GLP1 and GIP are involved in the action of synbiotics in broiler chickens

Background: In order to discover new strategies to replace antibiotics in the post-antibiotic era in meat-type chicken production, two new synbiotics were tested:(Lactobacillus salivarius IBB3154 plus galactooligosaccharide(Syn1) and Lactobacillus plantarum IBB3036 plus raffinose family oligosaccharides(Syn2).Methods: The synbiotics were administered via syringe, using a special automatic system, into the egg air chamber of Cobb 500 broiler chicks on the 12 thday of egg incubation(2 mg of prebiotics + 105 cfu bacteria per egg).Hatched roosters(total 2,400) were reared on an experimental farm, kept in pens(75 animals per pen), with free access to feed and water. After 42 d animals were slaughtered. Blood serum, pancreas, duodenum and duodenum content were ***: Syn2 increased trypsin activity by 2.5-fold in the pancreas and 1.5-fold in the duodenal content. In the duodenum content, Syn2 resulted in ca 30% elevation in lipase activity and 70% reduction in amylase ***1 and Syn2 strongly decreased expression of m RNA for GLP-1 and GIP in the duodenum and for GLP-1 receptors in the pancreas. Simultaneously, concentrations of the incretins significantly diminished in the blood serum(P < 0.05). The decreased expression of incretins coincides with changed activity of digestive enzymes in the pancreas and in the duodenal content. The results indicate that incretins are involved in the action of Syn1 and Syn2 or that they may even be their target. No changes were observed in key hormones regulating metabolism(insulin, glucagon, corticosterone, thyroid hormones, and leptin) or in metabolic indices(glucose,NEFA, triglycerides, cholesterol). Additionally, synbiotics did not cause significant changes in the activities of alanine and aspartate aminotransferases in broiler chickens. Background: Two differently charged polypeptides, α-poly-L-lysine and poly-L-glutamate, have previously been shown to effectively reduce postoperative intraabdominal adhesions. Though α-poly-L-lysine showed toxicity in doses too close to the lowest therapeutic dose, the aim in the present study was to investigate the possible antiadhesive effect of another four cationic polypeptides. Materials/Methods: 125 mice were studied with a standardized and reproducible adhesion model and given epsilon poly-L-lysine, lactoferrin, lysozyme and polyarginine respectively in a combination with poly-L-glutamate. Epsilon poly-L-lysine was also tested in different concentrations and as single treatment. Results: All four cationic polypeptides above showed a significantly better anti-adhesive effect than the controls receiving saline (p<0.05). Epsilon poly-L-lysine had the best antiadhesive effect of the new substances tested in the experiment. Single treatment with the epsilon poly-L-lysine showed toxic side effects. Discussion: We have shown that epsilon poly-L-lysine, polyarginine, lysozyme and lactoferrin, in descending order, all can reduce postoperative intraabdominal adhesions in mice when combined with poly-L-glutamate. There were side effects of epsilon poly-L-lysine resembling those of α-poly-L-lysine, although less toxic. The antiadhesive effect of epsilon poly-L-lysine did not reach the level of α-poly-L-lysine. Further studies will concentrate on additional investigation, trying to modify the α-poly-L-lysine to lower its toxicity. The less toxic epsilon poly-L-lysine also needs further attention in our research of antiadhesive bioactive polypeptides.

