This study aims to uncover the hormetic process of luteolin， a common dietary flavone， in neuronal cells through its role in inducing mitochondrial stress. In rat pheochromocytoma PC12 cells， luteolin at low concentrations caused a mild and reversible loss of mitochondrial membrane potential (MMP)， while high concentrations of luteolin triggered intense and sustained depolarization of MMP. The MMP disturbance was shown to have a close association with the trophic and/or toxic effects triggered by luteolin; because the common mitochondrial uncouplers shared similar bi-phase dose-response on cell viability， as that of luteolin. Along with the induced MMP pertubation， luteolin triggered the development of autophagy and mitophagy， as determined by mCherry-GFP-LC3B tandem protein， and by the co-localization of mitochondrial/lysosomal staining. Subsequent application of autophagy inhibitors in the cultures blocked the luteolin-induced neurotrophic activities and sensitized the cells to be less resistant to luteolin-mediated cytotoxicity. Other flavonoids also displayed similar properties in the cultures， indicating that these compounds act as hormetic pharmacological inducers that stimulate cells to become more resilient and adapt to threats. This study provides a unifying mechanistic explanation for the neuro-beneficial effects of luteolin and other flavonoids.

OBJECTIVES:Proteins are the major component and play a key role in nutritious and therapeutic functions of edible bird’s nest(EBN);however,limited studies have been conducted on the protein due to difficulties in extraction,isolation as well as *** study aimed to provide comprehensive information for the quality evaluation of EBN peptides,which would be a valuable reference for further study on EBN ***:Here,we developed a quality control method using high performance liquid chromatography(HPLC)peptide fingerprints deriving from EBN being digested with simulated gastric *** characteristic peptide peaks were collected and identified by LC-MS/***:The characteristic peptide peaks,corresponding to the protein fragments of acidic mammalian chitinase-like,lysyl oxidase,and Mucin-5AC-like,were identified and ***,the principal component analysis indicated that the fingerprints were able to discriminate colour of EBN(white/red)and production sites(cave/house)of White EBN on the same weight *** proposed by the model developed in this study,Muc-5AC-like and AMCaselike proteins were the markers with the highest discriminative ***:The overall findings suggest that HPLC peptide fingerprints were able to clearly demonstrate peptide profile differences between genuine and adulterated EBN samples;and classify EBN samples by its color and production *** addition,the protein identification results suggested that Muc-5AC-like protein was the major protein in EBN.

Amino acids are naturally occurring compounds in many edible or medicinal plants,which possess a variety of nutraceutical effects in ***,a method of ultrasound-assisted extraction was optimized using Box-Behnken design of response surface methodology in maximizing the yield of free amino acids deriving from flowers of Apocynum venetum *** the optimal condition of ultrasound-assisted extraction,i.e.187.5 W of ultrasonic power,31.93 min of extraction time,and 0.47 mg/ml of solid-liquid ratio,the experimental yield of extractive was 287.17±0.205 mg/g,which was in close agreement with the value,as predicted by the established *** addition,a hydrophilic interaction ultra-high performance liquid chromatography coupled with QqQ-MS/MS method was developed and validated for simultaneous quantification of 20 types of amino acids without derivatization contained in *** *** analytical method was validated by matrix effect,linearity,limit of detection,limit of quantification,precision,repeatability,stability,and *** analysis results showed that *** flower was rich in free amino acids of~3%of total dried weight,and which contained leucine(13.7 g/mg),isoleucine(7.9 g/mg),and lysine(2.2 g/mg)as the most abundant amino ***,*** flower could provide beneficial nutrient values for human health,*** weight maintenance or glucose homeostasis.

Panax notoginseng (Burk) F.H. Chen is one of the most famous Chinese traditional medicinal plants, which belongs to Panax genus under Araliaceae family. At the present, Panax notoginseng cultivated is a mix colony. Using DALP makers, this article studied on genetic diversity of cultivated populations of Panax notoginseng in Wenshan County of Yunnan province and Napo country of Guangxi province. And the results showed that there were 260 polymorphic loci detected from the total of 292 in 13 populations, and there was great part of genetic diversity found between populations and the genetic differentiations were lower within populations. So there is broad prospect in good species breeding. And it can provide basic information for resource protection and sustainable use of Panax notoginseng.

Edible bird’s nest (EBN;Yan Wo), or cubilose, is originated from the salivary secretion of Aerodramus fuciphagus. In Asia, EBN is famous for its unproven skin lightening function. Here, we aim to reveal the active ingredients of EBN responsible for skin lightening function. Three major fractions were isolated from EBN water extract by chromatography using LC-MS/MS, bioactivities of these isolated fractions were analyzed by assays of tyrosinase, melanocytes and 3D human skin model, from which, N-acetylneuraminic acid (NANA), the second isolated fraction showed an inhibition effect on tyrosinase activity in a dose-dependent manner. The IC50 of tyrosinase originated from mushroom and human was 16.93 mM and 0.10 mM respectively. Furthermore, only EBN with higher content of NANA (e.g. White and Red EBN), but not EBN with less NANA (e.g. Grass EBN), showed promising skin lightening function. This is the first report to reveal NANA being an active ingredient of EBN on skin lightening function.

