Environmental exposure to cadmium but not lead is associated with decreased semen quality parameters:quality regionalism of sperm properties

Environmental factors may negatively contribute to a progressive worsening of semen quality,and differences in semen quality may result from different environmental exposures(regional differences)or lifestyle differences.Heavy metals are factors with a confirmed negative influence on male fertility.Among them,lead and cadmium are commonly found in human surroundings.Thus,we analyzed semen parameters(according to the World Health Organization 2010 recommendations)and semen lead and cadmium concentrations in 188 men from two different regions in Poland,a typical agricultural area and an industrial area,in couples that had been diagnosed with infertility.The assays were performed using flameless electrothermal atomic absorption spectrometry.In the statistical analysis,regional comparisons and then taxonomic comparisons based on three parameters(age,semen concentration,and sperm morphology)were applied.We showed that more cadmium than lead accumulated in semen,a higher cadmium concentration was observed in semen obtained from men from the agricultural region,and better semen quality and lower cadmium concentrations were found in the semen of men from the industrial,more polluted region.We thus showed an existing regionalism in the sperm quality properties.However,semen parameters such as morphology and progressive and nonprogressive motility followed the same trends,regardless of the patient’s age,region,or class.We could conclude that the environment has a minor impact on sperm morphology and progressive and nonprogressive motility and that other existing factors could have an indirect influence on semen quality.

The performance of Polish healthcare is still far from satisfactory,which is connected with growing indebtedness that adversely affects not only the development of healthcare but also the quality of medical services provided.In turn,the financial situation of hospitals depends on various indicators and the relationship between them.This paper investigates the determinants of healthcare sector functioning in the context of hospitals’financial situation(dependent variable measured by total debt ratio).321 individual public units were examined between years 2007–2017.More specifically,the fixed effects panel data model in question measures and evaluates the impact of economic,regional,institutional,and social factors in determining the dependent variable.This study confirmed dependencies between financial and non-financial ratios along with their impact on the financial condition of studied entities.The management implications of our study lay the groundwork for the improvement of efficient resource management for public hospitals.

The typical mammalian neck consisting of seven cervical vertebrae(C1–C7)was established by the Late Permian in the cynodont forerunners of modern mammals.This structure is precisely adapted to facilitate movements of the head during feeding,locomotion,predator evasion,and social interactions.Eutheria,the clade including crown placentals,has a fossil record extending back more than 125 million years revealing significant morphological diversification in the Mesozoic.Yet very little is known concerning the early evolution of eutherian cervical morphology and its functional adaptations.A specimen of Zalambdalestes lechei from the Late Cretaceous of Mongolia boasts exceptional preservation of an almost complete series of cervical vertebrae(C2–C7)revealing a highly modified axis(C2).The significance of this cervical morphology is explored utilizing an integrated approach combining comparative anatomical examination across mammals,muscle reconstruction,geometric morphometrics and virtual range of motion analysis.We compared the shape of the axis in Zalambdalestes to a dataset of 88 mammalian species(monotremes,marsupials,and placentals)using three-dimensional landmark analysis.The results indicate that the unique axis morphology of Zalambdalestes has no close analog among living mammals.Virtual range of motion analysis of the neck strongly implies Zalambdalestes was capable of exerting very forceful head movements and had a high degree of ventral flexion for an animal its size.These findings reveal unexpected complexity in the early evolution of the eutherian cervical morphology and suggest a feeding behavior similar to insectivores specialized in vermivory and defensive behaviors in Zalambdalestes akin to modern spiniferous mammals.

Autophagy is a prosurvival mechanism for the clearance of accumulated abnormal proteins,damaged organelles,and excessive lipids within mammalian cells.A growing body of data indicates that autophagy is reduced in aging cells.This reduction leads to various diseases,such as myocardial hypertrophy,infarction,and atherosclerosis.Recent studies in animal models of an aging heart showed that fasting-induced autophagy improved cardiac function and longevity.This improvement is related to autophagic clearance of damaged cellular components via either bulk or selective autophagy(such as mitophagy).In this editorial,we summarize the mechanisms of autophagy in normal and aging hearts.In addition,the protective effect of fasting-induced autophagy in cardiac aging has been highlighted.

This narrative review provides an overview of the utilization of endoscopic ultrasound-guided gallbladder drainage(EUS-GBD)as a salvage approach in cases of unsuccessful conventional management.EUS-GBD is a minimally invasive and effective technique for drainage in patients with acute cholecystitis with high risk of surgery.The procedure has demonstrated impressive technical and clinical success rates with low rates of adverse events,making it a safe and effective option for appropriate candidates.Furthermore,EUS-GBD can also serve as a rescue option for patients who have failed endoscopic retrograde cholangiopancreatography or EUS biliary drainage for relief of jaundice in malignant biliary stricture.However,patient selection is critical for the success of EUS-GBD,and proper patient selection and risk assessment are important to ensure the safety and efficacy of the procedure.As the field continues to evolve and mature,ongoing research will further refine our understanding of the benefits and limitations of EUS-GBD,ultimately leading to improved outcomes for patients.

