Aberrant glycosylation is considered to be a hallmark of colorectal cancer(CRC),as demonstrated by various *** the N-glycosylation of cell lines and serum has been widely examined,the analysis of cancer-associated N-glycans from tissues has been hampered by the heterogeneity of tumors and the complexity of N-glycan *** overcome these obstacles,we present a study using laser capture microdissection that makes it possible to largely deconvolute distinct N-glycomic signatures originating from different regions of heterogeneous tissues including cancerous,stromal,and healthy mucosa cells.N-glycan alditols were analyzed by means of porous graphitized carbon liquid chromatographyelectrospray ionization tandem mass spectrometry,enabling the differentiation and structural characterization of isomeric *** total,116 N-glycans were identified that showed profound differences in expression among cancer,stroma,and normal *** comparison with healthy mucosa,the cancer cells showed an increase in a2-6 sialylation and monoantennary N-glycans,as well as a decrease in bisected ***,specific sialylated and(sialyl-)LewisA/X antigen-carrying N-glycans were exclusively expressed in *** comparison with cancer,the stroma showed lower levels of oligomannosidic and monoantennary N-glycans,LewisA/X epitopes,and sulfation,as well as increased expression of(core-)fucosylation and a2-3 *** study reveals the distinct N-glycomic profiles of different cell types in CRC and control tissues,proving the necessity of their separate analysis for the discovery of cancer-associated glycans.

AIMTo establish the role of nitric oxide (NO), ascorbic acid and tumour necrosis factor-α (TNF-α) in the pathogenesis of pseudoexfoliation glaucoma (XFG).METHODSOur study included 120 patients who were referred for cataract surgery. All patients were divided into four groups according to clinical findings: XFG, early and late pseudoexfoliation syndrome (XFS), and cataract (without pseudoexfoliation). Serum and aqueous humour levels of the ascorbic acid, NO and TNF-α were measured. The concentrations of the ascorbic acid and NO were measured by an appropriate spectrophotometric method. Enzyme-linked immunosorbent assay (ELISA) was used to determine TNF-α *** humour concentration of ascorbic acid was significantly lower in patients with late XFS (0.61±0.11 mmol/L) and XFG (0.48±0.15 mmol/L) compared to patients with early XFS (0.9±0.15 mmol/L) and cataract (1.16±0.22 mmol/L), while there was no difference in serum concentration in all examined groups. Aqueous humour concentration of NO was significantly higher in patients with XFG (77.7±11.4 µmol/L) compared to patients with early XFS (50.27±9.34 µmol/L) and cataract (49.77±7.1 µmol/L), while serum concentration was increased in the early stage of XFS (73.26±8.29 µmol/L). Aqueous humour level of proinflammatory cytokine TNF-α was increased in patients with XFS (early 460.04±18.32 pg/mL; late 502.42±53.23 pg/mL) and XFG (510.34±43.07 pg/mL), while there was no difference in serum level in all examined groups of *** ascorbic acid and elevated NO and inflammation related cytokine TNF-α level in aqueous humour of the patients with developed XFG suggest that oxidative stress induces local inflammation.

Reconstruction of defects on the chin represents a challenge given the functional, multidirectional movement of the chin subunit. In this article, we describe the surgical technique of a bilateral O to Z rotation flap on the chin, as well as complications, cosmetic outcome, and patient satisfaction. A retrospective chart review from January 2009-June 2011 identified twelve patients where an O to Z flap was used to repair a surgical defect. Of the twelve patients identified, one patient had a presumed post-operative infection, and one patient required additional corrections. There were no complications of hematomas or flap necrosis. Overall patient cosmetic outcome was excellent. An O to Z closure is an option for chin reconstruction of small to medium defects that offers a satisfactory cosmetic outcome, high patient satisfaction and low complication rate.

Counteraction of perforated cecum lesions in rats: Effects of pentadecapeptide BPC 157,L-NAME and L-arginine