AIMTo investigate serum omentin and vaspin levels in cirrhotic patients; and to assess the relationship of these levels with hemostatic parameters, metabolic abnormalities, cirrhosis severity and ***-one cirrhotic patients (17 with portal vein thrombosis) were analyzed. Serum omentin and vaspin levels were measured with commercially available direct enzyme-linked immunosorbent assays (ELISAs). To assess platelet activity, the following tests were performed using a MULTIPLATE®PLATELET FUNCTION ANALYZER: (1) an ADP-induced platelet activation test; (2) a cyclooxygenase dependent aggregation test (ASPI test); (3) a von Willebrand factor and glycoprotein Ib-dependent aggregation (using ristocetin) test (RISTO test); and (4) a test for thrombin receptor-activating peptide-6 induced activation of the thrombin receptor, which is sensitive to IIb/IIIa receptor ***, but not vaspin, serum concentrations were significantly decreased in patients with portal vein thrombosis (PVT) (P = 0.01). Prothrombin levels were significantly increased in patients with PVT (P = 0.01). The thrombin receptor activating peptide (TRAP) test results were significantly lower in the PVT group (P = 0.03). No significant differences in adipokines serum levels were found regarding the etiology or severity of liver cirrhosis assessed according to the Child-Pugh or Model of End-Stage Liver Disease (MELD) scores. There was a significant increase in the TRAP (P = 0.03), ASPI (P = 0.001) and RISTO high-test (P = 0.02) results in patients with lower MELD scores. Serum omentin and vaspin levels were significantly down-regulated in patients without insulin resistance (P = 0.03, P = 0.02, respectively). A positive relationship between omentin and vaspin levels were found both when all of the patients were analyzed (r = 0.41, P = 0.01) and among those with PVT (r = 0.94, P < 0.001).CONCLUSIONSerum omentin levels are increased in patients without PVT. The situation of the contemporary economic systems is based on their ability to ensure optimal response to the customer needs. There are several important groups of clients, who may determine development processes and who are recipients of public services. They include： residents, entrepreneurs, tourists, and State Treasury. Local governments are equipped with a range of legal instruments to provide public services addressed to the selected clients. The objective of this paper is to present the application of the AHP method to assess the significance of the selected categories of public services for different groups of clients in Poland. The results allow setting priorities for future policy and adjust the tasks to the specific expectations of each group of clients. The model of assessment of public services was built using the analytic hierarchy process （AHP）. Since the 1970＇s, this multicriteria decision-support method has been used by researchers and practitioners in many areas of socioeconomic life. The case study presented in this paper involved 14 experts--public administration workers in one of the districts in Southern Poland. The survey consisted of two stages. First, they assessed--using the AHP--the influence of each group of customers on local development processes. The most important was ＂business＂（priority 38.7%）, followed by ＂tourists＂（29.7%）, ＂citizens＂（26.1%）, and ＂central administration＂（5.5%）. Second, they evaluated at the five-point Liken scale the influence of over 20 different public services with respect to different groups of clients. The results were aggregated using arithmetic mean and multiplied by the priorities of the clients. The synthesized final score indicated the most significant public activities overall, which were ＂roads＂（4.16）, ＂transport＂（3.93）, and ＂tourism＂（3.84）.

Interactive effect of combined exposure to ethylene glycol ethers and ethanol on hematological parameters in rats

The study of the interaction of three glycol ethers, i.e. 2-methoxyethanol (ME), 2-ethoxyethanol (EE) and 2-butoxyethanol (BE) administered subcutaneously for 4 weeks and ethanol simultaneously given as 10% w/v solution for drinking in male rats, was carried out from a toxicodynamic point of view. Administered alone, ME (2.5 and 5.0 mM/kg), EE (2.5 and 5.0 mM/kg) or BE (0.75 and 1.25 mM/kg) resulted in a decrease of red blood cells (RBC), packed cell volumes (PCV), and hemoglobin concentration (HGB), as well as an increase in mean corpuscular volume (MCV) and reticulocyte count (Ret). In the rats co-ex- posed to ethanol and EGAEs, a significantly less pronounced hematological changes in comparison with animal exposed to these ethers alone were seen. The rats simultaneously exposed to ethanol and both ME and EE at the lower dose demonstrated mainly protection from the alterations in leukocyte system. In contrast, in the rats which consumed ethanol and were simultaneously treated with the higher dose of ME or EE (5.0 mM/kg) the amelioration of same hematological parameters were displayed. The intake of ethanol along with BE treatment at both doses resulted in markedly ameliorated hematological parameters, compared to those which were changed by BE alone. In conclusion, the decrease of the hemolytic effects of EGAEs is ethanol dependent. Ethanol is a substrate of alcohol dehydrogenase (ADH), and affinity of this enzyme to ethanol is greater than that to glycol ethers. It is possible that ethanol results in the change in EGAEs metabolism.

