Dendritic cells(DCs)are indispensable for defense against pathogens but may also contribute to *** of DCs upon the sensing of pathogens by Toll-like receptors(TLRs)is largely mediated by pattern recognition receptor/nuclear factor-κB(NF-κB)signaling and depends on the appropriate ubiquitination of the respective signaling ***,the ubiquitinating and deubiquitinating enzymes involved and their interactions are only incompletely ***,we reveal that the deubiquitinase OTU domain,ubiquitin aldehyde binding 1(OTUB1)is upregulated in DCs upon murine Toxoplasma gondii infection and lipopolysaccharide *** of DCs with the TLR11/12 ligand *** profilin and the TLR4 ligand lipopolysaccharide induced an increase in NF-κB activation in OTUB1-competent cells,resulting in elevated interleukin-6(IL-6),IL-12,and tumor necrosis factor(TNF)production,which was also observed upon the specific stimulation of TLR2,TLR3,TLR7,and ***,OTUB1 promoted NF-κB activity in DCs by K48-linked deubiquitination and stabilization of the E2-conjugating enzyme UBC13,resulting in increased K63-linked ubiquitination of IRAK1(IL-1 receptor-associated kinase 1)and TRAF6(TNF receptor-associated factor 6).Consequently,DC-specific deletion of OTUB1 impaired the production of cytokines,in particular IL-12,by DCs over the first 2 days of *** infection,resulting in the diminished production of protective interferon-γ(IFN-γ)by natural killer cells,impaired control of parasite replication,and,finally,death from chronic ***,all of which could be prevented by low-dose IL-12 treatment in the first 3 days of *** contrast,impaired OTUB1-deficient DC activation and cytokine production by OTUB1-deficient DCs protected mice from lipopolysaccharide-induced ***,these findings identify OTUB1 as a potent novel regulator of DCs during infectious and inflammatory diseases.

There is increasing evidence that pathogens do not only elicit direct defense responses, but also cause pronounced changes in primary carbohydrate metabolism. Cell-wall-bound invertases belong to the key regulators of carbohydrate partitioning and source-sink relations. Whereas studies have focused so far only on the transcriptional induction of invertase genes in response to pathogen infection, the role of post-translational regulation of invertase activity has been neglected and was the focus of the present study. Expression analyses revealed that the high mRNA level of one out of three proteinaceous invertase inhibitors in source leaves of Arabidopsis thaliana is strongly repressed upon infection by a virulent strain of Pseudomonas syringae pv. tomato DC3000. This repression is paralleled by a decrease in invertase inhibitor activity. The physiological role of this regulatory mechanism is revealed by the finding that in situ invertase activity was detectable only upon infection by P. syringae. In contrast, a high invertase activity could be measured in vitro in crude and cell wall extracts prepared from both infected and non-infected leaves. The discrepancy between the in situ and in vitro invertase activity of control leaves and the high in situ invertase activity in infected leaves can be explained by the pathogen-dependent repression of invertase inhibitor expression and a concomitant reduction in invertase inhibitor activity. The functional importance of the release of invertase from post-translational inhibition for the defense response was substantiated by the application of the competitive chemical invertase inhibitor acarbose. Posttranslational inhibition of extracellular invertase activity by infiltration of acarbose in leaves was shown to increase the susceptibility to P. syringae. Polyether and polyether/ester based TPU （thermoplastic polyurethanes） were investigated with wide-angle XRD （X-ray diffraction） and SAXS （small angle X-ray scattering）. Furthermore, SAXS measurements were performed in the temperature range of 30 ℃ to 130 ℃. Polyether based polymers exhibit only one broad diffraction signal in a region of 2 θ 15° to 25°. In case of polyurethanes with ether/ester modification, the broad diffraction signal arises with small sharp diffraction signals. SAXS measurements of polymers reveal the size and shape of the crystalline zones of the polymer. Between 30 ℃ and 130 ℃ the size of the crystalline zone changes significantly. The size decreases in most of investigated TPU. In the case of Desmopan 9365D an increase of the particle size was observed.

AIC-Based Selection of Growth Models: The Case of Piglets from Organic Farming

The selection and comparison of different growth models for describing weight gain of piglets raised in organic farming is investigated by using the Akaike’s Information Criterion (AIC). In total, 49,699 data points of 5188 piglets recorded between 2007 and 2013 were considered. From the day of birth, up to 40 days (i.e. until weaning) the model of von Bertalanffy was favored by the AIC. This model is with 60.32% more likely to truly reflect reality than any other of the analyzed models. Up to 105 days, the two-linear model was favored by the AIC (probability 99.75%). The intersection point of the two-linear model was calculated by 53.8 days, which fitted well to the actual change in the food situations.

The paper searched for raw data about wild-caught fish, where a sigmoidal growth function described the mass growth significantly better than non-sigmoidal functions. Specifically, von Bertalanffy’s sigmoidal growth function (metabolic exponent-pair a = 2/3, b = 1) was compared with unbounded linear growth and with bounded exponential growth using the Akaike information criterion. Thereby the maximum likelihood fits were compared, assuming a lognormal distribution of mass (i.e. a higher variance for heavier animals). Starting from 70+ size-at-age data, the paper focused on 15 data coming from large datasets. Of them, six data with 400 - 20,000 data-points were suitable for sigmoidal growth modeling. For these, a custom-made optimization tool identified the best fitting growth function from the general von Bertalanffy-Pütter class of models. This class generalizes the well-known models of Verhulst (logistic growth), Gompertz and von Bertalanffy. Whereas the best-fitting models varied widely, their exponent-pairs displayed a remarkable pattern, as their difference was close to 1/3 (example: von Bertalanffy exponent-pair). This defined a new class of models, for which the paper provided a biological motivation that relates growth to food consumption.
