The Or Gene enhances Carotenoid Accumulation and Stability During Post-Harvest Storage of Potato Tubers

Provitamin A carotenoids in staple crops are not very stable during storage and their loss compromises nutritional quality. To elucidate the fundamental mechanisms underlying carotenoid accumulation and stability, we investigated transgenic potato tubers that expressed the cauliflower Orange （Or） gene. We found that the Or transgene not only promoted retention of 13-carotene level, but also continuously stimulated its accumulation during 5 months of cold storage. In contrast, no increased levels of carotenoids were observed in the tubers of vector-only controls or a yellow- flesh variety during the same period of storage. The increased carotenoid accumulation was found to be associated with the formation of lipoprotein-carotenoid sequestering structures, as well as with the enhanced abundance of phytoene synthase, a key enzyme in the carotenoid biosynthetic pathway. Furthermore, the provitamin A carotenoids stored were shown to be stable during simulated digestion and accessible for uptake by human intestinal absorptive cells. Proteomic analysis identified three major functional groups of proteins （i.e. heat shock proteins, glutathione-S-transferases, and carbohydrate metabolic proteins） that are potentially important in the Or-regulated carotenoid accumulation. Our results show that regulation of carotenoid sequestration capacity is an important mechanism by which carotenoid stability is regulated. Our findings suggest that induction of a proper sink structure formation in staple crops may provide the crops with a unique ability to promote and/or stabilize provitamin A accumulation during plant growth and post-harvest storage.

There are five vital signs that healthcare providers assess: temperature, pulse, respiration, blood pressure, and pain. Normal levels for the five vital signs are published by the American Heart Association, and other specialty organizations, however, the sixth vital sign (resilience) which adopts the measure of immune resilience is suggested in this paper. Resilience is the ability of the immune system to respond to attacks and defend effectively against infections and inflammatory stressors, and psychological resilience is the capacity to resist, adapt, recover, thrive, and grow from a challenge or a stressor. Individuals with better optimal immune resilience had better health outcomes than those with minimal immune resilience. The purpose of this paper is to conceptualize, contextualize, and operationalize all six vital signs. We suggest measuring resilience subjectively and objectively. Subjectively, use a 5-item guided interview revised from the Connor-Davidson Resilience Scale (CDRC), a scale of 10 items. The revised CDRC scale is a 5-item scale. The scale is rated on a 5-point Likert scale from 0 (not true) to 4 (true all the time). The total score ranges from 0 to 20, with higher total scores indicating greater resilience. The scale demonstrated good construct validity and internal consistency (α = 0.85) during the development of the scale. The CD-RISC had a good Cronbach’s alpha level of 0.85. The Revised CD-RISC can be completed in 2 - 4 minutes. To measure resilience objectively, we suggest using Immune Resilience (IR) levels, the level of resilience to preserve and/or rapidly restore immune resilience functions that promote disease resistance and control inflammation and other inflammatory stress. IR levels are gauged with two peripheral blood metrics that quantify the balance between CD8 and CD4 T-cell levels and gene expression signatures tracking longevity-associated immunocompetence and mortality- or entropy-associated inflammation. The study examined the contamination levels of the soil and plantain leaves in three communities in Atakunmosa west local government area of Osun State in southwest Nigeria, where gold mining activities have recently become intensive. Plantain is a major food crop, whose leaves are used to wrap food items for household consumption. The objectives were to examine the heavy metal concentrations in the soil and plantain leaves around the gold mine site and compare them with standard recommended safe limits for the environment. Soil and plantain leaves were sampled at different locations around the sites and at a control site from a neighboring local government area with no history of gold mining activities between March 2015 and February 2016. Both soil and leave samples were processed and analyzed for selected heavy metals (Cd, As, Cu, Zn, Cr, Mn and Fe) using the Particle Induced X-ray emission (PIXe) in the laboratory. The study showed higher concentrations than the World Health Organization’s recommended safe limits of the heavy metals in the soils and plantain leaves. Dry season concentrations of the variables were also higher than the wet season and the heavy metal concentrations at the control station were significantly (p er than those of the mining environment. The study concluded that the ecosystems in the artisanal gold mining region are vulnerable to bioaccumulation of heavy metals and the leaves from the sites are sources of heavy metal contamination if consumed or used to wrap food items.

背景：心房纤颤（简称房颤）与死亡和心血管事件（特别是卒中）危险性增加相关，因此，预防新发房颤是一个重要的临床问题。虽然心电图左心室肥厚（electrocardiographic left ventricular hypertrophy，LVH）的存在及其严重程度可以预测房颤的发生，但是心电图LVH消退是否与房颤发生率下降相关尚不清楚。目的：验证降压治疗期间心电图LVH消退或持续消失与房颤发生率下降相关且独立于血压水平及治疗模式的假设。设计、方法及参试者：1995～2001年于8831例男性和女性高血压患者中进行双盲、随机、平行分组研究。患者年龄55～80岁（中位数，67岁），均根据心电图Comell电压-间期乘积或Sokolow—Lyon电压标准诊断LVH。患者无房颤史，基线心电图无房颤，已入选氯沙坦干预降低终点事件高血压研究。干预：氯沙坦或阿替洛尔治疗方案，6个月时进行随访评估，之后每年随访，直至患者死亡或试验终止。主要观测指标：基线及随访各时点心电图LVH与新发房颤的相关性。心电图LVH的测量使用性别校正的Comell乘积标准（{RavL＋Sv3[女性＋6mm]}×QRS间期）。结果：经过平均（SD）4．7（1．1）年的随访，治疗期间心电图Comell乘积标准LVH消退或持续消失的患者有290例新发房颤（发生率为14．9／1000病人-年）；而心电图LVH持续或进展的患者则有411例新发房颤（19．0／1000病人-年）。时间依赖Cox分析显示，在校正治疗效果、基线房颤危险因素差异、基线及治疗期间血压以及基线LVH严重程度后，治疗期间Comell乘积标准LVH降低作为一种时间变异协变量与新发房颤下降相关。Comell乘积每下降1050mm×msec（每个SD），新发房颤就下降12．4％（校正风险比[hazard ratio，HR]，0．88；95％可信区间[confidence interval，CI]0．80～0．97；P=0．007）；氯沙坦减少高血压患者发生房颤的益处优于阿替洛尔（HR，0．83；95％CI，0．71～0．97；P=0．01）。结论：原发性高血压患者在降压治疗期间，心电图Comell乘积标准LVH消退与新发房颤下降相关，而且独立于血压和治疗方案。这些发现提示，以降低或预防心电图LVH为目标的降压治疗可以降低新发房颤的发生率。
