(43) or standard therapy(71). In this paper we demonstrated a method to reconstruct vector-valued lattice distortion fields within nanoscale crystals by optimization of a forward model of multi-reflection Bragg coherent diffraction imaging(MR-BCDI)*** method flexibly accounts for geometric factors that arise when making BCDI measurements,is amenable to efficient inversion with modern optimization toolkits,and allows for globally constraining a single image reconstruction to multiple Bragg peak *** is enabled by a forward model that emulates the multiple Bragg peaks of a MR-BCDI experiment from a single estimate of the 3D crystal *** present this forward model,we implement it within the stochastic gradient descent optimization framework,and we demonstrate it with simulated and experimental data of nanocrystals with inhomogeneous internal lattice *** find that utilizing a global optimization approach to MR-BCDI affords a reliable path to convergence of data which is otherwise challenging to reconstruct.

oR^His, a Natural Variant of oR, Specifically Interacts with Plastid Division Factor ARC3 to Regulate Chromoplast Number and Carotenoid Accumulation

Chromoplasts are colored plastids that synthesize and store massive amounts of *** number and size define the sink strength for carotenoid accumulation in ***,nothing is known about the mechanisms controlling chromoplast ***,a natural allele of orange(oR),oR^His,was found to promote carotenoid accumulation by activating chromoplast differentiation and increasing carotenoid biosynthesis,but cells in orange tissues in melon fruit and cauliflower oR mutant have only one or two enlarged *** this study,we investigated an oR^His variant of Arabidopsis oR,genetically mimicking the melon oR^His allele,and found that it also constrains chromoplast number in Arabidopsis *** in vitro and in vivo experiments demonstrate that oR^His specifically interacts with the Membrane occupation and Recognition Nexus domain of ACCUMULATIoN AND REPLICATIoN oF CHLoRoPLASTS 3(ARC3),a crucial regulator of chloroplast *** further showed that oR^His interferes with the interaction between ARC3 and PARALoG oF ARC6(PARC6),another key regulator of chloroplast division,suggesting a role of oR^His in competing with PARC6 for binding to ARC3 to restrict chromoplast *** or knockout of ARC3 in Arabidopsis oR^His plants significantly alters total carotenoid ***,overexpression of the plastid division factor PLASTID DIVISIoN 1 greatly enhances carotenoid *** division factors likely alter carotenoid levels via their influence on chromoplast number and/or *** together,our findings provide novel mechanistic insights into the machinery controlling chromoplast number and highlight a potential new strategy for enhancing carotenoid accumulation and nutritional value in food crops.

Gold Mining and Mercury Bioaccumulation in a Floodplain Lake and Main Channel of the Tambopata River, Perú

Contamination of water bodies by inorganic mercury (Hg[II]) used in placer mining of gold deposits in the Madre de Dios Department, Perú, contributes to the bioaccumulation of methylmercury (MeHg) in fish tissue. We measured MeHg and total Hg (THg) concentrations (mg/kg wet weight [ww] tissue) of thirteen fish species from the Tambopata River, Perú, and the connected oxbow lake Tres Chimbadas. We also used stable isotope analysis (δ15N and δ13C) to estimate trophic positions of fishes. Average MeHg concentrations of fish species ranged from 0.042 mg/kg ww (Satanoperca jurupari) to 0.463 mg/kg ww (Hoplias malabaricus) in the main channel and from 0.090 mg/kg ww (Parauchenipterus sp.) to 1.282 mg/kg ww (Pimelodina flavipinnis) in the lake. Spearman rank correlation indicated that trophic position had no influence on MeHg concentrations of species in the main channel, but in the lake, trophic positions of species were positively associated with MeHg. Migrations of the pimelodid catfish surveyed from the main channel are well documented. Because little gold mining occurs at our study site, fishes from the main channel may be bioaccumulating MeHg from areas where mining is widespread. Fish species that reside in the lake are relatively sedentary and migration is limited by the brief period of floodplain inundation and the long, narrow corridor that connects the lake to the main channel;lake sediments are therefore the likely source for MeHg bioaccumulation. Five out of the eight fish species surveyed from the main channel and two out of the five species from the lake had MeHg levels higher than United States Environmental Protection Agency fish tissue criterion for human consumption.