目的对中药材墨旱莲中有效成分进行定性定量鉴别,对其水分、总灰分、酸不溶性灰分、浸出物及重金属含量进行定量测定,完善墨旱莲药材的质量标准.方法采用薄层色谱法定性鉴别墨旱莲中的蟛蜞菊内酯,采用高效液相色谱法定量测定蟛蜞菊内酯含量,采用杂质检查法、烘干法、灰分测定法、浸出物测定法、重金属检查法进行常规项目检查.结果 10批墨旱莲样品中MHLS-005的蟛蜞菊内酯含量低于《中华人民共和国药典》规定的 0.04%,MHLC-001 酸不溶性灰分的含量高于《中华人民共和国药典》标准,MHLS-004和MHLS-006中水分含量高于《中华人民共和国药典》规定的标准.结论所建立的方法简便、准确,可作为质量标准以控制墨旱莲药材的质量.

目的对穿心莲药材的有效成分进行定性定量鉴别,对水分、总灰分、酸不溶性灰分、浸出物及重金属含量进行定量测定,进一步完善穿心莲药材的质量标准。方法采用薄层色谱法对穿心莲中的穿心莲内酯和脱水穿心莲内酯进行定性鉴别,高效液相色谱法进行定量测定,常规检查项目按2010年版《中华人民共和国药典》(一部)附录ⅨA杂质检查法、ⅨH水分测定法中的烘干法、附录ⅨK灰分测定法、附录ⅩA浸出物测定法及ⅨE重金属检查法测定。结果 10批穿心莲样品中穿心莲内酯和脱水穿心莲内酯及醇溶性浸出物含量均符合我国药典规定的标准。结论所建立的方法简便、准确,能作为质量标准以控制穿心莲药材的质量。

Flavonoids exhibit a wide range of biological activities including anti-inflammatory activity, while some 1,3-benzoxazine derivatives show anti-inflammatory activity. In order to explore the novel anti-inflammatory agents, based on the principles of bioisosterism and hybridization, ten novel ehromeno[8,7-e][1,3]oxazin-4-ones were syn- thesized and characterized with IR, ^1H NMR, MS and elemental analyses. The anti-inflammatory activities of the target compounds were evaluated via the croton oil-induced ear oedema test in Swiss mice. According to screened resuits, some target compounds show potent anti-inflammatory activity.

目的:测定大青叶及其3种类似品中靛蓝、靛玉红的含量,并建立指纹图谱以评价药材质量。方法:采用HPLC建立含量测定方法和指纹图谱的分析条件,比较国内不同产地的大青叶及其3种类似品共17批样品中靛蓝和靛玉红的含量,并计算其HPLC指纹图谱相似度。结果:总体上,蓼蓝中靛蓝含量高于菘蓝,而靛玉红含量低于菘蓝,马蓝和路边青均没有检测到靛蓝和靛玉红。另外,17批样品中,不论是同品种还是不同品种间靛蓝和靛玉红或指纹图谱都有很大的差异。结论:上述方法可有效区分4种称为"大青叶"的商品,并为大青叶药材的质量评价提供了科学依据。

Guizhi Fuling capsule(GFC), a traditional Chinese medicine(TCM) with effects of promoting blood circulation and dissipating blood stasis, has been widely used in the clinic. Because of the complex matrix and various chemical structure types, quality control of GFC remains great challenge. In the present study, an ultra performance liquid chromatography hybrid triple-quadrupole mass spectrometry(UPLC-QQQ MS) method with ultrafast positive/negative ionization switching was developed for simultaneous determination of 18 bioactive components in GFC, including methyl gallate, ethyl gallate, oxypaeoniflorin, benzoic acid, albiflorin, paeonolide, paeoniflorin, 1, 2, 3, 4, 6-pentagalloylglucose, mudanpioside C, benzoyloxypaeoniflorin, benzoylpaeoniflorin, pachymic acid, amygdalin, cinnamaldehyde, paeonol, cinnamic acid, 4-hydroxybenzoic acid, and gallic acid. Separation was performed on an Agilent Zorbax Extend–C18 column(2.1 mm × 50 mm, 1.8 μm), using a gradient elution with acetonitrile and water containing 0.1% formic acid. Cholic acid was selected as the internal standard. This newly developed method was fully validated for linearity, precision, accuracy, and stability, and then applied to quality assessment of GFC. Finally, the batch-to-batch reproducibility of GFC samples was evaluated by the cosine ration and Euclidean distance method, which showed high quality consistency. The results demonstrated that the developed method provided a reasonable and powerful manner for quality control of GFC.