Background Medication adherence is an integral part of the comprehensive care of patients with atrial fibrillation （AF） receiving oral anticoagulations （OACs） therapy. Many patients with AF are elderly and may suffer from some form of cognitive impairment. This study was conducted to investigate whether cognitive impairment affects the level of adherence to anticoagulation treatment in AF patients. Me- thods The study involved 111 AF patients （mean age, 73.5±8.3 years） treated with OACs. Cognitive function was assessed using the Mini Mental State Examination （MMSE）. The level of adherence was assessed by the 8-item Mot^sky Medication Adherence Scale （MMAS-8）. Scores on the MMAS-8 range from 0 to 8, with scores 〈 6 reflecting low adherence, 6 to 〈 8 medium adherence, and 8 high adherence. Re- sults 46.9% of AF patients had low adherence, 18.8% had moderate adherence, and 33.3% had high adherence to OACs. Patients with lower adherence were older than those with moderate or high adherence （76.6 ±8.7 vs. 71.3 ~ 6.4 vs. 71.1 ± 6.7 years） and obtained low MMSE scores, indicating cognitive disorders or dementia （MMSE = 22.3 ± 4.2）. Patients with moderate or high adherence obtained high MMSE test results （27.5 ±1.7 and 27,5 ± 3.6）. According to Spearman＇s rank correlation, worse adherence to treatment with OACs was determined by older age （rs = -0.372） and lower MMSE scores （rs = 0.717）. According to multivariate regression analysis, the level of cognitive function was a significant independent predictor of adherence （b = 1.139）. Conclusions Cognitive impairment is an independent determinant of compliance with pharmacological therapy in elderly patients with AF. Lower adherence, beyond the assessment of cognitive function, is related to the age of patients.

The first-ever case of a 54-year-old woman who overdosed on non-steroidal anti-inflammatory drugs in an attempt at *** that incident,she had not been treated for coexisting diseases such as rheumatoid arthritis or *** the time of admission to the General Surgery Department,the patient reported pains in the epigastric region with accompanying nausea and vomiting with mucous content as well as the inability to ingest food *** parenteral and enteral feeding,the patient exhibited a drop in body *** histopathologic examination of a sample taken from the stomach during gastroscopy showed some non-specific necrotic and inflammatory masses with ***,a very small,infiltrated stomach with an initial section of duodenum was identified.A total stomach resection together with the reconstruction of digestive tract continuity was performed using the Roux-Y *** examination of the stomach revealed a deep,chronic and exacerbated inflammatory condition with an extensive ulceration over the entire length of the stomach,reaching up to the ***,numerous lymphatic glands with inflammatory reaction changes were observed.

Glutamate as a Neural Stress Factor in Humans and Animals

L-glutamic acid (glutamate-Glu) serves as one form with very strong stimulatory neurotransmitter (near aspartic, kainic, alpha-amino-3-hydroxy-5-me- thyl-4-izoxazole propionic acid (AMPA), chinolic and L-homocysteinic acid, glycine and D-serine) at the majority of neural excitatory synapse in the mammals and nonmammals central, sympathetic nervous system (CNS and SNS, respectively) and in different peripheral tissues and organ. It mediates interactions via stimulation a variety ionotropic N-methyl-D-As- partate (NMDA), AMPA and kainate receptors (ligand gated calcium channels) and III groups of the metabotropic glutamate receptors (mGluR1-8) family members (G-protein coupled receptors). It is good known different neuromodula-tion/interaction between Glu and norepinephrine (NE), dopamine (DA), gamma-amino-butyric acid (GABA), oxytocin/vasopressin (Oxy/AVP) and steroid receptors during stress in the central nervous system. In this review we describe the molecular structure of these glutamatergic receptors and discuss they neuropharmacology and clinical use probability of their antagonist, in stress particularly. On the other hand it was interesting if Glu can increase catecholamine (CA) release from motivational structures as stressoric factor in hypothalamo-pituitary adrenal axis (HPA) in the stress inducing processes. Our findings show that Glu more influences the brain’s motivational structure, which may indicate its contribution to the stress response by direct modulating the amount of catecholamine released.