FtsZ1 and FtsZ2 are phylogenetically distinct families of FtsZ in plants that co-localize to mid-plastid rings and facilitate division of chloroplasts. In plants, altered levels of either FtsZ1 or FtsZ2 cause dose-dependent defects in chloroplast division; thus, studies on the functional relationship between FtsZgenes require careful manipulation of FtsZ levels in vivo. To define the functional relationship between the two FtsZ2 genes in Arabidopsis thaliana, FtsZ2-1 and FtsZ2-2, we expressed FtsZ2-1 in an ftsZ2-2 null mutant, and vice versa, and determined whether the chloroplast division defects were rescued in plants expressing different total levels of FtsZ2. Full rescue was observed when either the FtsZ2-1 or FtsZ2-2 level approximated total FtsZ2 levels in wild-type （WT）. Additionally, FtsZ2-2 interacts with ARC6, as shown previously for FtsZ2- 1. These data indicate that FtsZ2-1 and FtsZ2-2 are functionally redundant for chloroplast division in Arabidopsis. To rigorously validate the requirement of each FtsZ family for chloroplast division, we replaced FtsZ1 with FtsZ2 in vivo, and vice versa, while maintaining the FtsZ level in the transgenic plants equal to that of the total level in WT. Chloroplast division defects were not rescued, demonstrating conclusively that FtsZ1 and FtsZ2 are non-redundant for maintenance of WT chloroplast numbers. Finally, we generated ftsZtriple null mutants and show that plants completely devoid of FtsZ protein are viable and fertile. As plastids are presumably essential organelles, these findings suggest that an FtsZ-independent mode of plastid partitioning may occur in higher plants.

Nodal segments from secondary branches of saplings of Phyllostachys bambusoides were inoculated in MS medium to assess the in vitro morphogenic response of leaf sheath through the induction to callogenesis by Picloram（4-amino-3,5,6-trichloropicolinic acid） at different concentrations of carbohydrate under the same conditions with presence or absence of *** our experiment,secondary explants were kept in MS medium containing 8.0 mg·L-1 of Picloram for the callus *** were transferred in MS medium supplemented with sucrose,fructose and glucose（control,2%,4% and 6%）.Results show that Picloram induced the callogenesis in leaf *** secondary embryogenesis was formed in yellow-globular *** sucrose as carbohydrate source in the absence of light was more efficient to induce *** was more efficiency in the presence of *** induction and further embryogenesis evidenced the competence and determination of leaf sheath cells.

目的探讨诱导SW872脂肪细胞分化的最优方法。方法根据不同的分化诱导刺激分为6组:①对照组:无分化刺激,仅有DMEM/F12培养基;②胰岛素组:含2 mg/L Insulin;③1-甲基3-异丁基黄嘌呤组:含0.5 mmol/L IBMX;④促酰化蛋白组:含50 mg/L ASP;⑤激素鸡尾酒(胰岛素+1-甲基3-异丁基黄嘌呤+地塞米松组:含2 mg/L insulin+0.5 mmol/L IBMX+1.0μmol/L DEX;⑥油酸(oleic acid)组:含0.2 mg/L oleic acid。通过形态学观察、油红染色、测定细胞甘油三酯总量的方法。结果多组分化刺激中,油酸组刺激可在24 h内诱导SW872细胞分化,而且分化均匀,分化率高,分化后形成的脂肪细胞形态典型;而且油酸组促进SW872细胞分化过程中细胞内甘油三酯的积累。结论油酸是诱导SW872细胞分化的最佳方法,该方法的建立和统一,为进一步利用SW872细胞进行脂代谢、脂肪细胞功能的研究奠定了基础。

Pancreatic cancer surgery: past, present, and future

The history of pancreatic cancer surgery, though fraught with failure and setbacks, is punctuated by periods of incremental progress dependent upon the state of the art and the mettle of the surgeons daring enough to attempt it. Surgical anesthesia and the aseptic techniques developed during the latter half of the 19 th century were instrumental in establishing a viable setting for pancreatic surgery to develop. Together, they allowed for bolder interventions and improved survival through the postoperative period. Surgical management began with palliative procedures to address biliary obstruction in advanced disease. By the turn of the century, surgical pioneers such as Alessandro Codivilla and Walther Kausch were demonstrating the technical feasibility of pancreatic head resections and applying principles learned from palliation to perform complicated anatomical reconstructions. Allen o. Whipple, the namesake of the pancreaticoduodenectomy（PD）, was the first to take a systematic approach to refining the procedure. Perhaps his greatest contribution was sparking a renewed interest in the surgical management of periampullary cancers and engendering a community of surgeons who advanced the field through their collective efforts. Though the work of Whipple and his contemporaries legitimized PD as an accepted surgical option, it was the establishment of high-volume centers of excellence and a multidisciplinary approach in the later decades of the 20^th century that made it a viable surgical option. Today, pancreatic surgeons are experimenting with minimally invasive surgical techniques, expanding indications for resection, and investigating new methods for screening and early detection. In the future, the effective management of pancreatic cancer will depend upon our ability to reliably detect the earliest cancers and precursor lesions to allow for truly curative resections.
