A nuclear import inhibitory peptide ameliorates the severity of cholecystokinin-induced acute pancreatitis

AIM: To assess the effect of our novel cell-permeable nuclear factor-kappaB (NF-κB) inhibitor peptide PN50 in an experimental model of acute pancreatitis. PN50 was produced by conjugating the cell-penetrating penetratin peptide with the nuclear localization signal of the NF-κB p50 ***: Pancreatitis was induced in male Wistar rats by administering 2×100 μg/kg body weight of cholecystokininoctapeptide (CCK) intraperitoneally (IP) at an interval of 1 h. PN50-treated animals received 1 mg/kg of PN50 IP 30 min before or after the CCK injections. The animals were sacrificed 4 h after the first injection of ***: All the examined laboratory (the pancreatic weight/body weight ratio, serum amylase activity,pancreatic levels of TNF-α and IL-6, degree of lipid peroxidation, reduced glutathione levels, NF-κB binding activity, pancreatic and lung myeloperoxidase activity) and morphological parameters of the disease were improved before and after treatment with the PN50 *** to the histological findings, PN50 protected the animals against acute pancreatitis by favoring the induction of apoptotic, as opposed to necrotic acinar cell death associated with severe acute ***: Our study implies that reversible inhibitors of stress-responsive transcription factors like NF-κB might be clinically useful for the suppression of the severity of acute pancreatitis.

Background:It remains unclear if the vascular and connective tissue structures of primary and metastatic tumors are intrinsically determined or whether these characteristics are defined by the host *** we examined the microanatomical steps of vasculature and connective tissue development of C38 colon carcinoma in different ***:Tumors produced in mice at five different locations(the cecal wall,skin,liver,lung,and brain)were ana-lyzed using fluorescent immunohistochemistry,electron microscopy and quantitative real-time polymerase chain ***:We found that in the cecal wall,skin,liver,and lung,resident fibroblasts differentiate into collagenous matrix-producing myofibroblasts at the tumor *** activated fibroblasts together with the produced matrix were incorporated by the *** connective tissue development culminated in the appearance of intratumoral tissue columns(centrally located single microvessels embedded in connective tissue and smooth muscle actin-expressing myofibroblasts surrounded by basement membrane).Conversely,in the brain(which lacks fibroblasts),C38 metasta-ses only induced the development of vascularized desmoplastic tissue columns when the growing tumor reached the fibroblast-containing ***:Our data suggest that the desmoplastic host tissue response is induced by tumor-derived fibrogenic molecules acting on host tissue *** concluded that not only the host tissue characteristics but also the tumor-derived fibrogenic signals determine the vascular and connective tissue structure of tumors.

背景与目的关于原发和转移性肿瘤的血管和结缔组织结构是由本身决定还是由宿主组织决定,目前尚不清楚。因此,我们研究了不同组织中C38结直肠癌血管和结缔组织发展的显微解剖过程。方法采用免疫荧光染色、电镜和实时定量聚合酶链反应等方法对小鼠盲肠壁、皮肤、肝脏、肺和大脑5个不同部位生成的肿瘤进行分析。结果我们发现在盲肠壁、皮肤、肝脏和肺的成纤维细胞分化成胶原蛋白基质,在肿瘤周围产生肌成纤维细胞。这些活化的成纤维细胞和生成的基质被肿瘤吸收利用。结缔组织的发展最终引起瘤内组织柱的出现(位于中心位置的单个微血管包埋在结缔组织和表达平滑肌肌动蛋白的肌成纤维细胞中,最外围由基底膜包围)。相反,在缺乏成纤维细胞的大脑中,仅当增生的肿瘤接触到含有成纤维细胞的脑膜时,C38转移才能诱导血管生成的促结缔组织增生性纤维柱的形成。结论我们的数据表明,作用于宿主组织成纤维细胞的肿瘤源性纤维生成分子诱导了促结缔组织增生的宿主组织反应。我们认为宿主组织特征和肿瘤来源的纤维生成信号共同决定了肿瘤的血管和结缔组织的结构。

In Central-and Eastern Europe,the collapse of socialist regimes resulted in a transformation of state-owned agricultural cooperatives to privately owned lands from the early 1990s *** socioeconomic processes resulted in landscape-scale changes in biodiversity,ecosystem services and agricultural *** parallel,large-scale abandonment of croplands,especially on sandy,salty or fre-quently inundated areas,became *** croplands are usually sensitive to species invasions,and are hotspots of noxious weeds,posing threats both to agriculture and nature *** restoration on former croplands can be an effective strategy for suppressing these ***,a common goal of nature conservation and agriculture can be the restoration of grasslands on former croplands to(1)suppress weed and/or invasive species in line with the EU policy“Good Farming Practices”,(2)support animal husbandry by creating meadows or pastures,and to(3)recover biodiversity and ecosystem *** the present paper we report“best practices”of grassland restoration projects from *** aim was to compare the effectiveness of spontaneous grassland recovery *** grassland restoration by seed sowing in terms of the recovery of biodiversity and ecosystem services,such as weed control and biomass *** results showed that grassland restoration on abandoned fields offers a viable solution for restoring biodiversity and ecosystem *** sowing ensures higher weed control and biomass pro-duction,but results in lower biodiversity compared to spontaneous *** restoration methods can be cost-effective,or even profitable even within a relatively short period of a nature conservation project.