Acute coalescent mastoiditis in a 16-month-old child due to Streptococcus pneumoniae infection

Rationale:Acute otitis media is a common disease in early childhood,and is usually caused by Streptococcus pneumoniae(S.pneumoniae).Acute mastoiditis is a complication of acute otitis media and can involve not only the mucoperiosteum of the middle ear but can also spread to the periosteum by destroying the mastoid bone(acute coalescent mastoiditis).In addition,the infection can extend through the surrounding bones or the emissary veins beyond the mastoid’s air cells,leading to subperiosteal abscesses.Patient’s Concern:A 16-month-old female patient was hospitalized due to the purulent discharge of the left ear and the symptoms of right mastoiditis(swelling and redness of the skin).Diagnosis:Bilateral acute coalescent mastoiditis caused by S.pneumoniae infection.The computer tomography revealed bilateral bone destruction of the mastoid and abscesses found behind the auricle on both sides.Interventions:The patient underwent intravenous antibiotic therapy and surgical treatment.Outcomes:The patient was discharged 14 days after hospitalization with an improved condition.Lessons:Improperly treated acute coalescent mastoiditis can lead to extracranial and intracranial complications,sometimes serious and even life-threatening.Complications are prevalent in children under 2 years,in whom the disease progresses more rapidly and severely.The vaccination with a 13-valent vaccine may not result in sufficient immunity against S.pneumoniae,a predominant pathogen in children affected by acute coalescent mastoiditis.

Operational guidance for aeration and flow augmentation for the Chicago Area Waterway System—A case study

The Chicago Area Waterway System(CAWS)is a 133.9 km branching network of navigable waterways controlled by hydraulic structures,in which the majority of the flow is treated wastewater effluent and there are periods of substantial combined sewer overflows.The CAWS comprises a network of effluent dominated streams.More stringent dissolved oxygen(DO)standards and a reduced flow augmentation allowance have been recently applied to the CAWS.Therefore,a carefully calibrated and verified one-dimensional flow and water quality model was applied to the CAWS to determine emission-based real-time control guidelines for the operation of flow augmentation and aeration stations.The goal of these guidelines was to attain DO standards at least 95%of the time.The“optimal”guidelines were tested for representative normal,dry,and wet years.The finally proposed guidelines were found in the simulations to attain the 95%target for nearly all locations in the CAWS for the three test years.The developed operational guidelines have been applied since 2018 and have shown improved attainment of the DO standards throughout the CAWS while at the same time achieving similar energy use at the aeration stations on the Calumet River system,greatly lowered energy use on the Chicago River system,and greatly lowered discretionary diversion from Lake Michigan,meeting the recently enacted lower amount of allowed annual discretionary diversion.This case study indicates that emission-based real-time control developed from a well calibrated model holds potential to help many receiving water bodies achieve high attainment of water quality standards.

In their recently published study in Nature,Jenson et al.1 provide compelling,unexpected evidence that bacterial cGAMP synthase(cGAS)and other cGAS/DncV-like Nucleotidyltransferases(CD-NTase)undergo ubiquitin-like conjugation(ULC)in type-II cyclic-oligonucleotide-based anti-phage signaling systems(CBASS).cGAS conjugation promotes cyclic-nucleotide(CN)formation and thus antiphage defense.Furthermore,the authors also reveal a new class of phage-encoded CBASS inhibitor,T4 Vs.4,a CN-sponge which limits CBASS effector function and promotes phage replication,revealing another weapon in the“arms race”between phages and their bacterial hosts.

Mitochondrial transport of catalytic RNAs and targeting of the organellar transcriptome in human cells

Mutations in the small genome present in mitochondria often result in severe pathologies.Different genetic strategies have been explored,aiming to rescue such mutations.A number of these strategies were based on the capacity of human mitochondria to import RNAs from the cytosol and designed to repress the replication of the mutated genomes or to provide the organelles with wild-type versions of mutant transcripts.However,the mutant RNAs present in mitochondria turned out to be an obstacle to therapy and little attention has been devoted so far to their elimination.Here,we present the development of a strategy to knockdown mitochondrial RNAs in human cells using the transfer RNA-like structure of Brome mosaic virus or Tobacco mosaic virus as a shuttle to drive trans-cleaving ribozymes into the organelles in human cell lines.We obtained a specific knockdown of the targeted mitochondrial ATP6 mRNA,followed by a deep drop in ATP6 protein and a functional impairment of the oxidative phosphorylation chain.Our strategy provides a powerful approach to eliminate mutant organellar transcripts and to analyse the control and communication of the human organellar genetic system.

Assessing discharge periodicity in mountain catchments using classified environmental conditions(Tatra Mountains, Poland)

The periodicity of a river expressed in cycles of various lengths(monthly, seasonal,multiannual) is a result of climatic factors and overlapping environmental conditions within its catchment. In uncontrolled or poorly surveyed catchments, it is very difficult to determine the duration of a stream’s hydrological activity. This is especially relevant for catchments with complicated water circulation in karstic rocks. The present study concerns the small catchment of the Str??yski Potok river located in the area of the Tatra National Park, in the Western Tatras. The observation period covered the 2015 hydrological year, which differed hydrologically from average conditions. This study aims to develop a simple method to explain the processes shaping the mountain stream discharge periodicity. The research employed periodic field observations linked with climatic and non-climatic factors. Environmental conditions were assessed as four classes reflecting their influence on appearance or disappearance of mountain stream water. Class boundaries were the values of quartiles. The degree of correspondence between environmental factors and stream field observations was described via the Index DC(Degree Correspondence Index) approach.Complete correspondence(Index DC =0) was found in 23% catchments, a weak relationship between conditions favouring discharge and actual condition(Index DC=-1, +1) was noted within 11 catchments,while in 9 catchments, no such relationship was found(Index DC =-2, +2). The obtained results indicate a correspondence or lack thereof between the environmental potential of the catchment and its discharge periodicity. The discrepancies between the assessment of the influence of climatic and nonclimatic factors and the data collected during field observations provide a basis for more detailed studies.Continuation of these studies based on the proposed classifications will allow for a more complete explanation of water disappearance in river channels and the determination of their short-and long-term discha