AIM To study the counteraction of perforated cecum lesion using BPC 157 and nitric oxide(NO) system *** Alongside with the agents' application(after 1 min, medication(/kg, 10 ml/2 min bath/rat) includes: BPC 157(10 μg), L-NAME(5 mg), L-arginine(100mg) alone or combined, and saline baths(controls)) on the rat perforate cecum injury, we continuously assessed the gross reappearance of the vessels(USB microcamera) quickly propagating toward the defect at the cecum surface, defect contraction, bleeding attenuation, MDA-and NO-levels in cecum tissue at 15 min, and severity of cecum lesions and adhesions at 1 and 7 d. RESULTS Post-injury, during/after a saline bath, the number of vessels was significantly reduced, the defect was slightly narrowed, bleeding was significant and MDA-levels increased and NO-levels decreased. BPC 157 bath: the vessel presentation was markedly increased, the defect was noticeably narrowed, the bleeding time was shortened and MDA-and NO-levels remained normal. L-NAME: reduced vessel presentation but not more than the control, did not change defect and shortened bleeding. L-arginine: exhibited less vessel reduction, did not change the defect and prolonged bleeding. In combination, mutual counteraction occurred(L-NAME + L-arginine) or the presentation was similar to that of BPC 157 rats(BPC 157 + L-NAME; BPC 157 + L-arginine; BPC 157 + L-NAME + L-arginine), except the defect did not change. Thereby at day 1 and 7, saline, L-NAME, L-arginine and L-NAME + L-arginine failed(defect was still open and large adhesions present). CONCLUSION The therapeutic effect was achieved with BPC 157 alone or in combination with L-NAME and L-arginine as it was able to consolidate the stimulating and inhibiting effects of the NO-system towards more effective healing recruiting vessels.

Antibiotics and near-patient testing: Differences in habits between physicians completing or discontinuing a medical audit

Background and Aim: Respiratory tract infection (RTI) is a common reason for consulting primary health care. Antibiotic prescribing for RTIs varies among physicians indicating that national guidelines are not always adhered to. The aim was to study if antibiotic prescribing and use of near-patient tests were different among physicians who complete an audit registration and those who discontinue their participation. Method: A prospective cohort study where physicians participated in an APO (Audit Project Odense) process, making an audit registration for every appointment with a patient who had a respiratory tract infection during 4 weeks in 2008 and 4 weeks in 2009. Between the registrations, a limited educationally oriented intervention was made. 18 Primary Health Care Centres located in three counties in southern of Sweden with 77 primary health care physicians participated. When comparing proportions the Chisquare test was used. Mann Whitney U-test was used when comparing independent groups and Wilcoxon’s signed-rank test was used when comparing dependent groups. Results: Of the 77 physicians, 38 participated only at baseline (group 1) and 39 participated in both registrations (group 2). The overall use of CRP near-patient tests was 37% in group 1 and 28% in group 2 (Chisquare p < 0.001), and the overall use of Strep-A near-patient tests was 31% and 20%, respectively (Chisquare p < 0.001). When the Strep-A near-patient test was negative in pharyngitis/tonsillitis, antibiotics were prescribed to 45% in group 1 and to 12% by group 2 (Chisquare 0.003). Conclusion: In conclusion, this study showed that physicians, who were more inclined to complete audit participation, used near-patient tests and prescribed antibiotics more correctly, according to the national guidelines for respiratory tract infections, than physicians who discontinued the participation. Amorphous and nanocrystalline (Fe1–xNix)81Nb7B12 (x = 0, 0.25, 0.5, 0.75) alloys were measured by M?ssbauer spectrometry in the weak external magnetic field of 0.5 T. From structural analyses, ferromagnetic bcc-FeNi and fcc-FeNi and paramagnetic (Fe-Ni)23B6 phases were identified in the annealed samples. It was shown that in the external magnetic field the intensities of the 2nd and the 5th lines (A23 parameter) are the most sensitive M?ssbauer parameters. Rather small changes were observed in the values of internal magnetic field. Our results showed that the amorphous precursor is more sensitive to the influence of external magnetic field than the nanocrystalline alloy. All spectra of amorphous precursor showed the increase of A23 parameter and decrease of internal magnetic field values of about 1 T (±0.5 T) under influence of external magnetic field. In the case of nanocrystalline samples the tendency for the values of internal magnetic field is similar but the effect is not so pronounced. The measurements confirmed that even weak external magnetic field affected orientation of the net magnetic moments. Our results indicate that effect of the external magnetic field is stronger in the case of amorphous samples due to their disordered structure.

The aim of the current paper is to discuss the replacement of diesel oil (DO) consumption by natural gas (NG) in a cogeneration system. A specific industrial consumption case study was chosen to be the method used to accomplish a robust analysis. The results have shown the advantages in reducing CO2, CH4, N2O and particulate matter emissions, as well as the need to keep the NOx emission rates. After proceeding with theoretical studies concerning our case, we concluded that the diesel oil replacement by natural gas is beneficial for gas emission reduction. Public policies should consider local development based on the use of different fuels, such natural gas, to achieve the integration between decentralized energy generation and energy-efficient initiatives.