Marker-assisted selection(MAS)is often employed in crop breeding programs to accelerate and enhance cultivar development,via selection during the juvenile phase and parental selection prior to ***-generation sequencing and its derivative technologies have been used for genome-wide molecular marker *** bridge the gap between marker development and MAS implementation,this study developed a novel practical strategy with a semi-automated pipeline that incorporates trait-associated single nucleotide polymorphism marker discovery,low-cost genotyping through amplicon sequencing(AmpSeq)and decision *** results document the development of a MAS package derived from genotyping-by-sequencing using three traits(flower sex,disease resistance and acylated anthocyanins)in grapevine *** vast majority of sequence reads(⩾99%)were from the targeted *** 380 individuals and up to 31 amplicons sequenced in each lane of MiSeq data,most amplicons(83 to 87%)had<10%missing data,and read depth had a median of 220–244×.Several strengths of the AmpSeq platform that make this approach of broad interest in diverse crop species include accuracy,flexibility,speed,high-throughput,low-cost and easily automated analysis.

During the past few decades, the investigative technologies of molecular biology- especially sequencing-underwent huge advances, leading to the sequencing of the entire human genome, as well as the identification of several candidate genes and the causative geneticvariations that are responsible for monogenic skin diseases. These advances provided a solid basis for subsequent studies elucidating mechanisms of monogenic skin diseases and improving our understanding of common skin diseases. Furthermore, these discoveries also contributed to the development of novel therapeutic modalities for monogenic skin diseases. In this review, we have used the disease spectrum caused by mutations in the CYLD gene- Brooke-Spiegler syndrome, familial cylindromatosis and multiple familial trichoepithelioma type 1- as a model for demonstrating the knowledge explosion for this group of diseases.

背景：临床医师必须能够正确诊断重症肌无力，因为延迟诊断，可使患者面临这种可治疗疾病的并发症的危险。目的：确定有关病史和检查或简单试验结果足否能改变诊断重症肌无力的可能性。资料来源：对MEDLINE收录的英文文献（1966年1月-2005年1月）进行检索，所用检索词包括重症肌无力、诊断和检验，其他资料来源于检出文献的参考文献。研究选择：在患或未患重症肌无力的患者评估某一特定症状或体征的研究。在检索到的640篇文献中，有33篇审查合格。其中15篇符合入选标准，构成本综述的基础。数据提取：两位作者分别审读每篇研究，判断其适用性，使用标准方法摘录数据，根据以前发表的标准对研究的质量进行分类。数据综合：“长时间说话时讲话变得难以理解”病史以及“偷看”征提高了重症肌无力的可能性（分别为：似然比[likelihood ratio，LR]，4．5；95％可信区间[confidence intexwal，CI]，1．2—17．0以及LR，30．0；95％CI，3．2—278．0）。缺乏这些表现并未明显降低患重症肌无力的可能性。有些研究仅仅评估了其他一种病史特征和体征（分别为“食物在吞咽后仍留在嘴咀”和眼颤），它们均未明显改变患重症肌无力的可能性。冰块试验反应异常是有帮助的（总阳性LR，24．0；95％CI，8．5．-67；0），而正常反应则能降低重症肌无力的可能性（总阴性LR，0．16；95％CI，0．09—0．27）。对抗胆碱酯酶药物（主要为腾喜龙）阳性反应提高诊断重症肌无力的可能性（总阳性LR，15.0；95％CI，7．5—31.0），阴性反应则降低诊断重症肌无力的可能性（总阴性LR，0．11；95％CI，0.06—0．21）。睡眠试验结果异常有助于确认诊断（LR；53．0；95％CI，3．4—832．0）。休息和睡眠试验结果正常时，不太可能为重症肌无力（分别为：LR，0．52；95％CI，0．29—0．95和LR，0．01；95％CI，0．00—0．16）。结论：某些病史和体检项目以及在诊所进行的简单试验（冰块试验、睡眠试验和腾喜龙试验）有助于预测患重症肌无力的可能性。然而，鉴于在已报道的临床研究人群该病的高发生率，对这些结果必须谨慎解释。该综述受到文献中较少研究和报告体征和症状的限制;仍需要更多的研究，以便对常见的病史特点以及专家们普遍了解并实施的简单诊断方法的价值做进一步评价。
